Gene expression profile in breast cancer cells with opposing responses to TGF-beta

ABSTRACT: We aim to test whether our panel of breast cancer cell lines belong to the claudinlow subtype of breast cancer using the classifier published by Prat et al. Breast Cancer Research 12, 2010. For the analysis, we used gene expression data of our cell lines obtained with Illumina bead arrays grown in 2D as attahed and in 3D as mammosphere cultures.

The role of transforming growth factor-beta (TGFβ) in the progression of different molecular subtypes of breast cancer has not been clarified. Here we show that TGFβ increases breast tumour-initiating cell (BTIC) numbers but only in claudin(low) breast cancer cell lines by orchestrating a specific gene signature enriched in stem cell processes that predicts worse clinical outcome in breast cancer patients. NEDD9, a member of the Cas family of integrin scaffold proteins, is necessary to mediate t  ...[more]
