Transcription profiling of rat brain hippocampi of animals fed the ketogenic diet (KD) to provide insight into the anticonvulsant action of KD

ABSTRACT: The ketogenic diet (KD) is an anticonvulsant treatment that has been used to manage medically-intractable epilepsies. The KD requires 10-12 days to become maximally effective, suggesting that changes in gene expression are involved in its anticonvulsant action. Using the Affymetrix rat arrays (RAE230A), 6 control samples and 5 KD samples were run.

Objective
The full anticonvulsant effect of the ketogenic diet (KD) can require weeks to develop in rats, suggesting that altered gene expression is involved. The KD typically is used in pediatric epilepsies, but is effective also in adolescents and adults. Our goal was to use microarray and complementary technologies in adolescent rats to understand its anticonvulsant effect.
Methods
Microarrays were used to define patterns of gene expression in the hippocampus of rats fed a KD o ...[more]
