Project description:Detoxification of chlorophyll during leaf senescence in higher plants is a complex and tightly regulated process. It aims at opening the chlorophyll porphyrin ring to produce non-photoreactive degradation products, termed phyllobilins, which are stored in the vacuole. Here we are describing the influence on the transcriptome of dark-induced senescing leaf of the removal of three key enzymes of the PAO/phyllobilin pathway (namely: PAO, PPH and NYE/SGR).

Expression data from Arabidopsis thaliana senescent leaves treated with cytokinins

Project description:Cytokinins are plant hormones with biological functions ranging from coordination of plant growth and development to the regulation of senescence. A series of 2-chloro-N6-(halogenobenzylamino)purine ribosides was prepared and tested for cytokinin activity in selected bioassays. Several compounds showed significant activity, especially in delaying senescence in detached wheat leaves. We used microarrays to gather information about the reprogramming of gene transcription when senescent Arabidopsis leaves were treated with selected C2-substituted aromatic cytokinin ribosides that showed high activity in the senescence bioassay. Arabidopsis senescent leaves were treated with cytokinins and subsequently used for RNA extraction and hybridization on Affymetrix microarrays. 21-days old Arabidopsis leaves were treated with the appropriate cytokinin or left untreated (DMSO only).

For this, petioles of detached leaves were put in a solution containing either sucrose or sorbitol (control). Sugars were taken up into the leaf via the respiration stream for 13 hours. After that, leaves were frozen in liquid nitrogen and RNA was extracted for analysis. Project description:Virus infection and over expression of protein in cytosol induce a subset of HSP70s. We named this response the Cytosolic Protein Response (CPR) and have been investigating it in the context of a parallel mechanism in the soluble cytosol with the UPR, and as a subcomponent of the larger HS response. This experiment was carried out to study the transcriptional aspect of CPR. In this analysis, we have triggered CPR by infiltrating proline analogue, L-azetidine-2-carboxylic acid (AZC) into Arabidopsis mature leaves. Since AZC trigger unfolded protein response(UPR) in ER as well as CPR, we have included tunicamycin treatment, which is a specific inducer of UPR to subtract the effect of UPR from the AZC response. Heat shocked samples were included to identify CPR as a subcomponent of larger HS response. We used microarray data to identify the genes upregurated by CPR. These genes were commonly upregulated by AZC and HS but not by tunicamycin treatment. Experiment Overall Design: Arabidopsis mature leaves were infiltrated with AZC or L-Proline (control of AZC) and tunicamycin or DMF(solvent control of tunicamycin). For the heat shock treatment, six mature leaves were detached from plant and incubated at 37 Â°C or 20Â°C (as a control) for a period of 1h. The experiment was repeated three times for AZC and HS treatment (3 biological replication). Tunicamycin experiment was repeated five times due to large valiation in the responses (5 biological replication).

Comparative expression analysis of the Arabidopsis strigolactone (SL)-response mutant, max2-3, and wild type (WT) plants (Col-0) under dehydration conditions.

Project description:To understand the role of SL-signaling components in water stress response, we have carried out comparative expression analysis of the SL-response max2-3 mutant and WT plants under dehydration and well-watered (control) conditions. AligentM-bM-^@M-^Ys whole Arabidopsis Gene Expression Microarray (G2519F-021169, V4, 4x44K) was used. Two-week-old WT and max2-3 mutant plants were transferred from GM plates to soil and grown for 10 additional day. The aerial parts of 24-d-old plants were detached and exposed to dehydration on KimTowel papers for 0 (well-watered, control), 2 and 4 h. All rosette leaves of independent 24-d-old plants were collected. Total RNA was prepared and used for the microarray hybridization. Three independent biological replicates were used for each plant sample.

Project description:Disposing efficiently and safely chlorophyll derivatives during senescence requires a coordinated pathway that is well conserved throughout green plants. The PAO/phyllobilin pathway catalyzes the degradation of the chlorophyll during senescence and allows detoxification of the pigment and its subsequent export from the chloroplast. Although most of the chloroplastic reactions involved in chlorophyll degradation are well understood, the diversity of enzymes responsible for downstream modifications of non-phototoxic phyllobilins remains to be explored. More than 40 phyllobilins have been described to date, but only three enzymes catalyzing side-chain reactions have been identified in Arabidopsis thaliana, namely, TIC55, CYP89A9, and MES16. Here, by generating a triple mutant, we evaluate the extent to which these enzymes are influencing the rate and amplitude of chlorophyll degradation at the metabolite as well as its regulation at the transcriptome level. Our data show that major side-chain modifications of phyllobilins do not influence significantly chlorophyll degradation or leaf senescence, letting the physiological relevance of their striking diversity an open question.

Transcription profiling of Arabidopsis Col-0 and NDED3 knock out plants subjected to drought stress for different lengths of time

Project description:Arabidopsis thaliana ecotype Col-0 and NCED3 knockout mutant (nc3-2) were used for transcriptome analyses. We were provided the mutant line carrying a T-DNA in the AtNCED3 by GABI Kat Team in Max-Planck-Institute. We named it nc3-2. Drought stress treatment for the analysis was performed as follows. Plants were grown in soil under illumination (16 h of light and 8 h of dark cycle/ 40-60 umol photons/s/m2) at 22 Â°C for 4 weeks. The aerial parts were detached and exposed to drought stress on the paper for various hours at 22 Â°C under 50 % humidity.

Project description:To understand the role of the Arabidopsis type-B Response Regulators ARR1, ARR10 and ARR12 in water stress response, we have carried out comparative expression analysis of the arr1,10,12 mutant and WT plants under dehydration and well-watered (control) conditions. Agilentâs whole Arabidopsis Gene Expression Microarray (G2519F-021169, V4, 4x44K) was used. Two-week-old WT and arr1,10,12 mutant plants were transferred from GM plates to soil and grown for 10 additional day. Aerial portions of 24-d-old plants were detached and exposed to dehydration on KimTowel papers for 0 (well-watered, control), 2 and 4 h. Rosette leaves collected in 3 biological repeats from arr1,10,12 and WT plants treated by dehydration for 0, 2 and 4 h were used for microarray and expression analyses. Total RNA was prepared and used for the microarray hybridization. Three independent biological replicates were used for each plant sample.
