Transcription profiling of human epithelial stem cells co-cultured with the dermal papilla

ABSTRACT: Clonogenic keratinocyte stem cells isolated from the bulge area of human telogen follicles were co-cultured with dermal papilla cells in a transwell system. RNA was isolated from stem cells for different periods of time (day 0, 1, 2, and 5) after co-culture with DP and analyzed for changes in gene expression using Genechip microarrays.

The epithelial-mesenchymal interactions between keratinocyte stem cells and dermal papilla (DP) cells are crucial for normal development of the hair follicle as well as during hair cycling. During the cyclical regrowth of a new lower follicle, the multipotent hair follicle stem cells are stimulated to proliferate and differentiate through interactions with the underlying mesenchymal DP cells. To characterize the events occurring during the process of epithelial stem cell fate determination, we u  ...[more]
