KD of Fibroblast specific TF

ABSTRACT: Combinatorial knock down of LHX9, OSR1, PRRX1, TWIST2, fibroblast specific transcription factors and adipogenic induction by medium change induce transdifferentiation from fibroblast to adipocyte. Adipogenesis from meshencymal stem cell (MSC) and human preadipocyte (PreADP) were used for general reference of adipogenesis. 54 Samples total with 3 replicates each of sample and treatment types.

Effect of Twist2 knockdown on the transcriptional profile of differentiating 3T3-L1 cells

Project description:TWIST2 is a transcription factor heavily involved in proper development. We queried the role of TWIST2 in 3T3-L1 adipogenesis by silencing gene expression using a lentiviral-delivered shRNA and performed RNA-seq at various time points following initiation of differentiation. We found that previously identified TWIST2-target genes increase in transcription, suggesting that TWIST2 acts as a transcriptional repressor in this system.

Direct cell conversion from human dermal fibroblast to adipocyte

Project description:To investigate the effect of ZNF395 on adipogenesis, we tested whether ZNF395 enhance cell conversion from human dermal Fibroblast (FIB) to Adipocyte (ADP). PPARG2 was reported as a master regulator and can induce adipogenesis in non-adipogenic fibroblasts. We transduced PPARG2 with or without ZNF395 in FIB with lentivirus. Interestingly, co-transduction of PPARG2 and ZNF395 showed higher occurrence of adipocyte-like cells as compared with PPARG2 alone. Moreover, genes related with lipid metabolic process and lipid transport was significantly up-regulated in combination of PPARG2 and ZNF395. These results suggest that ZNF395 co-ordinate the transcriptional regulatory pathway with PPARG2, necessary for the induction of adipogenesis. Total RNA was obteined from human dermal fibroblast transduced with mock lentivirus vector (FIB_ctrl), PPARG2 (FIB_PPARG2) and co-transduced with PPARG2 and ZNF395 (FIB_PPARG2+ZNF395).

Regulation of p21 by TWIST2 contributes to its tumor suppressor function in human acute myeloid leukemia

Project description:TWIST2 is deregulated in about 1/3 human acute myeloid leukemia (AML)M-BM- patients partially due to hypermethylation. Overexpression of TWIST2 in various AML cell lines and primary AML cells leads to growth arrest, reduced colony forming cell (CFC) capacity, inhibited cell migration and tumorigenic ability in nude mice, suggesting its tumor suppressor role in AML. To explore the molecular insight of how TWIST2 acts as a tumor suppressor in AML cells, control and TWIST2 expressed THP-1 cells have been analyzed with microarray. 4 independent biological replicates of control and TWIST2 expressed THP-1 cells have been used for RNA extraction and microarray analysis with Agilent platform.

Transcription profiling of preadipocytes of wild type and IRS-1,IRS-2, IRS-3, IRS-4 knock out mice to reveal role of insulin receptor substrates and necdin

Project description:Insulin and IGF-1 promote adipocyte differentiation via complex and overlapping signalling networks. Here we used microarray analysis of brown preadipocytes derived from wild-type and insulin receptor substrate (IRS) knockout (KO) animals, which exhibited progressively impaired differentiation, to define the set of genes that predict adipogenic potential in these cells. 374 genes/ESTs were identified whose expression in preadipocytes correlated with their ultimate ability to differentiate. Many of these genes were related to early adipogenic events, including genes involved in extracellular matrix, cytoskeletal organization, growth arrest, post-mitotic clonal expansion, and inhibitors of adipogenesis, including preadipocyte factor-1 and multiple members of the Wnt-signalling pathway. Reconstitution of IRS-1 KO cells with IRS-1 reversed these changes and restored the ability to differentiate. Several of these genes showed concordant changes in brown adipose tissue in vivo. Necdin was markedly increased in IRS-1 KO cells that could not differentiate, and knockdown of necdin restored brown adipogenesis with down-regulation of Pref-1 and Wnt10a expression. We demonstrated a necdin-E2F4 interaction repressing PPARg transcription. IRS proteins regulated necdin via a CREB dependent pathway, defining a signalling network involved in brown preadipocyte determination.
