Project description:Rice tungro bacilliform virus (RTBV) is determinant of the rice tungro disease symptom. We compare the gene responses by RTBV infection between RTBV susceptive (TN1) and resistant cultivar (TW16). TN1 infected with RTBV shows the mild stunting and leaf yellowing, although TW16 shows only mild stunting at only early infection stage, and recovers the symptoms. Project description:Rice plants cultivated on mountain areas are frequently exposed to acid fog and natural fog events. In this report, we analyzed the expression profiles of the rice plant with acid fog (SiAF) or neutral fog (SiNF) treatment for 1 and 7 days. Microarray results suggested that ROS generation was induced by not only SiAF, but also SiNF treatment, and it occurred in apoplast, not in organelles. Genes for defense- and stress-responses was also induced by both SiAF and SiNF treatments. The induction occurred in plants treated with SiAF for both 1 day and 7 days, and it was also detected in plants treated with SiAF for 7 days. These results suggest that both SiAF and SiNF treatments are abiotic stresses accompanying ROS generation in apoplast. Comparison between continuous fog treated shoots and control shoots. Biological replicates: 3 simulated acid fog (pH 3.0) treated (SiAF) for 1 day; 3 simulated acid fog (pH 3.0) treated (SiAF) for 7 days; 3 simulated neutral fog (SiNF) treated for 1 day; 3 simulated neutral fog (SiNF) treated for 7 days; 3 controls for 1 day; 3 controls for 7 days. Project description:To investigate plant-fungus interactions in early stage of infection, we analyzed response of rice against Magnaporthe grisea infection deficient mutants. In M. grisea, Mgb1 and Mst12 are essential for development of infection structures. Deletion of MGB1 results in defect in appresorium formation, and MST12, in penetration peg development. Analysis of gene expression profiles in rice by microarray revealed the mutant-specific and R gene dependent gene expression, strongly suggesting that gene-for-gene interaction commences before the penetration into rice cell. Experiment Overall Design: Agilent rice oligo microarray was used to investigate the gene expression profiling in rice plants infected with Guy11, Mgb1 and Mst12. Total RNAs were isolated from fourth leaves at 24 and 48 hpi. Agilent rice DNA chips were used according to the manufactureâs protocol. After hybridization, arrays were scanned using an Agilent Microarray Scanner, and Feature Extraction version 9.1 was used for image analysis and data extraction. Signal intensities derived from inoculated leaved were compared with that of mock-inoculated leaves. In each treatment, the experiment was performed independently three times.

Project description:To dissect the molecular mechanisms underlying drought tolerance (DT) in rice, transcriptome differences of a DT introgression line H471, the DT donor P28 and the drought sensitive recurrent parent HHZ under drought stress were investigated using deep transcriptome sequencing. Results revealed a differential constitutive gene expression prior to stress and distinct global transcriptome reprogramming among three genotypes under time-series drought stress, consistent with their differential genotypes and DT phenotypes. DT introgression line H471, the DT donor P28 and the drought sensitive recurrent parent HHZ under drought stress were investigated using deep transcriptome sequencing.The drought stress treatment was started by withholding water at the tillering stage. The days were counted after the AWC in the soil reached 20% to allow drought measurements at precisely determined intervals, and the soil water content reached 15%, 10% and 7.5% after 1d, 3d and 4d drought treatment, respectively.Three top leaves for each sample were harvested for each genotype under 1d and 3d drought stress and control conditions. All samples were immediately frozen in liquid nitrogen and stored at -80C and then for transcriptome sequencing.

Project description:The ‘O3-responsive transcriptome’ behavior in the panicles and grains of rice plant was studied individually through high-throughput oligo-DNA microarray technique. Obtained results showed that O3 differentially regulated 620 and 130 genes in the panicles and grains separately, by at least two-fold changes. However, only five genes were found to be common in both the tissues, suggesting towards the tissue specific O3-sensitivity in rice plants. Among the O3-responsive genes, 176 and 444 genes were up- and down-regulated in rice panicle; whereas, 24 and 106 genes in rice grain, respectively. Further mapping onto various regulatory events revealed that, the majority of differentially expressed genes were mainly involved in signaling, hormonal, cell wall, transcription, proteolysis, and defense events. Many previously unknown O3-responsive novel genes were identified, including the brassinosteroid insensitive-1 receptor kinase, wall-associated kinase like receptor, calcium-dependent protein kinases, phosphatidylinositol kinases, G-protein components, ethylene insensitive-3, cellulose synthases, pectatelyase, etc. Inventory of 745 O3-responsive genes and their mapping will surely expand our knowledge on novel regulatory processes in both panicles and grains of rice; and, serve as a resource towards the designing of rice crops for future high-O3 world. Comparison between healthy rice plant panicles and ozone treated plant panicles (for 8 h) and seed (grain) of healthy rice plants and of rice plants grown under ozone for their lifeftime was performed. Three biological replicates (panicle or seed; pooled) were used, and dye-swaped.
