Project description:Experimental studies confirmed n-6 type polyunsaturated fatty acid as pro-carcinogenic factor and n-3 fatty acid as cancer restraining agent; though their mode of action on tumor cells are still unclear. To review the contrasting effect of omega-3 and omega-6 fatty acids over carcinoma by studying genomic alteration in treated cancer cells, two members from each family of PUFAs, namely EPA and DHA from n-3 PUFA family and, AA and LA from n-6 PUFA family was selected to treat four breast cancer cell lines: MDA-MB231, MDA-MB435S, HCC2218 and MCF-7 for two time period- 6hr and 24hr.

Project description:Experimental studies confirmed that seleium induced a global change of gene expression in HuH7 cells. Several novally induced secretory growth factors may contribute to the autocrine mode of growth elicited by selenium treatment. Keywords: Treatment HuH7 were cultured in 0.1 μM sodium selenite-containing medium for 15 passages (SELT) or in serum-free medium for 30 h (SF30) or in the regular complete medium containing 10 % fetal calf serum (HUH7) for 30 h. The total RNA were reverse transcried and hybridized to Human 8K-1 (Egenomix Technology Corp., Taiwan). The hybridization time and temperature was 16 h and 62â??, respectively. SELT and HUH7 samples were labeled with Cy5 while SF30 sample was labeled with Cy3. Local background correction and interchip global normalization were accomplished by GenePix Pro 5.0 (Molecular Devices, Sunnyvale, CA ). Genes with differential up- or down-regulation by 1.5 fold with respect to the SF30 cells were obtained using GeneSpring software 7.0 (Silicon Genetics, Agilent Technologies).

Project description:The optimal ratio of omega-6 to omega-3 polyunsaturated fatty acids (PUFAs) is important for keeping homeostasis of biological processes and metabolism, yet the underlying biological mechanism is poorly understood. The objective of this study was to identify changes in the pig liver transcriptome induced by a diet enriched with omega-6 and omega-3 fatty acids, and to characterize the biological mechanisms related to PUFA metabolism. Polish Landrace pigs (n =12) were fed diet enriched with linoleic acid (LA, omega-6) and alpha-linolenic acid (ALA, omega-3 family) or standard diet as a control. The fatty acids profiling was assayed in order to verify how feeding influenced the fatty acids content in liver, and subsequently next-generation sequencing (NGS) was used to identify differentially expressed genes (DEG) between transcriptomes between dietary groups. The biological mechanisms and pathway interaction networks were identified by analysis in DAVID and Cytoscape tools. Fatty acids profile analysis indicated a higher contribution of PUFAs in liver for LA and ALA-enriched diet group, particularly for the omega-3 fatty acids family, but not omega-6. Next-generation sequencing identified 3,565 DEG, 1,484 of which were induced and 2,081 were suppressed by PUFA supplemenation. Low ratio of omega-6/-3 fatty acids resulted in modulation of fatty acids metabolism pathways and over-representation of genes involved in membrane composition, signal transduction and immune response pathways. In conclusion, a diet enriched with omega-6 and omega-3 fatty acids altered the transcriptomic profile of the pig liver and affected a set of genes involved in metabolic pathways important to animal health status. Hepatic mRNA profiles of Polish Landrace pig breed fed two different diets, were generated by deep sequencing, using Illumina MiSeq. Experimental diet was enriched with polyunsaturated fatty acids (omega-6 and omega-3), while standard diet remain as a cotrol. 2 pooled samples each containing RNA extracts from 6 individuals livers were analyzed.

Project description:In this study, we have examined the effect of combined prenatal Lactobacillus reuteri and omega-3 fatty acid supplementation on DNA methylation patterns in neonatal immune cells from children with a high propensity of developing allergies. We addressed this matter by assessing epigenome-wide DNA methylation patterns in CD4+ T helper cells obtained at birth, using the Illumina MethylationEPIC 850K array. Samples were retrieved from all four arms of the on-going PRObiotics and OMega-3 (PROOM-3, ClinicalTrials.gov-ID: NCT01542970) allergy prevention trial, and treatment-induced differential methylation as well as network modules were assessed.

Project description:In this study, we present an investigation of the yeast response to the exposure to octanoic and decanoic acids, two major fermentation inhibitors. Aerobically grown cells of Saccharomyces cerevisiae U13 wine strain have been exposed to 0,05 mM of octanoic and decanoic acid. We have compared the variations of expression induced by the acid challenges in comparison to a reference non trated modality, as well as the two acid responses. The transcriptome analysis is build in a triangular design in which the RNA extracted from three modalities : reference (non treated cells) , cells exposed to octanoic acid, and cells exposed to decanoic acid are compared. 12 samples have been analyzed in a triangular design (4 samples per modalité), four references (non treated cells), four samples exposed to octanoic acid, four samples exposed to decanoic acid

Project description:Omega-3 and omega-6 polyunsaturated fatty acids (PUFA) have important signalling roles in the body. The goal of this study was to investigate the impact of linoleic acid (LA, omega-6) and alpha-linolenic (ALA, omega-3) on global skeletal muscle gene expression. We were also interested to study the impact of these fatty acids on myokine expression. To differentiate the roles of essential dietary PUFA on skeletal muscle function, we fed male rats a control diet (AIN-93G) or diets containing 10% safflower oil or flaxseed oil. Skeletal muscle gene expression was investigated by microrray.
