RNA-Seq profile of PSGL-1loCD4+ T cells from chronic GVHD mice

Project description:Our studies have shown that CD44hiCD62LloPSGL-1loCD4+ T (PSGL-1loCD4+ T) cells play an important role in chronic GVHD pathogenesis (R. Deng et al: Nature Communications 2017). Thus, PSGL-1loCD4+ T cells were sorted from spleen of chronic GVHD BALB/c recipient mice transplanted with spleen cells and T cell-depleted bone marrow (TCD-BM) cells from C57BL/6 donor mice. The control no GVHD BALB/c recipient mice were transplanted with C57BL/6 donor TCD-BM only. 21 days after transplantation, PSGL-1loCD4+ T cells from spleen of chronic GVHD and control mice were sorted and subjected to RNA-seq analysis.

Profiling circulating microRNA expression in a mouse model of nerve allotransplantation (part 2)

Project description:Background. The lack of noninvasive biomarkers of rejection remains a challenge in the accurate monitoring of deeply buried nerve allografts and precludes optimization of therapeutic intervention. This study aimed to establish the expression profile of circulating microRNAs (miRNAs) during nerve allotransplantation with or without immunosuppression. Methods. Balb/c mice were randomized into 3 experimental groups, that is, (1) untreated isograft (Balb/c → Balb/c), (2) untreated allograft (C57BL/6 → Balb/c), and (3) allograft (C57BL/6 → Balb/c) with FK506 immunosuppression. A 1-cm Balb/c or C57BL/6 donor sciatic nerve graft was transplanted into sciatic nerve gaps created in recipient mice. At 1, 3, 7, 10, and 14 d after nerve transplantation, nerve grafts, whole blood, and sera were obtained for miRNA expression analysis with an miRNA array and subsequent validation with quantitative PCR.

Transcriptomic profile of murine abdominal aorta allograft undergoing chronic rejection

Project description:Graft vascular disease (GVD) is a major problem limiting the long-term survival of grafts. To determine the underlying mechanisms of GVD during chronic phase, we performed murine abdominal aorta transplantation. Donor aortas from C57Bl/6 (H-2b) mice were transplanted into C57Bl/6 (H-2b) mice, and we call this group isograft (ISO). Donor aortas from BALB/c (H-2d) mice were transplanted into C57Bl/6 (H-2b) mice, and we call this group allograft (ALLO). In brief, donor abdominal aortas were isolated and the branch vessels were ligated, while the recipient vessels were cut from the midsection using microscopic scissors. The recipient and donor vessels were anastomosed with a cuff suture. 4 weeks after aorta transplantation, the mice were sacrificed for transcriptomic sequencing and analyzing of the aortic grafts.

Single cell RNA-sequencing of pleural cavity macrophages naïve and nematode-infected C57BL/6 and BALB/c mice

Project description:The recent revolution in tissue-resident macrophage biology has resulted largely from murine studies performed in the C57BL/6 strain. Here, we provide a comprehensive analysis of immune cells in the pleural cavity using both C57BL/6 and BALB/c mice. Unlike C57BL/6 mice, naïve tissue-resident Large Cavity Macrophages (LCM) of BALB/c mice failed to fully implement the tissue residency programme. Following infection with a pleural-dwelling nematode these pre-existing differences were accentuated with LCM expansion occurring in C57BL/6 but not BALB/c mice. While infection drove monocyte recruitment in both strains, only in C57BL/6 mice were monocytes able to integrate into the resident pool. Monocyte to macrophage conversion required both T cells and IL-4Rα signalling. Host genetics are therefore a key influence on tissue resident macrophage biology, and during nematode infection Th2 cells control the differentiation pathway of tissue resident macrophages.

Project description:During immune-mediated severe aplastic anemia (SAA) monocytes (CD11b+ Ly6C+ Ly6G-) signficantly increase by both frequency and number within the bone marrow. We isolated BM monocytes from F1 hybrid mice (C57BL/6;Balb/c) induced with SAA via the splenocyte-transfer model. We utilized single-cell RNA sequencing (scRNA-seq) to analyze the heterogenity of BM monocytes during SAA.
