Changes in exosomal circulating microRNAs in grazing cattle

Project description:To investigate the potential effect of grazing movement on miRNA circulation in cattle, here we profiled miRNA expression in centrifugally prepared exosomes from the plasma of both grazing and housed Japanese Shorthorn cattle. Microarray analysis of the c-miRNAs resulted in detection of a total of 231 bovine exosomal miRNAs in the plasma, with a constant expression level of let-7g across the duration and cattle groups. Expression of muscle-specific miRNAs such as miR-1, miR-133a, miR-206, miR-208a/b, and miR-499 were undetectable, suggesting the mildness of grazing movement as exercise. Changes in miRNA expression in plasma exosome of cattle was measured at during 0, 1, 2 and 4 months of grazing or housing. Plasma exosome samples prepared from three cattle for each treatment were collected and mixed within the treatment at each time for microarray analysis .

Expression data from cold stress in Sanhe cattle

Project description:While cold stress has been shown to seriously impact cattle industry, there are only a few reports investigating the effect of cold stress on cattle. Whether severe cold stress results in alterations in gene expression and affects molecular genetic mechanisms remains unknown. We used microarrays to analyze the alterations in gene expression in peripheral blood samples in response to cold exposure and identified differentially regulated genes. This study was performed on 30 healthy Sanhe heifers with similar genetic backgrounds, weight, and age. In order to induce cold stress, the cattle were transferred outdoor and were exposed to a temperature of -32Â°C for 3 hours followed by housing in cowshed at 5Â°C for 15 hours. Blood samples with EDTA were collected from each animal before and after the cold exposure. After total RNA was isolated from blood cells, six RNA samples (three derived before and three after the cold exposure), were collected from three animals randomly selected from the 30 healthy heifers for gene expression profiling in response to severe cold stress.

Early response of bovine alveolar macrophages to infection with live and heat-killed Mycobacterium bovis

Project description:Bovine tuberculosis, caused by Mycobacterium bovis, is a disease of considerable economic importance yet comparatively little is known about the bovine immune response to the disease. Alveolar macrophages are one of the first cells to encounter mycobacteria following infection. In this experiment we investigated the early transcriptional response of bovine alveolar macrophages following infection with M. bovis. The transcriptional response to heat-killed M. bovis was also investigated to look for genes that are only differentially transcribed in response to the live organism. Five-condition experiment, uninfected, live and heat-killed M. bovis-infected bovine alveolar macrophages from five cattle infected for two and four hours. Comparisons were within animal. Dye swaps were incorporated into the design.

Transcriptomic characterization of cattle primary hepatocytes and its comparison with Madin-Darby bovine kidney (MDBK) cells and liver tissue

Project description:Cattle primary cultured hepatocytes (CHs) have been used as a useful in vitro model for drug metabolism studies. However, they suffer some limitations, when compared with the ex vivo liver tissue, such as difficult adhesion to the substrate following isolation, a limited viability and a rapid loss of differentiated characteristics. Therefore, a reliable hepatic in vitro model is actually needed for cattle. As established bovine hepatic cell lines are not available, we characterized the cattle primary CHs' transcriptome by using a cDNA microarray approach and compared it with ex vivo cattle liver tissue as well as with the Madin-Darby Bovine Kidney (MDBK) cells (as an established cell line for transfection purposes).<br>This part of the study compares the primary CHs vs. liver tissue. The comparison of MDBK cells vs. the primary CHs is found under E-MTAB-4430.

Project description:Aging is a physiological process occurring in all living organisms. It is characterized by a pro-gressive deterioration of the physiological and cognitive functions of the organism, accompanied by a gradual impairment of mechanisms involved in the regulation of tissue and organ homeo-stasis, thus exacerbating the risk of developing pathologies, including cancer and neurodegener-ative disorders. In the present work, we sought to investigate the influence of aging upon the expression protein profile in the brain cortex of grazing cattle, through LC-MS/MS-based differ-ential proteomics and bioinformatic analysis approach. We found a significant downregulation of proteins involved in synaptic plasticity, myelination, dendritic process, and oxidative stress in the cortex of aged bovine when compared to the adult samples. In conclusion, our data pave the way for a better understanding of molecular mechanisms underlying brain aging in cattle.

Transcriptional profiling of whole-blood in Mycobacterium avium subsp. paratuberculosis-infected cattle

Project description:To provide novel insights into understanding the host-immune response during the different stages of progression of the disease, we performed gene expression profiling in Mycobacterium avium subsp. paratuberculosis infection in cattle. We analyzed the transcriptional profiles of whole-blood cells of cattle identified and grouped according to the presence of MAP-specific antibodies and the MAP shed by them. The grouping was based on the results obtained by ELISA and PCR analyses as follows: 1) Test1 group: MAP-negative results obtained by ELISA and positive results obtained by PCR; 2) Test2 group: MAP-positive results obtained by ELISA and negative results obtained by PCR; 3) Test3 group: MAP-positive results obtained by ELISA and positive results obtained by PCR; 4) uninfected control: MAP-negative results obtained both by ELISA and PCR analysis.
