Investigating the effects of Ceylon cinnamon water extract and metformin on HepG2 cells for type 2 diabetes therapy

ABSTRACT: performing high-throughput RNA sequencing (RNA-Seq) transcriptomic analysis of hepatic cells after metformin or CWE treatment to identify changes in diabetes-related gene expression. Outcomes of this project provide evidence for the effectiveness of Ceylon cinnamon water extract compared to the current standard therapy for type 2 diabetes, metformin.

Project description:Bone metastases is a common severe complication for breast cancer. We previously showed that conditioned medium (CM) from osteocytes stimulated with oscillatory fluid flow, mimicking bone mechanical loading during routine physical activities, reduced breast cancer cell extravasation across endothelial monolayers. Endothelial cells are situated at an ideal location to mediate signals between osteocytes in the bone matrix and metastasizing cancer cells in the blood vessels. Therefore, we used RNA sequencing to show that CM from endothelial cells conditioned in CM from flow-stimulated osteocytes significantly altered gene expression in bone-metastatic breast cancer cells. This This provides insights into the capability of bone-loading activity in preventing bone metastases.

Project description:To examine the effect of metformin on lung cancer biology, human lung H226 and H1299 squamous cell carcinoma cell-lines were grown in RPMI-1640 medium with 10% v/v fetal bovine serum and with or without 15 uM metformin hydrochloride for 7-8 days. Medium (with any metformin) was replaced every two days. Paired cultures with and without metformin were grown and maintained in parallel. Three separate paired cultures, all seeded with same stock of frozen cells, were grown.
