Transcription profiling of Drosophila w1118 flies treated with different doses of radiation at different time points

ABSTRACT: To determine what genes are affect to the reduction in mutation frequency by 500 µGy gamma irradiation of Drosophila melanogaster. The strain is w[1118] (a loss of function mutant of compound eye pigment gene white with Canton-S background). Experiment Overall Design: 15 samples are analyzed. Transcription profiling of Arabidopsis FNR1 mutants from different ecotypes

Project description:The mechanism by which plants regulate channeling of photosynthetically derived electrons into different areas of chloroplast metabolism remains obscure. In higher plants, these electrons are made accessible to stromal enzymes, or cyclic electron flow, as reduced ferredoxin (Fd), or NADPH. We investigated how the specific loss of an Arabidopsis (Arabidopsis thaliana) ferredoxin:NADPH reductase (FNR) isoprotein affected channeling of photosynthetic electrons into NADPH and Fd dependent metabolism. Affymetrix gene chip comparison of transcript changes in these mutants against two different ecotype backgrounds indicates that stress signaling is perturbed, and experiments revealed that the mutants are more susceptible to oxidative stress. Experiment Overall Design: We isolated arabidopsis mutants of the FNR1 gene in different ecotypes, extracted RNA from these and respective wild type plants and hybridised it to Affymetrix microarrays

Transcription profiling of rat neuronal progenitor cells after corticosterone treatment

Project description:Exposure to excessive glucocorticoids, which are major mediators of the stress reaction, is detrimental to brain development related to an increased risk of psychiatric disorders. However, the molecular mechanism underlying the effect of excessive glucocorticoids on the developing brain remains largely unclear. To clarify the mechanism, we examined the effects of glucocorticoids on cultured neuronal progenitor cells (NPCs) from cerebral cortices from rat embryo. We performed comparative analysis using gene expression profiling of corticosterone- or vehicle-treated NPCs. Results provide insight into the effects of glucocorticoids in regulating genetic programs important for controlling NPCsâ properties. Experiment Overall Design: Three couples of RNA samples from corticosterone- or vehicle-treated neuronal progenitor cells were analyzed.

Transcription profiling of human 293 cells

Project description:A long form (tRNase ZL) of tRNA 3' processing endoribonuclease (tRNase Z, or 3' tRNase) can cleave any target RNA at any desired site under the direction of artificial small guide RNA (sgRNA). We discovered in human kidney 293 cell extracts various new small noncoding RNAs (ncRNAs) including 5'-half-tRNAs and 28S rRNA fragments, co-immunoprecipitated with tRNase ZL, and demonstrated that two of these ncRNAs work as sgRNAs for tRNase ZL in vivo as well as in vitro. In order to find genuine mRNA targets of tRNase ZL guided by ncRNAs, we performed DNA microarray analysis for mRNAs from the 293 cells transfected with the tRNase ZL expression plasmid, and found that PPM1F and DYNC1H1 mRNAs are its genuine targets. Effect of mifepristone on gene expression in Drosophila

Project description:Illumina sequencing was used to assay the effect of mifepristone treatment on gene expression in adult Drosophila, including males, virgin females and mated females. Males of strain w[1118]; p53B[6] were crossed to virgins of w[1118]; rtTA(3)E2 and progeny males and virgins were collected over 48 hours. One half of the virgins were mated to w[1118] males at ratio of 1:1 virgins to males for 4 days. Mated females were then separated from the w[1118] males. The mated females, males and virgins females were then maintained at approximately 20 flies per vial, on food with and without supplementation with 160ug/ml mifepristone for 12 days. Total fly RNA was isolated from 20 animals per sample. Three replicate samples were generated for each type of flies: males, mated females and virgin females.
