Project description:Testis RNA from C57/BL6J mice at various ages post partum was compared to a common control consisting of adult (8 week) testis RNA from the same strain. This generates a time course showing the activation of genes on the array across the first wave of spermatogenesis, which can then be correlated with the appearance of specific germ cell types, allowing assignation of unknown genes to differing cell types and charting the evolution of the developing testis transcriptome. Adult testis RNA from six different genetic models of infertility (XXSxrb, mshi, Bax -/-, bs, Bcl-w -/- and azh) were compared to age- and strain-matched normal control testis RNA. These models possess different cellular complements within the testis, which can be interpreted within the framework established by the first wave analysis and used to refine the assignment of genes to cell type.

Project description:Temporal study of the first wave of spermatogenesis in juvenile mouse testis and analysis of four germ cell deficient mouse models. A series of purified testis cell-types (McCarrey libraries)constituting a focussed gene set was exploited to examine gene expression in prepubertal mouse testis. Testis RNA from C57/BL6J mice at various ages post partum was compared to a common control consisting of adult (8 week) testis RNA from the same strain(also corresponding to the last time-point).This generates a time course showing the activation of genes on the array across the first wave of spermatogenesis, which can then be correlated with the appearance of specific germ cell types, allowing assignation of unknown genes to differing cell types. Transcription profiling of left and right lateral plates of mouse 8.5dpc embryos to identify assymetrically expressed genes

Project description:C3H/HeH females were mated with 101/H males Mating was assessed by the presence of vaginal plugs the next morning. Identification of plug was designated day 0.5. At 8.5 days of development the dam was culled by cervical dislocation, the uterine horns disscted and placed into PBS. The uterus was dissected to release decuduae and these opened according to standard protocols. Morphologically normal looking embryos of between 4 and 6 somites were collected. The left and right lateral plates were dissected away with watchmakers forceps and pools of 4 left and 4 right lateral plates were snap frozen in LN2.

Project description:We have employed whole RNA microarray expression profiling as a discovery platform to identify genes regulated by overexpression of miR-145 in U87 glioma cell. Lentivirus containing miR-145 coding sequence was infected to U87 cell to make U87 overexpressing miR-145. We did genome microarray between U87 and U87 overexpressing miR-145. Total RNAs from U87 cell or U87 overexpressing miR-145 were extracted. Whole RNA microarray expression profiling was performed between them.

The response to substitution of soybean with faba bean protein concentrate on hepatic transcriptome of Atlantic salmon (Salmo salar)

Project description:The production of carnivorous fish such as Atlantic salmon (Salmo salar) is dependent on the availability of high quality protein required as a sustainable substitute for the formulation of the feeds. Plants have arguably the greatest potential, however a major limitation is associated with the presence of anti-nutritional factors. Investigating novel raw materials involves understanding the physiological consequences of the substitution. The primary aim of the present study was to assess the metabolic response of salmon to increasing inclusion of air-classified faba bean protein concentrate (BPC) in feeds as a replacement for soy (SPC). Specifically, we focused on the hepatic transcriptome response to dietary BPC inclusion over a range including commercially relevant levels (e.g. 11-22%) to levels giving impaired performance (45 %). The present investigation provided a profile of the salmon hepatic response to BPC indicating that fish tolerated moderate substitution of dietary SPC with BPC with no evident negative effects on the hepatic physiology of the fish. The analysis of extreme levels of substitution provided insights into physiological mechanisms that are significantly altered providing the basis for further investigation and improvement.
