We measured the body length of worms reaching the L4 stage after a-SWCNT exposure for 48 hr at 22°C. Compared to the untreated worms, we found that the average length of worms exposed to a-SWCNTs (500 μg mL-1) was significantly shorter than the untreated groups. In addition, the dose of a-SWCNTs also caused retarded growth, reduced lifespan and defective embryogenesis in worms. Genome-wide gene expression analysis using an Affymetrix GeneChip was performed to further investigate the molecular basis of these defects.

Project description:Single-walled carbon nanotubes (SWCNTs) are being explored and used for a wide range of applications in industrial and medical sectors, and the increasing exposure of SWCNTs necessitate the studies of their potential environmental and health effects. Considerable efforts have been made to improve the dispersion of SWCNTs by chemical modifications. The present study was designed to determine if the acid functionalization of SWCNTs enhanced the toxicity and more efforts were made to understand the molecular mechanisms. RAW264.7 cells were exposed to 0-100μg/mL of SWCNTs and AF-SWCNTs for 24 hours and cell proliferation, viability, and gene expression profiles were assessed and compared. Results showed that the magnitude of AF-SWCNT-induced cell proliferation inhibition was higher than that of SWCNTs. 10μg/mL of AF-SWCNTs was determined as non-toxic to cells by viability analysis. AF-SWCNTs could enter and aggregate in cell cytoplasm and nuclear areas. Microarray, real-time reverse-transcriptase polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assays (ELISA) demonstrated that AF-SWCNTs (1, 10, and 50μg/mL) altered gene and protein expression patterns. The most differentially expressed genes were related to pro-inflammatory cytokines and chemokines (such as CCL-3 and CCL-4), apoptosis (e.g., Caspase-1, -4, and Dffa), protective response (such as Hmox-1and GSTM3), and cell surface molecules such as ICAM-1, Itgb2, indicating a possible involvement of SWCNT in inflammation, anti-apoptotic and DNA fragmentation, carcinogenesis and biased T cell polarization. Furthermore, a substantial amount of down-regulated genes were found related to ribosomal assembly and mitochondrial respiration chain, implicating a possible mechanism of SWCNT-induced oxidative stress and gene expression inhibition. Project description:As environmental pollutants and possible carcinogens, carbon nanotubes (CNTs) have recently been found to promote tumorigenesis and tumor metastasis after long-term pulmonary exposure. However, whether CNT-induced carcinogenesis can be inherited and last for generations remains unknown. Here, we establish a post-chronic single-walled carbon nanotubes (SWCNTs) exposed human bronchial epithelium BEAS-2B cell model to investigate SWCNTs-induced carcinogenesis. At a tolerated sublethal dose level, post-chronic SWCNTs exposure significantly increases the migration and colony formation abilities of BEAS-2B cells, leading to cell malignant transformation. Notably, the malignant transformation of BEAS-2B cells is irreversible within 60 days recovery period after SWCNTs exposure, and the malignant transformation activities of cells gradually increase during the recovery period. Mechanism analyses show that post-chronic exposure to SWCNTs causes substantial DNA methylation and transcriptome dysregulation of BEAS-2B cells. Subsequent enrichment and clinical database analyses reveal that differentially expressed/methylated genes of BEAS-2B cells are enriched in cancer-related biological pathways, and several of these genes are validated in lung cancer patients. Project description:To further development of our gene expression approach to assess the effects of manufactured nanomaterials at the molecular level, we have employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential to distinguish characterization of physico-chemical properties of single-wall carbon nanotubes (SWCNTs). Three kinds of single-wall carbon nanotubes (SWCNTs), CNT-1, CNT-2 or CNT-3 induced gene expression in human alveolar epithelial cell lines were measured at 24 and 48 hours exposure to doses of 90.8μg/mL (CNT-1), 109.0μg/mL (CNT-2), and 77.0μg/mL (CNT-2). Three independent experiments were performed at each time (24 or 48 hours) using different untreated control for each experiment (n=4).

Project description:Carbon nanotubes (CNTs) are one of the most attractive nanomaterials that have many potential applications in industrial, medical, and environmental sectors. To investigate the toxic effect of single-walled CNTs (SWCNTs) on the immune system, we examined the influences of SWCNTs (depleted of metal impurities) on viability and global gene expression in murine macrophages. RAW264.7 Cells were exposed to various concentrations of SWCNTs, and cell viability was analyzed at various time points using WST-1 assay. SWCNT cytotoxicity in macrophages was concentration and time dependent, with significant viability loss occurring within 4 hr at 50μg/ml SWCNT exposure. As low as 1μg/ml SWCNTs reduced cell viability at 12 hr, whereas 50μg/ml SWCNT exposure completely abolished cell proliferation. Laser confocal microscopy indicated that SWCNTs could enter and aggregate in cell cytoplasm and nuclear areas. Microarray, real-time reverse-transcriptase polymerase chain reaction and enzyme-linked immunosorbent assay revealed that SWCNT altered gene and protein expression pattern. The most differentially expressed genes were related to proinflammatroy cytokines and chemokine, such as CCL-3 and CCL-4, apoptosis (e.g., Caspase-1, and -3), protective response such as Hmox-1and GSTM3) and cell surface molecules such as ICAM-1, Itgb2, indicating a possible involvement of SWCNT in inflammation and pulmonary granuloma formation, carcinogenesis and Th2 polarization of T cells. The results are a first step in identifying molecular targets for the toxicity of SWCNTs and the elucidating of the molecular mechanisms for the immune modulation ability of SWCNTs. Cells were exposure to 50ug/ml of acid-functionzalized SWCNTs for 24 hours before gene expression profiling, cells without SWCNT exposure were used as control.

Project description:Gene expression profiling of the rat lung following intratracheal instillation with SWCNTs was employed to gain insights into these molecular events. We attempted to characterize time-dependent changes in the gene expression until 754 days after intratracheal instillation with SWCNTs suspensions at 0.2 mg (L-SWCNT) and 0.4 mg (H-SWCNT) injected dose per rat, and to identify the shift from the acute-phase to the chronic-phase phase on the basis of evaluation at the molecular level. Groups of nine-week-old male Wistar rats (n= 6 per group/ time point) were intratracheally instilled with single-wall carbon nanotubes (SWCNTs) suspended in 0.4 ml distilled water including 0.1% Triton X-100 as a single injection 0.1 mg (L-SWCNT) and 0.4 mg (H-SWCNT) SWCNTs/ rat). Control groups received 0.1% Triton X-100 (vehicle control). After intratracheal instillation treatment, rats were housed within polycarbonate cages at a controlled temperature of 22 M-BM-0C with a chow diet ad libitum, and dissected at 3 days, 7 days, 30 days, 90 days, 180 days, 365 days and 754 days post-instillation. Right lungs of anesthetized rats were perfused with physiological saline, excised, and used for DNA microarray analysis.
