Heat stress UNR

ABSTRACT: To identify transcripts that are regulated by putative transcriptional co-activators, WT (Col) and KO of heat responsive transcriptional co-activator (Multiprotein Bridging Factor 1c) in Arabidopsis were subjeted to heat stress. These plants were compared to find transcripts that are regulated by the transcriptional co-activator during heat stress.

Over-expression of MBF1c enhances stress tolerance

Project description:Multiprotein bridging factor 1c MBF1c (At3g24500) is a stress-response transcription co-activator. To test the function of MBF1c, we over-expressed it in transgenic Arabidopsis plants using the 35S-CaMV promoter. T4 seeds form 3 independent lines were tested for their tolerance to biotic and abiotic stress conditions. Constitutive expression of MBF1c in Arabidopsis enhanced the tolerance of transgenic plants to bacterial infection, salinity, heat and osmotic stress. Moreover, the enhanced tolerance of transgenic plants to osmotic and heat stress was maintained even when these two stresses were combined. The expression of MBF1c in transgenic plants augmented the accumulation of a number of sugars and defense transcrtipts in response to heat stress. Transcriptome profiling and inhibitor studies suggest that MBF1c expression enhances the tolerance of transgenic plants to heat and osmotic stress by partially activating, or perturbing, the ethylene-response signal transduction pathway. Identification of Drosophila MBF1 regulated genes in embryos

Project description:Multiprotein bridging factor 1(MBF1), a transcriptional co-activator that can regulate transcription by forming a bridge between the TATA-box-binding protein and gene-specific transcription factors. MBF1 is required for organ development and environmental stress responses. However, the targets of MBF1 is unknown. To identify the MBF1 regulated genes, we compared expression profiles of mRNA purified from wild-type and mbf1 mutant embryos. yw and mbf1 mutant embryos were selected at later stage of development for RNA extraction and hybridization on Affymetrix microarrays.

The interactome of multiprotein bridging factor 1 in Beauveria bassiana

Project description:Beauveria basiana is a well-known filamentous entomopathogenic fungus. Oxidation tolerance is an important determinant to fungal pathogenicity and biocontrol potential. B. bassiana transcriptional co-activator multiprotein bridging factor 1 (BbMBF1) contributes to fungal resistance to the oxidative stress. The interactome of BbMBF1 was revealed by the qualitative proteomic analysis integrated with the immuno-precipitation. This study is sought to unveil the comprehensive protein interaction of BbMBF1 and isolate the BbMBF1-mediated transcription factor under oxidative stress.

Mechanistically, the mTORC1-activated SRPK2 phosphorylates a splicing factor SRSF1, enhancing its binding to FAM120A. FAM120A directly interacts with a lipogenic transcription factor SREBP1 at active promoters, thereby bridging newly transcribed lipogenic genes to the RNA splicing machinery.

Espression data of control and heat-stressed Arabidopsis WT, hd2c-3, brm-1 and brm-1 hd2c-3 plants

Project description:Studies in yeast and animals have revealed that histone deacetylases (HDACs) often act as components of multiprotein complexes, including chromatin remodeling complexes (CRCs). In our group, we found evidence for the interaction between Arabidopsis HD2C deacetylase and a BRM-containing SWI/SNF chromatin remodeling complex. Moreover, we revealed a novel function of HD2C as a regulator of the heat stress response. HD2C transcript levels were strongly induced in plants subjected to heat treatment and the expression of selected heat-responsive genes was up-regulated in heat-stressed hd2c mutant, suggesting that HD2C acts to down-regulate heat-activated genes. Microarray transcriptome analysis of brm and hd2c mutants identified a subset of commonly regulated heat-responsive genes, and the effect of the double brm hd2c mutation on the expression of these genes was non-additive. Moreover, heat treated hd2c, brm and brm hd2c mutants displayed similar rates of growth retardation. Taken together, our findings suggest that HD2C and BRM act in a common genetic pathway to regulate the Arabidopsis heat stress response. 3-week-old WT (Col-0), hd2c-3, brm-1 and brm-1 hd3c-3 plants were collected before and after 1 hour of heat (38Â°C) treatment. For each variant, three biological replicates with similar expression levels of selected marker genes, were chosen for microarray analysis.

mRNA-seq data of 14 days old Arabidopsis seedlings in Col-0 and At rz-1b At rz-1c double mutant

Project description:Nuclear-localized RNA binding proteins are involved in various aspects of RNA metabolism, which in turn modulates gene expression. However, the functions of nuclear-localized RNA binding proteins in plants are poorly understood. Here we report the functions of two proteins containing RNA recognition motifs, At RZ-1B and At RZ-1C, in Arabidopsis. At RZ-1B and At RZ-1C were localized to nuclear speckles and interacted with a spectrum of serine/arginine-rich (SR) proteins through their C-termini. At RZ-1C preferentially bound to purine-rich RNA sequences in vitro through its N-terminal RNA recognition motif. Disrupting the RNA-binding activity of At RZ-1C with SR proteins through over-expression of the C-terminus of At RZ-1C conferred defective phenotypes similar to those observed in At rz-1b/At rz-1c double mutants, including delayed seed germination, reduced stature, and serrated leaves. Loss of function of At RZ-1B and At RZ-1C was accompanied by defective splicing of many genes and global perturbation of gene expression. In addition, we found that At RZ-1C directly targeted FLC, promoting efficient splicing of FLC introns and likely also repressing FLC transcription. Functional metabolomics as a tool to analyze Mediator function and structure in plants

Project description:Mediator is a multiprotein transcriptional co-regulator complex composed of four modules; Head, Middle, Tail, and Kinase. It conveys signals from promoter-bound transcriptional regulators to RNA polymerase II and thus plays an essential role in eukaryotic gene regulation. We describe subunit localization and activities of Mediator in Arabidopsis through metabolome and transcriptome analyses from a set of Mediator mutants. Functional metabolomic analysis based on the metabolite profiles of Mediator mutants using multivariate statistical analysis and heat-map visualization shows that different subunit mutants display distinct metabolite profiles, which cluster according to the reported localization of the corresponding subunits in yeast. Based on these results, we suggest localization of previously unassigned plant Mediator subunits to specific modules. We also describe novel roles for individual subunits in development, and demonstrate changes in gene expression patterns and specific metabolite levels in med18 and med25, which can explain their phenotypes. We find that med18 displays levels of phytoalexins normally found in wild type plants only after exposure to pathogens. Our results indicate that different Mediator subunits are involved in specific signaling pathways that control developmental processes and tolerance to pathogen infections.
