Transcriptome study of rice early response to rice blast fungus

Project description:Rice blast disease is a major threat to rice production worldwide, but the mechanisms underlying rice resistance to the causal agent Magnaporthe oryzae remain elusive. In this whole-genome transcriptome study of rice early defense response to M. oryzae, we applied Affymetrix Rice Genome Genechip to compare the compatible and incompatible rice-M. oryzae interactions in 24 hours post-inoculation. Leaf samples were harvested from three biological replicates of fungal- and mock-inoculated seedlings at 24 hours post-inoculation, from which RNA were extracted and analyzed with Genechip Rice Genome Array.

Project description:In order to understand the mechanisms of Drought induced susceptibility (DIS) we’ve conducted a dual RNAseq experiment on rice infected tissues by Magnaporthe oryzae. At 4 days post inoculation tissues have been collected on mock inoculated and M. oryzae inoculated plants. Rice were conducted under two type of water regime: DIS Drought during three days before inoculation, NoDIS no drought before inoculation. RNAseq was conducted both on rice and fungal RNA.

Project description:In order to understand the mechanisms of Nitrogen induced susceptibility (NIS) we’ve conducted a dual RNAseq experiment on rice infected tissues by Magnaporthe oryzae. At 0 day post inoculation and 2 days post inoculation tissues have been collected on mock inoculated and M. oryzae inoculated plants. Rice were conducted under two type of nitrogen fertilization: 0N all fertilization but nitrogen, 1N all fertilization and NH4NO3. The fertilization was applied one day before inoculation. RNAseq was conducted both on rice and fungal RNA.

Project description:affy_riz_2011_7 - affy_riz_2011_7 - The Bacterial Leaf Blight disease of rice is due to Xanthomonas oryzae pv. oryzae. As for many pathogenic bacteria, it relies on a type 3 secretion system (TTSS) that is devoted to the injection of type 3 effectors (T3Es) into the eukaryotic host cell. These proteins are meant to suppress host basal defense responses and/or mimic some host regulatory function promoting bacterial survey in the plant. During an incompatible interaction, T3Es may act as Avr proteins and stimulate Effector-Triggered-Immunity. We aim at evaluating the transcriptomic response of rice leaves challenged with avirulent strains of Xoo BAI3 and MAI1 on resistant lines IR64 and IRBB4 versus the reference susceptible rice line Nipponbare. In addition, we investigated the transcriptomic response of rice leaves upon inoculation of an XoohrcC mutant strain affected in the production of a functional TTSS.-The goal of the experiment is to characterize the rice leaf transcriptome response, upon the inoculation of susceptible and resistant rice leaves 24 hours post-infection. To that end, the experimental design includes the inoculation of susceptible Nipponbare rice leaves with Xoo strains BAI3 (race A1) and MAI1 (race A3), that will be compared to the response of resistant lines IRBB4 and IR64 rice lines. In addition, Nipponbare rice leaves will also be challenged with the BAI3hrcC mutant that is affected in the production of a functional TTSS.

Transcription profiling of rice cv. Nipponbare roots infected with Magnaporthe oryzae strain Guy11

Project description:Magnaporthe oryzae causes rice blast, the most devastating foliar fungal disease of cultivated rice. During disease development the fungus simultaneously maintains both biotrophic and necrotrophic growth corresponding to a hemi-biotrophic life style. The ability of M. oryzae to also colonize roots and subsequently develop blast symptoms on aerial tissue has been recognized. The fungal root infection strategy and the respective host responses are currently unknown. Global temporal expression analysis suggested a purely biotrophic infection process reflected by the rapid induction of defense response-associated genes at the early stage of root invasion and subsequent repression coinciding with the onset of intracellular fungal growth. The same group of down-regulated defense genes was increasingly induced upon leaf infection by M. oryzae where symptom development occurs shortly post tissue penetration. Our molecular analysis therefore demonstrates the existence of fundamentally different tissue-specific fungal infection strategies and provides the basis for enhancing our understanding of the pathogen life style. Experiment Overall Design: We investigated global transcriptome response overtime of Mock- and M. oryzae inoculated rice root tissue in vitro. Two independant replicates were perfomed for each treatments and samples were collected at 2, 4 and 6 days post-inoculation.

Data-independent acquisition (DIA)-MS analysis for identification of Magnaporthe oryzae secreted protein induces cell death and triggers defense responses in rice

Project description:The rice blast disease, caused by Magnaporthe oryzae , devastates cultivated rice (Oryza sativa L.), resulting in extensive global crop loss. We employed a label-free quantitative proteomics approach to discover novel proteins associated with M. oryzae pathogenicity and rice defense. We identified 990 significantly modulated proteins in rice leaves including various pattern recognition receptors (PRRs) and pathogenesis-related (PR) proteins that were induced in response to M. oryzae inoculation. Additionally, 123 M. oryzae proteins were also identified and screened for their cell death-inducing activity by an in-silico approach. Among these, we found a novel protein MoXYL1 (endo-1,4-beta-xylanase) protein, which induces cell death in Nicotiana benthamiana leaves. Transgenic rice plants (PDUF26::MoXYL1) expressing MoXYL1 derived by rice domain of unknown function protein 26 (DUF26) promoter exhibited resistance against the M. oryzae and Cochliobolus miyabeanus and enhanced expression of pathogen-responsive genes and hormone-related genes. Furthermore, the application of data-independent acquisition (DIA) mass spectrometry (MS)-based proteomics on these transgenic rice plants revealed 1,833 significantly modulated proteins in response to M. oryzae, with 219 and 410 proteins responsive to MoXYL1 and M. oryzae, respectively. Based on these results, we propose a signaling network model induced by MoXYL1 and M. oryzae. In summary, our findings highlight the crucial role of MoXYL1 in rice innate immunity against M. oryzae and its potential to enhance rice disease resistance.
