Project description:To analyze gene expression we isolated total mRNA from the core, periinfarct and contralateral cortex of 60 sprague-dawley rats after 24 hours or 3 days of permanent middle cerebral artery occlusion (pMCAo). Also ipsilateral and contralateral cortex was obtained from sham-operated and healthy animals. A total of 44 microarrays were performed with RNA pooled from 3 independent animals. For each experimental condition the total n number is of 12. Project description:Transcriptome analysis of RNAs extracted from early passage and late passage (immortalized) mouse embryonic fibroblasts (MEFs) We compared the gene expression profiles of early (p.6-7) and immortalized (p.28-33) Smurf2KO cells to those of their wild-type counter parts, and selected the genes that were differentially expressed at a cut-off of 1.54 folds in both B6 and BL backgrounds. The results showed a progressive increase in differential gene activities with the number of cell passages. We detected 101 differentially expressed genes in the early passage and 668 in the immortalized Smurf2KO cells, with 49 in common between these two categories. These data pointed to a global deregulation of gene expression as the cells became immortalized in the absence of Smurf2. We analyzed MEFs from wild type and Smurf2-/- embryos from both C57B6/L (B6) and mixed 129SVJ x NIH Black Swiss (BL) backgrounds using the Affymetrix mouse Exon 1.0 ST platform. No techinical replicates were performed.

Project description:The transcription factor β-catenin has been shown to be active in different types of dendritic cells (DCs) with ability to induce tolerogenic or anti-inflammatory features. Monocyte-derived dendritic cells (moDCs) have been widely used in dendritic cell-based cancer therapy, but so far with limited clinical efficacy. It is possible that aberrant differentiation or induction of dual pro- and anti-inflammatory features may decrease the moDCs efficiency to stage the immune attack on cancer cells. Here we show, using moDCs derived from healthy buffycoats, that β-catenin is detectable in both immature and lipopolysaccharide (LPS)-matured DCs.

Project description:Background: BMPER, an orthologue of Drosophila melanogaster crossveinless-2, is a secreted factor that regulates BMP activity in endothelial cell precursors and during early cardiomyocyte differentiation. Although previously described in the heart, the role of Bmper in cardiac development and function remained unknown. Methods: BMPER deficient hearts were phenotyped histologically and functionally using echocardiography and Doppler analysis. Since BMPER -/- mice die perinatally, BMPER +/- mice were then challenged to pressure overload induced cardiac hypertrophy and hind limb ischemia to determine changes in angiogensis and regulation of cardiomyocyte size. Results: We identified for the first time the cardiac phenotype associated with BMPER haploinsufficiency. BMPER mRNA and protein are present in the heart during cardiac development through at least E14.5 but is lost by E18.5. BMPER +/- ventricles are thinner and less compact than sibling wild-type hearts. In the adult, BMPER +/- hearts present with decreased anterior and posterior wall thickness, decreased cardiomyocyte size, and an increase in cardiac vessel density. Despite these changes, BMPER +/- mice respond to pressure overload-induced cardiac hypertrophy challenge largely to the same extent as wild-type mice. Conclusion: BMPER appears to play a role in regulating both vessel density and cardiac development in vivo; however, BMPER haploinsufficiency does not result in marked effects on cardiac function or adaptation to pressure overload hypertrophy. Unpaired, two-condition experiment, wild-type vs BMPER+/- adult hearts. Biological replicates: 4 per condition.

Project description:The androgen receptor is a steroid receptor belonging to the superfamily of hormone-activated transcriptional factors, displaying distinct expression profiles in Sertoli cells during testis development, tightly correlated to the stages of spermatogenesis.The aim of the project was to better understand the AR signaling pathways, and identify androgen regulated genes in a mature Sertoli cell line (ST38c). In order to identify androgen regulated candidate genes we compared the gene expression of mature Sertoli cells (ST38c) in the absence (condition B) and in the presence of a androgenic ligand, dihydrotestosterone (condition ST). In order to understand the AMH gene repression in mature Sertoli cells, which occurs at puberty, soon after the augmentation of the AR expression, we also compared the gene profile of an immature, not expressing the androgen receptor, prepubertal Sertoli cell line (SMAT1-condition A) with the mature Sertoli cell line (ST38c- condition C), expressing the androgen receptor. The Sertoli cell line SMAT1 has been obtaiend from a 6-day old transgenic male mouse (using the targeted oncogenesis SV40). This cell line has been kindly provided to us by Dr Jean Yves Picard (UMR 782), Universite Paris Sud. (Dutertre M, et al., Mol Cell Endocrinol. 1997 Dec 31;136(1):57-65. PMID 9510068).The Sertoli cell line ST38c was obtained in the INSERM U-693 from 8-wk-old male murine testis; from a transgenic mouse carrying the construct in which the SV40 large T Antigen (TAg) was placed under the control of the human vimentin promoter. The development and description of this new cell line have not yet been published.
