Project description:The rationale: Activation of the immune response antagonizes plant growth and development in absence of pathogen, and such autoimmune phenotype is often suppressed by the elevation of ambient temperature. However, the molecular regulation of ambient temperature-sensitive intersection of immune response and growth is largely elusive. Methods: A genetic screen identified an Arabidopsis mutant, zed1-D, by its high temperature-dependent growth retardation. A combination of molecular, cytological and genetic approaches was used to investigate the molecular basis behind the temperature-sensitive growth and immune response in zed1-D. Key results: A dominant mutation in HOPZ-ETI-DEFICIENT 1 (ZED1) is responsible for a high temperature-dependent autoimmunity and growth retardation in the zed1-D. The autoimmune phenotype in the zed1-D is dependent on the HOPZ-ACTIVATED RESISTANCE 1 (ZAR1). ZED1 and some ZED1-related kinases (ZRKs) are induced by elevated temperature and function cooperatively to suppress the immune response by modulating the transcription of SUPPRESSOR OF NPR1-1 CONSTITUTIVE 1 (SNC1) in absence of pathogen. Main conclusion: Our data reveal a previously unidentified role of ZRKs in ambient temperature-sensitive immune response in absence of pathogen, and thus disclose a possible molecular mechanism underlying temperature-mediated intersection of immune response and growth in plants.

Project description:High ambient temperature regulated the plant systemic response to the beneficial endophytic fungus Serendipita indica. Most plants in nature establish symbiotic associations with endophytic fungi in soil. Beneficial endophytic fungi induce a systemic response in the aboveground parts of the host plant, thus promoting the growth and fitness of host plants. Meanwhile, temperature elevation from climate change widely affects global plant biodiversity as well as crop quality and yield. Over the past decades, great progresses have been made in the response of plants to high ambient temperature and to symbiosis with endophytic fungi. However, little is known about their synergistic effect on host plants. The endophytic fungus Serendipita indica colonizes the roots of a wide range of plants, including Arabidopsis. Based on the Arabidopsis-S. indica symbiosis experimental system, we analyzed the synergistic effect of high ambient temperature and endophytic fungal symbiosis on host plants. By transcriptome analysis, we found that DNA replication-related genes were significantly upregulated during the systemic response of Arabidopsis aboveground parts to S. indica colonization. Plant hormones, such as jasmonic acid (JA) and ethylene (ET), play important roles in plant growth and systemic responses. We found that high ambient temperature repressed the JA and ET signaling pathways of Arabidopsis aboveground parts during the systemic response to S. indica colonization in roots. Meanwhile, PIF4 is the central hub transcription factor controlling plant thermosensory growth under high ambient temperature in Arabidopsis. PIF4 is also involving JA and/or ET signaling pathway. We found that PIF4 target genes overlapped with many differentially expressed genes (DEGs) during the systemic response, and further showed that the growth promotion efficiency of S. indica on the pif4 mutant was higher than that on the wild type plants.

Abstract:
Circadian clocks exhibit 'temperature compensation', meaning
that they show only small changes in period over a broad
temperature range. Several clock genes have been implicated in
the temperature-dependent control of period in Arabidopsis. We
show that blue light is essential for this, suggesting that the
effects of light and temperature interact or converge upon
common targets in the circadian clock. Our data demonstrate
that two cryptochrome photoreceptors differentially control
circadian period and sustain rhythmicity across the
physiological temperature range. In order to test the
hypothesis that the targets of light regulation are sufficient
to mediate temperature compensation, we constructed a
temperature-compensated clock model by adding passive
temperature effects into only the light-sensitive processes in
the model. Remarkably, this model was not only capable of full
temperature compensation and consistent with mRNA profiles
across a temperature range, but also predicted the
temperature-dependent change in the level of LATE ELONGATED
HYPOCOTYL, a key clock protein. Our analysis provides a
systems-level understanding of period control in the plant
circadian oscillator.

Project description:The increasing ambient temperature significantly impacts plant growth, development, and reproduction. Uncovering the temperature-regulating mechanisms in plants is of high importance, not only for boosting our plant biology knowledge but also for assisting plant breeders in improving plant resilience to these stress conditions. Numerous studies on the molecular mechanisms by which plants regulate temperature responses revealed that plants employ distinct transcription factors to regulate thermomorphogenesis specific to each tissue type. A significant discovery in this field was the identification of PHYTOCHROME-INTERACTING FACTORs (PIFs) as key regulators of thermomorphogenesis during vegetative growth. PIF4, a regulator of auxin-mediated signaling pathways, is crucial in controlling high-temperature responses. In this study, we screened the temperature responses of the wild type and several PhyB-PIF4 pathway Arabidopsis mutant lines in combined and integrative phenotyping platforms for root in soil, shoot, inflorescence, and seed. We demonstrated that high ambient temperature differentially impacts vegetative and reproductive organs through this pathway. Suppression of the PhyB-PIF4 components mimics the response to a high ambient temperature in wild-type plants. We also identified correlative responses to high ambient temperature between shoot and root tissues. This integrative and automated phenotyping was complemented by monitoring the changes in transcript levels in reproductive organs. Transcriptomic profiling of the pistils from plants grown under high ambient temperature identified key elements that may provide clues to the molecular mechanisms behind temperature-induced reduced fertilization rate, such as a downregulation of auxin metabolism, upregulation of genes involved auxin signalling, miRNA156 and miRN160 pathways, pollen tube attractants.

Project description:Temperature profoundly impacts plant growth and development. Response to temperature by plants is one of most crucial ecological factors determining the natural distribution of species, and the yield potential of crops. Among various temperature responses of plants, there remains a gap in understanding molecular mechanisms underlying growth retardation caused by low temperature. Here, we addressed genetic and epigenetic determinants that mediate growth retardation under low ambient temperature in plants. Transcriptome changes mediated by low ambient temperatures include upregulation of a series of regulatory genes, consequently resulting in growth retardation. Our study identified a regulatory network in which a Polycomb protein, VERNALIZATION INSENSITIVE 3-LIKE 1 (VIL1), and a transcription factor, C-REPEAT BINDING FACTOR 1 (CBF1), operate to control growth under suboptimal low temperature. Under suboptimal low ambient temperature, VIL1 controls dynamic chromatin remodeling through transient chromatin loop formation and nucleosome rearrangement at the CBF cluster. Our study provides insights into how plants reprogram growth and development in response to suboptimal low ambient temperatures.

Project description:Dynamic trimethylation of histone H3 at Lys27 (H3K27me3) affects gene expression and controls plant development and environmental responses. In Arabidopsis thaliana, RELATIVE OF EARLY FLOWERING 6/JUMONJI DOMAIN-CONTAINING PROTEIN 12 (REF6/JMJ12) demethylates H3K27me3 by recognizing a specific DNA motif; however, little is known about how REF6 activates target gene expression after recognition, especially in environmental responses. In response to warm ambient temperature, plants undergo thermomorphogenesis, which involves accelerated growth, early flowering, and changes in morphology. Here we show that REF6 regulates thermomorphogenesis and cooperates with the transcription factor PHYTOCHROME INTERACTING FACTOR 4 (PIF4) to synergistically activate thermo-responsive genes under warm ambient temperature. The ref6 loss-of-function mutants exhibited attenuated hypocotyl elongation at warm temperature, partially due to down-regulation of GIBBERELLIN 20-OXIDASE2 (GA20ox2) and BASIC HELIX-LOOP-HELIX 87 (bHLH87). REF6 enzymatic activity is necessary for warm ambient temperature responses. Together, our results provide direct evidence of an epigenetic modifier and a transcription factor working together to respond to the environment.
