Affy_med_2012_03-Characterization of new phytostimulatrices molecules for the improvement of plants products and crop resistance to diseases

ABSTRACT: affy_med_2012_03 - affy_med_2012_03 - Aphanomyces euteiches is a major legume parasite related to S. parasitica. Analysis of its cell wall showed the presence of an unsusual high content in GlcNAc, representing 10% of the total carbohydrates. Interestingly, this compound does not form crystalline chitin, but amorphous chitosaccharides. The aim goal of this study is to compare the response to a preparation of crab shell chitin chitooligosaccharides with a mean DP of 7 and to A.euteiches chitosaccharides at a transcriptomic level. This comparison will be done on Medicago truncatula wild type compared to the nfp-1 and nfp-2 mutants. - 7-day-old A17 wild type or mutant nfp-1 or nfp-2 plants were grown in vitro on liquid M medium. The entire roots were then treated (or not = controls) with a preparation of crab shell chitin chitooligosaccharides of a mean DP of 7 or with chitosaccharides from A. euteiches cell wall, names fraction P2F1, and harvested after 4 hours of incubation. Project description:affy_infection_medicago - affy_infection_medicago - HAP2a is a transcription factor involved in the regulation of nitrogen fixing nodule organogenesis. hap2a-1 mutant has been shown to be more resistant to Aphanomyces euteiches (Ae), a root pathogen, than the A17 wild type (WT) line. In this study we want to compare both WT and mutant plant responses, 1 and 6 days post inoculation (dpi) between WT and the hap2a-1 mutant, in order to identify HAP2a-dependent gene networks during infection. Data about non inoculated A17 control plants and infected A17 at 1 dpi have been already obtained in another experiment in which the conditions of in vitro culture of M. truncatula plants and ofA.euteiches(Ae) inoculation were exactly the same than the ones used in these experiments.-Fifteen-day-old A17 or hap2a-1 plants were grown in vitro on M medium. Influence of Nod Factors on early developmental stages of root development in Medicago truncatula

