Global transcriptional profiling in porcine liver and lymph node tissues during acute pleuropneumoni

Project description:Expression profiles of porcine liver and tracheobronchial lung lymph node tissues were studied to identify genes being significantly affected by bacterial infection of the lungs. Samples were taken from liver and tracheobronchial lung lymph node tissues in pigs experimentally infected with A. pleuropneumoniae, serotype 5b, and from healthy non-inoculated control pigs from the same herd. Samples were investigated for changes in gene expression by means of global cDNA microarrays. Expression profiling of samples of liver and lung lymph node tissues from ten pigs challenged with Actinobacillus pleuropneumoniae and from five healthy control pigs was conducted by hybridising all 2x15 samples against a tissue specific common reference consisting of a pool of equal amount of total-RNA from all samples of each tissue type and were balanced with respect to control and challenged animals.

Early pig response to experimental infection with Actinobacillus pleuropneomuniae

Project description:The objective of this study was to identify porcine genes which expression was affected by experimental infection with A. pleuropneumoniae within 24 hours after experimental challenge. Microarray experiments were conducted to reveal genes being significantly differentially expressed in infected versus non-infected lung tissue and in liver and lung lymph node tissue from three challenged versus two non-challenged animals. Project description:Postweaning multisystemic wasting syndrome (PMWS) is one of the pig diseases with major economic impact worldwide. Clinical, pathologic and some immunologic aspects of this disease are well-known, but the molecular mechanisms underlying pathogenic mechanisms of the disease are still poorly understood. The objective of the present study was to investigate the global changes in gene expression in the mediastinal lymph nodes from pigs naturally affected by PMWS and healthy counterparts, using the Affymetrix Porcine Genechip®. This is the first study on gene expression in pigs naturally affected by PMWS. The present results allowed identifying potential mechanisms underlying the inflammation, lymphocyte depletion in lymphoid tissues and immune suppression, which are key features of PMWS. A total of twenty-five conventional 13 to 15-week-old pigs were used in this study. Animals were selected from three farms with historical records of PMWS and free of Porcine reproductive and respiratory syndrome virus (PRRSV) and Pseudorabies virus (PRV) infections. Twelve out of the 25 pigs fulfilled the PMWS diagnosis and were selected as cases, and the remaining thirteen pigs were selected as healthy control animals. At least one case and one control were used from each farm. 25 microarrays were used in the experiment, corresponding to the RNAs from mediastinal lymph nodes of 12 PMWS-affected pigs and 13 healthy controls.

Expression data of muscle and draining lymph node after intramuscular ISCOM-Matrix vaccine adjuvant treatment

Project description:ISCOM vaccines induce a balanced Th1/Th2 response and cytotoxic T lymphocytes. The adjuvant component, ISCOM-Matrix, consists of purified saponin fractions, cholesterol and phospholipids. The mode of action for the ISCOM-Matrix is known to some extent but still we lack knowledge of important segments in initiation of the immune response. The study was performed to analyze the early transcriptional responses to the ISCOM-Matrix alone, without the use of co-administered antigen. Matrix M (AbISCOÂ® 100) was given as intramuscular injection and after 24 hours the pigs was sacrificed for gene expression analysis, performed for the injection site and the draining lymph node. Results revealed 594 and 362 genes to be differentially expressed (FC > 2; q < 0.05) at the injection site and in the lymph node, respectively. This show that the ISCOM-Matrix, without any antigen added, has a clear immunomodulatory effect at the site of administration as well as in the draining lymph node. Six pigs, castrated males, 11 weeks old was used for the experiment. They were sorted two weeks prior the treatment and kept separate throughout the experiment. 3 pigs were intramuscularly (biceps femoris) injected with ISCOM-Matrix (AbISCOÂ® 100) suspended in 1 ml sterile saline. 3 control animals were similarly injected with saline only. The pigs were sacrificed after 24 hours and muscle tissue from injection site and draining (internal iliac) lymph nodes were collected and transferred to RNAlater for RNA isolation.

Quantification of microRNA expression in early-stage oral squamous cell carcinoma

Furthermore, we compared cases of early-stage oral squamous cell carcinoma with lymph node metastasis with cases without lymph node metastasis. The miRNAs obtained may not only serve as predictive biomarkers for lymph node metastasis, but may also be used further to understand disease.
