Transcription profiling of rat PC12 cells treated with cholecystokinin for 2, 4, 8, and 16 hours


ABSTRACT: 1 x 106 PC12-CCK-2R cells/100 mm or 1 x 106 PC12 cells/100 mm were seeded in DMEM containing 0.5% serum for 2 days before cells were untreated or treated with 200 nM CCK-8S for 2, 4, 8, and 16 hours. Total cellular RNA was isolated. Three independent series containing RNA from four 100 mm petri dishes were pooled to minimize differences in cell cultures and treatments.

Cholecystokinin (CCK) is abundantly expressed in the CNS, in which it regulates feeding behavior and long-term memory. Moreover, CCK has been implicated in mental disorders, such as anxiety and schizophrenia. Despite its manifest physiological and pathophysiological role, the molecular targets of neuronal CCK are incompletely understood.