Project description:We would like to know how symbiotic molecules such as Nod Factors (NF) influence lateral root (LR) development in M. truncatula. We have preliminary evidence that this action is through early stages of root development. Auxin is the major phytohormone controlling LR development and we also have evidence that NF interfere with auxin for the control of LR development. This transcriptomic study aims at finding new molecular targets that would be responsive to auxin and NF treatment, even at a higher level by the combination of both auxin and NF. Such targets could be involved in the control or early development stages of LR that would be controlled both by auxin and NF treatment. - 2 day old M. truncatula (accession A17) plantlets grown on M medium were transferred for 2 days on M medium+ 10-5M NPA (1-N-Naphthylphthalamic acid, auxin transport inhibitor) then transferred for 10 h on M medium containing 10-6 M NAA (naphthalene-1-acetic acid, permeant auxin analog) or 10-7 M Nod factors (NF) or a combination of both (10-6 M NAA + 10-7 M NF). Project description:The involvement of ROS in the legume – Rhizobium symbiotic interaction has been highlighted (Santos et al., 2001; Rubio et al., 2004). This interaction is characterized by the formation of a new organ on the root, the nodule and by the penetration, in parallel, of the bacteria into the root tissue via an infection thread (IT) (Parniske and Downie, 2003; Gage, 2004). H2O2 production has been shown in ITs during the Medicago – Sinorhizobium meliloti interaction (Santos et al., 2001). Moreover, S. meliloti mutants impaired in H2O2 detoxification mechanism possess in planta symbiotic phenotypes. As H2O2 production is known to orchestrate plant gene expression, our goal is focused on identifying the H2O2 regulated genes in the symbiotic process. We focus our analysis in the M. truncatula transcriptome as we are characterising the S. meliloti H2O2 transcriptome. -M. truncatula seedlings were grown in axenic condition on modified Fahraeus medium during seven days. Then they were transferred on new plates supplemented either with dimethyl sulfoxide (DMSO, mock treatement) or diphenylene iodonium (DPI, 10 µM). Twenty four hours later, they were inoculated either with water (mock inoculation; DMSO-H2O and DPI-H2O) or wild type S. meliloti 2011 strain (DMSO-INOC and DPI-INOC). Roots without apexes were harvested 48 hours after inoculation. Project description:affy_rice_2011_03 - affy_compartimentation_rice_albumen_embryon - During germination, the rice seed goes from a dry quiescent state to an active metabolism. As with all cereals, the rice seed is highly differentiated between the embryo (that will give rise to the future plantlet) and the endosperm (that contains the seed storage compounds and that will degenerate). The molecular mechanisms operating in the rice seed embryo have begun to be described. Yet, very few studies have focused specifically on the endosperm during the germination process. In particular, the endosperm is mostly addressed with regards to its storage proteins but we have detected a large protein diversity by two-dimensional electrophoresis. Similarly, the endosperm is rich in total RNA which suggest that gene expression coming from seed maturation could play a role during the germination process. In this context, we want to compare the transcriptome of the embryo and the endosperm during rice seed germination. -We germinate rice seeds of the first sequenced rice cultivar i.e. Nipponbare during 0, 4, 8, 12, 16 and 24h of imbibition in sterile distilled water. Germination occurs under constant air bubbling, in the dark at 30M-BM-0C. These rice seeds are then manually dissected into embryo and endosperm fractions. -The embryo-derived samples are abbreviated in M-bM-^@M-^\EM-bM-^@M-^] while the endosperm samples are abbreviated M-bM-^@M-^\AM-bM-^@M-^]. The germination time-point is indicated after the letter (e.g. E8 for embryo samples harvested after 8 hours of germination). Project description:affy_strigolactone_sunflower - affy_strigolactone_sunflower - Abiotic stress and more specifically drought is the major limiting factor for sunflower production. Sunflower response to drought includes root plasticity to adapt to water availability and reach soil water. We identified genotype-specific responses of root architecture to strigolactone application. This experiment aims at identifying strigolactone responsive pathways in 6 genotypes in order to better understand molecular control of root development in sunflower and therefore its response to drought. The ultimate goal will be to improve sunflower breeding through selection of key drought response genes.-The experiment consisted of 2 repeats of four 12-day-old-plantlets of 6 sunflower genotype SF193 (INRA code: XRQ), SF326 (INRA code: PSC8), SF056 (INRA code: FU), SF306 (INRA code: PAZ2), SF302 (INRA code: PAC2), SF268 (INRA code: RHA266), grown in growth chamber conditions and submitted to a 24-hour-treatment of 100 nM strigolactone analogue rac-GR24 (Chiralix, Nijmegen, Netherland) or not. Growth conditions were 14h light at 23Â°C and 10h night at 20Â°C under fluorescent bulbs. Plants were grown in hydroponic boxes containing 20 litres of aerated liquid culture medium (as described in Massonneau et al., 2001 Planta). The small RNA diversity from Medicago truncatula roots under biotic interactions evidences the environmental plasticity of the miRNAome

Project description:Plants show a remarkable plasticity to adapt their root architecture to biotic and abiotic constraints of the soil environment. Although some of these modifications are fine-tuned by miRNAs, there are still shadow zones in these regulations. In the model legume Medicago truncatula, we analyzed the small RNA (smRNA) transcriptome of roots submitted to symbiotic and pathogenic interactions. Mapping on the genome and prediction of pre-miRNA hairpins allowed the identification of 416 candidates. Out of them, we found known and novel variants of 77 miRNA families, already reported in miRBase. In addition, thanks stringent criteria of miRNA prediction, 53 mtr-miRNAs were discovered, including 27 putative miRtrons. Exploring polymorphism in 26 M. truncatula ecotypes, higher polymorphism was observed in conserved rather than legume-specific miRNA genes. An average of 19 targets, mainly involved in environmental responses and signaling, was predicted per novel miRNA. In addition, taking advantage of our large number of smRNA libraries, we identified sets of miRNAs responsive to root pathogens or to symbiotic interactions and the related Nod and myc-LCO signals. 23 libraries of small RNA (smRNA) of roots submitted to symbiotic and pathogenic interactions.
