Project description:We utilize a finite element model of implantation mechanics to guide the in vitro micropatterning human embryonic stem cells to simulate in vivo stress gradients that form during development. Micropatterned stem cells showed early formation of GATA4+/SOX17+ primitive endoderm due to migration toward regions of high stress, while NANOG+/OCT4+ epiblast was retained in regions of low stress under spontaneous differentiation.

Project description:Synovial fluid was obtained weekly from nine horses throughout a 70-day study period and subjected to small RNA sequencing. Nine skeletally mature Standardbred trotters (seven mares and two geldings, 2.5-7 years old, weighing 397-528 kg) were included in this study. OA was surgically induced in the left middle carpal joint and the right middle carpal joint underwent sham surgery. Under arthroscopic guidance, an osteochondral fragment was created in the left middle carpal joint by chiselling off a fragment on the dorsodistal surface of the radial carpal bone using an 8 mm curved osteotome. The fragment was left adherent to the joint capsule proximally, and an incongruent defect bed of approximately 15 mm in width was created in the exposed subchondral bone between the fragment and radial carpal bone by drilling with an arthroscopic burr. Debris from the procedure was left inside the joint. Sham surgery (arthroscopy alone) was performed in the right middle carpal joint. All portals were sutured using 2-0 propylene (Surgipro, Medtronic Danmark A/S, Copenhagen, Denmark). Both carpi were bandaged with sterile dressings. Bandages were changed at day 2 and removed at day 5. Sutures were removed 10 days after surgery. From 2 weeks following OA induction and onwards, horses were exercised in 2 min of trot (4.4-5.3 m/sec) , 2 min of fast trot/gallop (9 m/sec) and 2 min of trot (4.4-5.3 m/sec) 5 days a week on a treadmill. The horses were euthanized on day 71 or 72 with an overdose of pentobarbital sodium (140 mg/kg, Euthasol Vet, Dechra Veterinary Products, Uldum, Denmark). One horse, Horse 5, was euthanized according to predefined humane endpoints at day 49, as it became lame at the walk.

Project description:Brachydactyly type A1 (BDA1), due to mutations in IHH, is characterized with shortened and/or missing middle phalangeal bones. These mutations alter the capacity and range of IHH signaling, reducing mesenchymal cells recruiting into the cartilage anlagen of a growing digit, result in a shorter middle phalange. However, reason for a missing middle phalange is unclear. Here, in IhhE95K BDA1 mouse, we showed a link to impaired apoptosis normally needed for interzone cavitation, thus a joint is not formed and the middle phalange is deemed absence. Hedgehog interacting partners (PTCH, CDON, GAS1and HHIP) are expressed in the interzone. Our data support a model for interzone cavitation, where CDON/GAS1 function as dependency receptors. Normally, IHH level is low at the center of the interzone, and the “ligand-free” CDON/GAS1 activate cell death for cavitation. In BDA1 joint, this is suppressed due to excess IHH. Our findings provided new insights into the role of IHH and CDON in joint formation, with relevance to hedgehog signaling in developmental biology.

Project description:Cartilage destruction in osteoarthritis (OA) results from disturbed chondrocyte metabolism. Here, we used microarrays to show that TGF alpha and CCL2 are simultaneously upregulated in a rat model of OA and cooperate to drive cartilage degradation. The goals of the experiments included here were to a) characterize gene expression in knee joint articular chondrocytes at various stages of development of OA (2 and 8 weeks after surgical induction of OA), and b) to establish trends in gene expression among groups of genes related to the TGF alpha-EGFR axis, over time, in OA. The model chosen to study these results has been previously validated (Appleton, CT et al, 2007, Arthritis Rheum) and used to describe similar gene expression results at a different time point (4 weeks) after induction of OA. The rat model of OA involves surgical destabilization of the knee joint, followed by forced low-intensity mobilization over several weeks; a sham surgery is used as the control (representing a healthy non-OA knee joint) wherein a surgical incision is made but not structural (i.e. ligamentous) modification is made to the joint. Altogether, our data indicate that TGF and CCL2 cooperate to drive cartilage degradation in osteoarthritis. A total of 12 samples were analyzed. 3 replicates were used per condition: OA surgery at 2 weeks, OA surgery at 8 weeks, Sham (control) surgery at 2 weeks and Sham surgery at 8 weeks. Expression of OA samples was assessed relaitve to Sham (control) expression levels.

Project description:Mounting evidence points to a link between a cancer possessing stem-like properties and a worse prognosis. To understand the biology, a common approach is to integrate network biology with signal processing mechanics. That said, even with the right tools, predicting the risk for a highly susceptible target using only a handful of gene signatures remains very difficult. By compiling the expression profiles of a panel of tumor stem-like cells (TSLCs) originating in different tissues, comparing these to their parental tumor cells (PTCs) and the human embryonic stem cells (hESCs), and integrating network analysis with signaling mechanics, we propose that network topologically-weighted signaling processing measurements under tissue-specific conditions can provide scalable and predicable target identification. All function codes related to this project could be accessed at the supplementary website. We hypothesized that the transcriptional stochastic element of lung tumors modeled by the lung-TSLC networks could be utilized to estimate the prognostic survival times. A scalable, network-based signal-processing model was devised to analyze the transcriptional signal for a single gene or for a set of genes relating to other gene players in the networks. We tested our hypothesis in the following three steps: (1) construct the consensus TSLC networks and the lung-TSLC networks by analyzing the transcriptomes of three panels of cells: the parental tumor cells (PTCs), the cultivated TSLCs, and the human embryonic stem cels (hESCs); (2) develop network topologically-weighted signal processing measurements and apply such measurements to find the survival correlation and to identify potential targets for biological validation; and (3) verify a potential regulatory target - Cbx5 - in another clinical cohort of LACs as well as in knock-down experiments in lung TSLCs functional assays and in animal models.
