The effect of XUESAITONG on gene expression profile after focal cerebral ischemia in rats

Project description:Xuesaitong injection (XST), a standardized patent Chinese medicine of Panax notoginseng roots (Sanqi in Chinese), has long been used for effective prevention and treatment of stroke in China. However, the mechanisms underlying its effects against ischemic stroke are still poorly understood. In this study, we focused on the polypharmacology of XST against ischemic stroke with a XST-regulated stroke network analysis. Male Sprague Dawley rat model of MCAO and reperfusion were administered XST for 7 days while the control group was not treated. Three conditions were compared with three replicates each. These are: (1) sham; (2) model (3) XST treatment. Whole genome microarray analysis was performed using Affymetrix rat Genome 230 2.0 chips.

Comparison of miRNA between normal rat plasma exosomes and melatonin pretreated rat plasma exosomes

Project description:Stroke places a huge burden on society today, and great of studies were devoted for seeking safe and effective therapeutic strategy to improve the prognosis of stroke. Plasma exosome has exhibited its therapeutic potential against ischemia and reperfusion injury via ameliorating inflammation. To enhance therapeutic potential in patients with ischemic injury, we isolated exosomes from melatonin pretreated rat plasma and assessed the neurological protective effect in a rat model of focal cerebral ischemia. Treatment with melatonin enhanced plasma exosome therapeutic effect against ischemia induced inflammatory response and inflammasome mediated pyroptosis. In addition, we confirmed ischemic stroke induced pyroptotic cell death mainly occurred in microglia, while administration of melatonin treated exosome further effectively decreased infract volume and improved function recovery via regulation of TLR-4/NF-κB signaling pathway. Finally, the altered miRNAs profile in melatonin treated plasma exosomes demonstrated the regulatory mechanisms. This study suggests plasma exosome with melatonin pretreatment might be a more effective strategy for patients with ischemic brain injury. Further exploration of key molecules in plasma exosome may devote more therapeutic value for cerebral ischemic injury.

The transcriptome analysis of Peri-infarct areas of infarction for sham, PT, and Pushen treatment

Project description:We investigated the functional recovery of in ischemic stroke by Photothrombotic stroke (PT) model. RNA-Seq of the peri-infarct cortex at 28 day after PT to clarify the potential mechanisms of pushen capsule against ischemic stroke. Pushen (drug approval number: Z20040074; batch number: 20082403; Suzhong pharma) was dissolved in the vehicle of ultrapure water, and it was administered by oral gavage at 24 hours after surgery continuously for 28 days. Mice in the Sham and Vehicle group were administered with a vehicle of ultrapure water equal to the Pushen group. By applying the cutoﬀs of P <0.05 and |log2 (fold change)|>1, 2369 differentially expressed genes (DEGs) were identified between sham vs PT, and 231 differentially expressed genes (DEGs) were identified between PT vs Pushen.
