Project description:affy_cinetique_lyon_rose. The objective is to identify genes involved in petal development and senescence. R. chinensis cv Old Blush (OB) was used for the following reasons: it is a diploid Chinese rose that participated in the generation of modern roses (recurrent flowering, scent, etc.). The objective here is to identify genes whose expression is associated with different flower development stages, from floral meristem to senescing flower. These genes are putative candidates involved in floral initiation, development and senescence. All samples were collected at the same time early in the afternoon. Meristems and early flower development stages were dissected under a microscope. Total RNA was extracted from harvested tissues using the Plant RNA kit (Macherey Nagel), and then used to hybridize Rosa-Affymetrix microarrays. Project description:Rosa chinensis ‘Pallida’ (Rosa L.) is one of the most important ancient rose cultivars originating from China. It contributed the ‘tea scent’ trait to modern roses. However, little information is available on the gene regulatory networks involved in scent biosynthesis and metabolism in Rosa. In this study, the transcriptome of R. chinensis ‘Pallida’ petals at different developmental stages, from flower buds to senescent flowers, was investigated using Illumina sequencing technology. De novo assembly generated 89,614 clusters with an average length of 428 bp. Based on sequence similarity search with known proteins, 62.9% of total clusters were annotated. Out of these annotated transcripts, 25,705 and 37,159 sequences were assigned to gene ontology and clusters of orthologous groups, respectively. The dataset provides information on transcripts putatively associated with known scent metabolic pathways. Digital gene expression (DGE) was obtained using RNA samples from flower bud, open flower and senescent flower stages. Comparative DGE and quantitative real time PCR permitted the identification of five transcripts encoding proteins putatively associated with scent biosynthesis in roses. The study provides a foundation for scent-related genes discovery in roses.

Project description:affy_brachy_2011_11 - affy_brachy_2011_11 - Fusarium graminearum is the causal agent of Fusarium head blight (FHB) of small-grain cereals, including wheat. Besides direct grain losses, this disease is of major concern because of the production by the pathogen of mycotoxins which are hazardous to animals, thus making the grain unfit for food or feed. Major mycotoxins produced by the fungus are trichothecens, including deoxynivalenol (DON). In our laboratory, we use Brachypodium distachyon as a model plant for cereals because of its amenability (short life cycle, numerous genomic and genetic resources, ...). We have recently shown that F. graminearum does induce head blight symptoms on this species and that DON is produced on infected spikes. We have also evidenced that a F. graminearum strain unable to produce DON exhibits reduced virulence on B. distachyon spikes, as previously shown on wheat. The aim of this project is to analyse and compare the plant response to DON producing and non-producing strains of F. graminearum. This analysis will allow to decipher the mechanisms of detoxification set up by the plant and also more specific responses due to the impact of the mycotoxin on plant metabolism and physiology. -Three conditions on B. distachyon spikes: 1-Mock inoculation (Tween 20 0,01%) 2-Inoculation by a F. graminearum wild-type strain 3-Inoculation by a F. graminearum mutant strain, unable to produce DON Spikes were point inoculated with 3ul of either Tween 20 0.01%, wild-type strain or mutant strain (300 spores) and incubated for 96 hours. Six inoculated spikes were collected and pooled for each condition and biological replicate. Project description:affy_duplicature_lyon_rose. The objective is to identify the genes involved in petal doubling in rose. In this study we are using two rosa gallica genotypes: wild-type (simple flower rose) and Cardinal de Richelieu (double flower rose), and two rosa hybrida genotypes : Souvenir de la Malmaison, which has about 110 petal, and its bud sport cultivar, Souvenir de St Anne’s. In this study, we used a microarray approach to compare the transcriptome of double flower rose (CDR) versus simple flower rose (G). The objective is to identify genes whose expression is associated with the double flower phenotype. These genes are putative candidates involved in the control of petal organ number per flower. Floral buds were dissected under a microscope and pooled in eppendorf tubes. Tissue samples were harvested at the same time during 3 weeks in April 2007. Total RNA was extracted from the pools of flowers using the Plant RNA kit (Macherey Nagel), and then used to hybridize Rosa-Affymetrix microarrays. Project description:affy_ralstonia_peeters_medicago - We have identified two essential virulence determinants (GALA7, a type III secretion effector and HpaP, a chaperone-like protein) of Ralstonia solanacearum for the infection and colonisation of the host plant Medicago truncatula. The scope of this project is to identify the GALA7 and HpaP-specific transcriptome alteration. For this purpose wild type and mutant infected root material (13h and 72h postinfection) will be analysed on M. truncatula affymetrix chips. Medicago truncatula (A17 line) are grown in vitro on Farheus medium (with Nitrogen source) plantlets are inoculated with water R. solanacearum wt, gala7 and hpap mutants, and root tips are collected at 13h and 72h postinoculation. Project description:Chemical communication in mammals is in part mediated through the deposition of scent marks in the environment. Volatile and non-volatile moleculesin the scent marksconvey important information about the donor animal, such as sex, social and reproductive status,and stimulatea response in the receptor animal. In rodents, most of the work about deciphering the molecular composition of scent markshas been focused on mice. In rats, by contrast, knowledge is limitedand the molecular complexity of scent marks is not as well described as in mice. In particular, the sources and roles of proteins in scent marks are largely undefined, with even less knowledge when we consider female rats. We therefore conducted a global proteomic analysis of scent marks from femalerats (Rattus norvegicus)to define the protein composition and then we investigated the role of scent marks in sexual communication. Our study revealed that protein pattern inscent marks greatly differsfrom that in naturally voided urine, indicatinga different tissue of41origin. In particular, female scent marks are dominated by lipocalins, notablymajor urinary proteins, whereas urinary output of these proteins is almost imperceptible.Further analysis revealed that the most plausible source of the components in scent marks are the clitoral glands,implying a critical role of theseaccessory glands in sexual communication in the rat.
