Effects of purine de novo synthesis knockdown on gene expression in Drosophila melanogaster S2 tissue culture cells

ABSTRACT: The effect of inhibiting expression of the ade2 gene by RNA interference in Drosophila S2 tissue culture cells was examined. S2 cells underwnt three different dsRNA treatments. Two were for non-overlapping sequences of ade2 transcripts and the third was for a non-specific Chlamydomonas reinhardtii lhcb sequence. 3 samples from ade2 dsRNA-1 treated cells, 2 samples from ade2 dsRNA-2, and 3 samples from lhcb dsRNA treated cells were used on two 4x44K format slides (G2519F).

Endocytosed dsRNAs induce lysosomal membrane permeabilization that allows cytosolic dsRNA translocation for Drosophila RNAi response

Project description:RNA interference (RNAi) is a gene-silencing mechanism triggered by the cytosolic entry of double-stranded RNAs (dsRNAs). Many animal cells internalize extracellular dsRNAs via endocytosis for RNAi induction. However, it is not clear how the endocytosed dsRNAs are translocated into the cytosol across the endo/lysosomal membrane. Herein, we show that in Drosophila S2 cells, endocytosed dsRNAs induce lysosomal membrane permeabilization (LMP) that allows cytosolic dsRNA translocation. LMP mediated by dsRNAs requires the lysosomal Cl−/H+ antiporter ClC-b/DmOstm1. In clc-b or dmostm1 knockout S2 cells, extracellular dsRNAs are endocytosed and reach the lysosomes normally but fail to enter the cytosol. Pharmacological induction of LMP restores extracellular dsRNA-directed RNAi in clc-b or dmostm1-knockout cells. Furthermore, clc-b or dmostm1 mutant flies are defective in extracellular dsRNA-directed RNAi and its associated antiviral immunity. Therefore, endocytosed dsRNAs have an intrinsic ability to induce ClC-b/DmOstm1-dependent LMP that allows cytosolic dsRNA translocation for RNAi responses in Drosophila cells.

Defining an evolutionarily conserved role of GW182 in circular RNA degradation.

Project description:By using dsRNA screening, we found that GW182 is critical for the degradation of circRNA in the Drosophila cells. To further confirm our finding genome widely, S2 cells were treated with either β-gal dsRNA (as a control) or GW182 dsRNA for 3 days and total RNA was extracted and sequenced after ribosome depletion. By comparing the expression level of both circRNAs and linear mRNAs between controls and GW182 depleted samples, we confirmed that GW182 depletion led to accumulation circRNAs but not their parental linear mRNAs, indicating that GW182 may play an import role in the degradation of circRNAs.
