Transcription profiling of soft tissue sarcoma: Expression data from Complex genetics sarcomas (cohort 1 and 2)

ABSTRACT: For this study, we selected, from the French Sarcoma Group (FSG) database, soft tissue sarcomas with no recurrent chromosomal translocations and for which a frozen tissue of the untreated primary tumor was available. Three hundred and ten sarcomas have been studied. They are split in two cohorts. We used microarrays to detail the global programme of gene expression underlying the tumor biology Every cases has been histologically reviewed by the pathologist subgroup and classified according to the 2002 WHO classification using histology, immunohistochemistry and molecular genetic and cytogenetic when needed

Sarcomas are heterogeneous and aggressive mesenchymal tumors. Histological grading has so far been the best predictor for metastasis-free survival, but it has several limitations, such as moderate reproducibility and poor prognostic value for some histological types. To improve patient grading, we performed genomic and expression profiling in a training set of 183 sarcomas and established a prognostic gene expression signature, complexity index in sarcomas (CINSARC), composed of 67 genes related  ...[more]
