Effect of Griffithsin on gene expression profile in a human ecto-cervical cell line Ect1/E6E7

ABSTRACT: Treatment of cells by different concentrations of Griffithsin resulted in minor alterations in gene expression Cells were treated with Griffithsin or controls (Cyanovirin-N, lectin deficient Griffithsin termed mGRFT and PBS) and incubated overnight (16 h). Gene expression was compared to that of the PBS treated cells to derive fold changes. Each treatment was done in triplicates. Benjammini and Hochberg correction was used and we considered a fold change of at least 2.0 compared to the PBS treated samples to represent a meaningfully altered gene expression.

Transcription factors MYOCD, SRF, Mesp1 and SMARCD3 significantly enhance the cardio-inducing effect of GATA4, TBX5, and MEF2C during direct cellular reprogramming

Project description:Transient over-expression of defined combinations of master regulator genes can effectively induce cellular reprogramming: the acquisition of an alternative predicted phenotype from a differentiated cell lineage. This can be of particular importance in cardiac regenerative medicine wherein the heart lacks the capacity to heal itself, but simultaneously contains a large pool of fibroblasts. In this study we determined the cardio-inducing capacity of ten transcription factors to actuate cellular reprogramming of mouse embryonic fibroblasts into cardiomyocyte-like cells. Over-expression of transcription factors MYOCD and SRF alone or in conjunction with Mesp1 and SMARCD3 significantly enhanced the basal but necessary cardio-inducing effect of the previously reported GATA4, TBX5, and MEF2C. In particular, combinations of five or seven transcription factors significantly enhanced the activation of cardiac reporter vectors, and induced an upregulation of cardiac-specific genes. Global gene expression analysis also demonstrated a significantly greater cardio-inducing effect when the transcription factors MYOCD and SRF were used. Detection of cross-striated cells was highly dependent on the cell culture conditions and was enhanced by the addition of valproic acid and JAK inhibitor. Although we detected Ca2+ transient oscillations in the reprogrammed cells, we did not detect significant changes in resting membrane potential or spontaneously contracting cells. This study further elucidates the cardio-inducing effect of the transcriptional networks involved in cardiac cellular reprogramming, contributing to the ongoing rational design of a robust protocol required for cardiac regenerative therapies. Mouse embryonic fibroblasts were transduced lentiviruses allowing the inducible overexpression of three unique sets of transcription factors and negative control. Project description:To gain deeper insight into the mechanism of toxicity, it is important to identify and characterize miRNAs profiles involved in responses to specific classes of toxicants in conjunction with their impact on gene expression levels. However, few reports have described the effects of toxicants on miRNA expression profiles. Taking into account the prominent role of miRNAs in cancer development, progression, cell cycle control, and proliferation-related processes, it is likely that miRNAs are involved in the toxic response induced by carcinogens. Polycyclic aromatic hydrocarbons (PAHs) are a well-characterized class of human carcinogens. In the present study, we documented the different expression profiles of miRNAs in environmental carcinogen-exposed HepG2 cells by miRNA microarray analysis. To evaluate the change in miRNA expression levels, human hepatocellular carcinoma (HepG2) cells were exposed to two PAHs (benzo[a]anthracene, benzo[k]fluoranthene) for 48 h. miRNA expression analysis was conducted using a 8x16k human miRNA microarray (Agilent Technologies, USA).

Analysis of milk removal frequency on bovine mammary gene expression

Project description:Regulation of milk synthesis and secretion is controlled mostly through local (intra-mammary) mechanisms. To gain insight into the molecular pathways comprising this response, an analysis of mammary gene expression was conducted in 12 lactating cows shifted from twice daily to once daily milking. Tissues were sampled by biopsy from adjacent mammary quarters of these animals during the two milking frequencies, allowing changes in gene expression to be assessed within each animal. Using bovine-specific, oligonucleotide arrays representing 21,495 unique transcripts, a range of differentially expressed genes were found as a result of less frequent milk removal. A total of 12 microarrays were used in this study. Each array was hybridized with RNA derived from a mammary sample taken at each biopsy time point, such that the twice-a-day milked sample and once-a-day milked sample were contained on the same slide for each animal (12 animals). Six of the microarrays represented animals previously shown to demonstrate a âlargeâ reduction in milk yield, whereas the other six arrays represented animals that previously displayed a âsmallâ reduction in milk yield when milked once per day. Half of each yield loss group (three animals) was represented by once-a-day milked/twice-daily-milked samples for Cy5/Cy3 labelling, with the other half labelled with the opposite dye configuration. This meant that six of 12 arrays were 'dye swapped', but with biological replicates as opposed to identical replicates.

Transcription profiling of liver, kidney and brain from male and female Zebra Finches

Project description:Each array hybridization involved one male sample hybridized in competition to one female sample. For each hybridization one sample was labeled with Cy5 and the other with Cy3. Half of the male samples were labeled with Cy5 and rest with Cy3. Each hybridization was tissue-specific (i.e., hybridize male brain vs. female brain, male liver vs. female liver, etc.). A total of 10 male-female hybridizations were performed using adult brain, 7 for liver, 9 for kidney and 8 for postnatal day1 brain. One hybridization each from adult brain and liver were discarded because the hybridization intensities were far out of range.

Gene expression of human skin fibroblast cells derived from hypospadias patients in response to low-dose bisphenol A exposure

Project description:To better understand the molecular basis of the reproductive health effects of bisphenol A (BPA) on humans, a genome-wide screening was applied to identify novel targets of low-dose bisphenol A exposure in huamn skin fibroblast cells (hSFCs) derived from hypospadias patient children. Three hSFCs were collected at National Research Institute for Child Health and Development, Japan. Gene expression profiles of hSFCs were measured at 24 hours after exposure to 10nM BPA, 0.01nM 17β-estradiol (E2) and 1nM 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). Differentially expressed genes following chemical treatments were identified by unpaired Student’s t test with P values cut off by 0.05 and fold change of more than 1.2 and selected for the network generation and pathway analysis using Ingenuity Pathways Analysis (IPA) program. As the result, 71 genes (42 downregulated and 29 upregulated), 814 genes (371 downregulated and 443 upregulated), and 824 genes (344 downregulated and 480 upregulated) were identified significantly differently expressed in response to BPA, E2, and TCDD, respectively. The network analysis indicated that the most associated network fuctions of genes genes with altered expression profile derived from microarray analysis were “Endocrine System Disorders, Gastrointestinal Disease, Genetic Disorder”, “Cellular Growth and Proliferation, Skeletal and Muscular System Development and Function, Cell Cycle”, and “Post-Translational Modification, Genetic Disorder, Hematological Disease” in response to BPA, E2, and TCDD, respectively. Gene expression profiles of 3 hSFCs derived from hypospadias patient children were measured at 24 hours after exposure to 10nM BPA, 0.01nM E2 and 1nM TCDD.
