Single-cell RNA sequencing of effector memory CD8+ T cells of nasal-associated lymphoid tissue (NALT), spleen, submandibular & cervical lymph nodes and blood of Epstein Barr Virus (EBV)-infected NSG mice engrafted with a human immune system (HIS mice)


ABSTRACT: To identify the difference in transcriptome of effector memory CD8+ T cells in different tissues of EBV-infected HIS mice, we sorted CD8+ CD62L- CD45RA- cells from NALT, spleen, lymph node and blood via flow cytometry-assisted sorting. We pooled the cells of three individual mice together in equal cell count to gain heterogenous samples. Using the 10X Genomics platform, we performed single-cell RNA sequencing of the cells to identify their transcriptome.
