Project description:Transcriptional profiling of genes recoverd by STOP2-complementation in the stop1-mutant (STOP2 expression is repressed in the stop1-mutant). The roots were exposed to Al and low pH stressed conditions, then genes recovered expression were analyzed. Goal is identifying the genes can be actevated the expression by STOP2. Two-condition experiment, STOP2-complemented lines (stop1-mutant backgoround) v.s. stop1-mutant. Biological replicates: 3 of low pH treatments replicates, 3 Al treatment replicates.

Project description:Primary alveolar type II cells were either treated with vehicle or NaHS for 6 h and 24 h (RS-266 samples). Primary alveolar type II cells treated with GYY4137 at 100 M-BM-5M concentration were exposed to either 21% O2 or 85% O2 for 24 h (RS-284 samples). Overall no changes in gene expressionwere found in the corrected p values of both microarray studies. Four vehicle samples at 6 h of exposure, four NaHS treated samples at 6 h of exposure, four vehicle samples at 24 h of exposure and four NaHS treated samples at 24 h of exposure were used for the RS-266 microarray study. four GYY4137 100 M-BM-5M samples in 21% O2 after 24 h of exposure were compared to four GYY4137 100 M-BM-5M samples in 85% O2 after 24 h of exposure and used for the RS-284 microarray study.

A set of 72 differentially expressed miRNAs provide a molecular signature for pancreatic cancer

Project description:MicroRNAs (miRNAs) are short single-stranded RNA molecules that have a critical role in the regulation of gene expression. Alterations in miRNA expression levels have been observed in multiple tumor types and there is clear evidence on their active involvement in cancer development. Here, a comprehensive miRNA expression profiling in 16 pancreatic cancer cell lines and four normal pancreatic samples provided a specific molecular signature for pancreatic cancer and enabled us to identify 72 differentially expressed miRNAs with approximately half of them being up- and half downregulated in cancer cells as compared to normal samples. Of these, miR-31 was selected for further functional analyses based on its interesting M-bM-^@M-^\on-offM-bM-^@M-^] type expression profile, i.e. very low or even absent expression in normal pancreas and in six of the pancreatic cancer samples but extremely high expression in the remaining ten cell lines. Quite unexpectedly, both the inhibition of miR-31 in AsPC-1 and HPAF-II pancreatic cancer cells with high endogenous expression and forced expression of miR-31 in MIA PaCa-2 with low endogenous levels led to reduced cell proliferation, migration and invasion. More importantly, in AsPC-1 cells further enhancement of miR-31 also resulted in reduced cell migration and invasion, implicating that the level of miR-31 is critical for these phenotypes. We also identified novel miR-31 target genes, APBB2 and RSBN1, that might contribute to cancer pathogenesis. This study highlights a specific miRNA expression pattern in pancreatic cancer and reveals that manipulation of miR-31 expression leads to reduced cell migration and invasion in pancreatic cancer. 16 pancreatic cancer cell lines and 4 normal pancreatic RNA samples were hybridized on Agilent vs1 miRNA microarrays. Pool of four normal samples was hybridized on each slide to allow normalization between slides.

Molecular crosstalk between hepatocytes and hepatic stellate cells.

Project description:Aim of the study was to characterize at a molecular level (changes in transcriptomes) the crosstalk between tumor hepatocytes and activated hepatic stellate cells (HSC) in liver cancer. This was adressed by using a coculture model system of HepaRG cell line (tumor hepatocytes, human), and LX2 cell line (HSC, human). By using genome-wide expression profiling, we demonstrated that hepatocyte-HSC crosstalk is bidirectional and results in the deregulation of functionally relevant gene networks. HepaRG and LX2 cells were cultured alone in serum- and DMSO-free William's E medium or together using 1 M-BM-5m pore size transwell inserts which allow diffusion of media components but prevent cell migration (BD Biosciences, San Jose, CA). Triplicate experiments were performed: HepaRG (culture versus coculture), LX2 (culture versus coculture).
