Ancistrocladinium A induces cell death in drug-resistant myeloma cells via targeting transcription and RNA splicing machinery and ATF4-dependent apoptotic pathway: a promising candidate as a drug for multiple myeloma

Project description:N,C-coupled naphthylisoquinoline alkaloids like the natural product ancistrocladinium A, was known for its strong antiprotozoal activity, but had never explored for its antitumor activity. We found the alkaloid to be a very efficient anti-myeloma agent in various cell lines, including proteasome inhibitor-resistant cell lines and patient's primary CD138+ myeloma cells either alone or in combination approaches with the compounds carfilzomib or panobinostat. To gain further insight, we used an MS-based approach with the biotinylated derivative of Ancistrocladinium A, leading to an enrichment of RNA splicing associated proteins. We validated alternative pre-mRNA splicing by analyzing RNA gene expression and splicing using the human Clariom D gene array after treatment of INA-6 cells with the alkaloid. Important pathways with altered RNA expression levels included RNA processing, transcription and splicing, and the ribonucleoprotein complex. Pathways with altered RNA splicing included PSMB5 target genes and, therefore, a driver of unfolded protein response (UPR) along with transcription-associated and cell cycle-associated genes. Furthermore, we confirmed the splicing-induced protein expression of activating tran-scription factor 4 (ATF4), a key regulator of UPR. Finally, histone 2A (H2A) phosphorylation and PARP-1 cleavage as an indicator of apoptosis indicator was also detected. Taken together, an-cistrocladinium A can be considered as a potent agent against PI-resistance in MM either alone or in combination with clinically approved drugs.

Gene expression in human umbilical cord endothelial cells following FoxP1 knockdown

Project description:To search for genes regulated by the transcription factor Forkhead box Protein P1 (FoxP1) in endothelial cells, we transfected human umbilical cord endothelial cells (HUVECs) with a combination of three siRNA oligonucleotides directed against human FoxP1 . Human umbilical vein endothelial cells (HUVECs) were isolated from donated umbilical cords, pooled from two donors and cultivated up to passage 5. For transfection with FoxP1-siRNA cells were cultured to 70% confluence and transfected with a combination of three FoxP1-targeting siRNAs or an irrelevant control oligonucleotide (all from invitrogen) using Lipofectamin RNAiMax (Invitrogen) according to the manufacturers instructions. Total RNA was isolated 48h after transfection.

[E-MEXP-3786] IGF-I-induced chronic gliosis and retinal stress lead to neurodegeneration in an animal model of retinopathy

Project description:Transcription profiling by array of mouse male retinas to investigate IGF-I-induced chronic gliosis and retinal stress IGF-I exert multiple effects in different retinal cell populations in both physiological and pathological conditions. Transgenic mice overexpressing IGF-I in the retina showed impaired electroretinographic responses at 6-7 months of age that worsen with age. This retinal neuronal dysfunction was correlated with the loss of rod photoreceptors, bipolar, ganglion and amacrines cells. Neuronal alterations were preceded by the overexpression of retinal stress markers, acute phase proteins and gliosis-related genes. IGF-I overexpression leads to chronic gliosis and microgliosis in TgIGF-I retinas, with mild oxidative stress, impaired recycling of glutamate and defective potassium buffering. These impaired supportive functions can contribute to neurodegeneration in TgIGF-I retinas, together with the increased production of pro-inflammatory cytokines, potential mediators of neuronal death.
