Transcription profiling of diurnal responses in CRY2-OX and wild type tomato plants

Project description:In order to study possible effects of cryptochrome 2-mediated light signals on the global expression profiles of tomato genes, we performed large scale transcription comparisons in wt and CRY2-OX by using a DNA microarray of more than 90,000 tomato oligo-probes. Tomato plants were grown under a daily light cycle of 16h light/8h darkness (LD) and leaves from both genotypes were sampled at presumptive dawn (Zeitgeber time (ZT) 0), eight hours after dawn (ZT8), at presumptive dusk (ZT16) and four hours after dusk (ZT20).

Transcript profiling of the wild type Arabidopsis grown under different photoperiods

Project description:Arabidopsis rosette leaves were harvested from plants grown under different photoperiods under 100 M-5mol photons m-2 s-1 at 20 M-0C. In the first experiment plants grown under short day conditions 8L/16D (8 h light / 16 h dark) for 4 weeks were compared with plants grown under long day (16L/8D) for 3 weeks. In the second experiment plants grown under 12L/12D for 2 weeks were compared with plants grown first 2 weeks under 12L/12D and then two days under short day (8L/16D) conditions.

Project description:To monitor gene expression changes pre floral transition and during early flower development col-0 and tps1-2 GVG::TPS1 (von Dijken, 2004) plants were grown under SD (8 hr light, 16 hr dark) for 21 days. Plants were then shifted to LD (16 hr light, 8 hr dark) conditions to induce flowering. RNA was isolated from micro-dissected apical tissue harvested 0 and 5 days after the shift to LD and double-stranded cDNA was synthesized. Biotinylated cRNA probes were prepared and hybridized to the Affymetrix ATH1 array in duplicate (biological replicates).
