F1 allele specific gene expression

Project description:By comparing mouse fibroblasts from two parental strains (Bl6 and Spretus) with fibroblasts from their first generation offspring (F1) we can detect allele specific expression of proteins. The Bl6 and Spretus lines are evolutionary distant and harbour many SNPs in their genomes which when synonomous we can detect on the protein level using mass spectrometry. By mixing SILAC labeled Bl6, Spretus and F1 offspring cell lines we can detect peptides shared between all three cell lines and also SNP peptides that are only expressed in the F1 cells and either Bl6 or Spretus cells. By comparing the abundance of the shared peptides and the SNP peptides we can quantify how much of a protein in the F1 cells that comes from the paternal or maternal allele. This data were then further compared to polysome profiling data. Azidohomoalanine labeling was used to enrich newly synthesized proteins from the three cell lines.

Activation of Ftz-F1-responsive genes through Ftz/Ftz-F1 dependent enhancers

Project description:The orphan nuclear receptor Ftz-F1 is expressed in all somatic nuclei in Drosophila embryos but mutations result in a pair-rule phenotype. Previously characterized Ftz-F1 target genes were co-regulated by Ftz, which is expressed in stripes, consistent with the pair-rule phenotype observed for ftz-f1 and ftz mutants. However, attempts to identify new target genes on the basis of Ftz-F1 and Ftz binding sites alone has met with only limited success. To discern the rules for Ftz-F1 target site selection in vivo and to identify additional target genes, a microarray analysis was performed comparing wildtype and ftz-f1 mutant embryos.

Gene expression in leaf blades of the rice Os-LBD37 overexpressor line RK16331-13

Project description:The 5th and 6th leaf blades of the rice Os-LBD37 overexpressor line RK16331-13 and the empty vector control line FOX3 were examined. LBD37 belongs to the plant- specific LOB- (Lateral Organ Boundary) domain family proteins first characterized in Arabidopsis. Results point towards an involvement of the rice LBD37 (OsLBD37) ortholog of Arabidopsis in nitrogen metabolism- and senescence- related processes.

Recipe for a busy bee: MicroRNAs in honey bee caste determination (microRNA array)

Project description:Social caste determination in the honey bee is assumed to be determined by the dietary status of the young larvae and translated into physiological and epigenetic changes through nutrient-sensing pathways. We have employed microRNA gene-microarray, and observed that both worker jelly and royal jelly showed dynamic changes in miRNA content during the 4th to 6th day of larval development . Adding specific miRNAs to royal jelly elicited significant changes in queen larval mRNA expression and in morphological characters of the emerging adult queen bee. We propose that miRNAs in the nurse bee secretions constitute an additional element in the regulatory control of caste determination in the honey bee. We collected worker and royal jelly of the Italian honeybee (ZND No.1, Apis mellifera ligustica) at 73~90 hours (4th-day larvae), 97~114 hours (5th-day larvae), and 121~138 hours (6th-day larvae) after hatching. After total RNA was extractedM-BM- and quantified , equal amounts of total RNAs from each of the three sampling days were analyzed on the LC Science miRNA-array to observe the expression variation of miRNAs between worker jelly and royal jelly along with the development time points (4th-day, 5th-day and 6th-day).

Project description:Intervention1: Warm foot bath therapy: In experimental arm, Warm foot bath therapy Each session-15min. Participants will receive warm foot bath therapy prior oxaliplatin for 15min.and post oxaliplatin for 15min.on 1st day of 2nd,3rd,4th,5th,6th chemotherapy cycle and in home at night once for 15min.till completion of 6cycles. In control arm, Participants will not receive the above intervention; but only receive routine treatment. Control Intervention1: standard routine treatment No interventions: In experimental arm, Warm foot bath therapy Each session-15min. Participants will receive warm foot bath therapy prior oxaliplatin for 15min.and post oxaliplatin for 15min.on 1st day of 2nd,3rd,4th,5th,6th chemotherapy cycle and in home at night once for 15min.till completion of 6cycles. In control arm, Participants will not receive the above intervention; but only receive standard routine treatment. Primary outcome(s): To evaluate the effects of foot bathing on peripheral neuropathy in patients receiving oxaliplatin containing chemotherapy for colorectal cancer.Timepoint: Baseline assessment- 1st day of first CT cycle using FACT-GOG NTX tool -Assessment of CIPN on 1st day of 2nd,3rd,4th,5th,6th chemotherapy cycle and assessment of CIPN and associated QOL using FACT-GOG NTX tool on 1st day of 2nd and 6th chemotherapy cycle and three months after completion of 6th cycle and six months after completion of 6th cycle.

F1 hybridization effects on mouse oocytes

Project description:This dataset consists of germinal vesicle (GV) and metaphase II (MII) oocytes from B6, D2, and B6xD2 F1 mice. Oocytes from a single mother were pooled as one biological replicate to become one sample. The goal of the study was to explore the genetic effects on oocyte quality, and more specifically, to determine a viable explanation for the hybrid vigor phenotype displayed in F1 mice. This study is the first to combine oocytes of different stages and different genotypes (two parental inbred strains and F1 hybrid) into a single analysis capable of identifying molecular characteristics that distinguish F1 hybrids.
