Transcription profiling by array of muscle biopsies from patients with dermatomyositis or with polymyositis

ABSTRACT: Muscle biopsies were taken from 6 patients with dermatomyositis, 4 with polymyositis and 5 not myopathic subjects as controls. The genome-wide expression patterns were compared using Affymetrix HG-U133A chips. Experiment Overall Design: Gene expression profiles were generated for 15 individuals.

Project description:Differences in the expression profile of hepatic and pancreatic stellate cells are investigated. Aim is to identify organ and disease specific transcriptome signatures of stellate cells, comparing hepatic and pancreatic stellate cells obtained from tissues of chronic inflammation, and primary or metastatic cancers of the pancreas. Tissues of chronic pancreatitis (n=6), pancreatic ductal adenocarcinoma (n=5), liver cirrhosis (n=5) and liver metastasis of pancreatic ductal adenocarcinoma (n=6) were collected and stellate cells were isolated by the outgrowth method. Using cDNA microarrays, differentially expressed genes are identified.

Project description:At eight weeks of age, mice were divided into 10 groups with six mice per group and fed either the control-diet or the same diet supplemented with quercetin, hesperidin, naringenin, anthocyanin, catechin and curcumin at 0.006% (w/w) corresponding to an equivalent intake in humans of 30 mg/day, while proanthocyanin, caffeic acid and ferulic acid were added at 0.06% (w/w) corresponding to an equivalent intake in humans of 300 mg/day. After two weeks of supplementation, mice were sacrificed under pentobarbital anesthesia. The organs were washed with physiological saline solution maintained at 37C by direct injection in the heartM-^Rs left ventricle. Livers were collected, immediately frozen in liquid nitrogen and stored at -80C until the time of analysis.

Project description:The impact on gene expression by the alarmone (p)ppGpp during growth and non-growth was determined by comparing the transcriptome of R. etli CFN42 wild type and rel mutant. This allowed us to better understand the pleiotropic stress phenotype of the rel mutant as well as identifying specific (p)ppGpp-dependent stress regulators. In order to study expression, R. etli CFN42 wild type and rel mutant was grown at 30˚C in acid minimal salts medium supplied with 10 mM NH4Cl and 10 mM succinate while monitoring the optical density (OD) of the culture. Samples were taken at OD600 = 0.3, 0.7 and 6 hours after reaching the maximum OD, representing early/late exponential and stationary phase, respectively.
