Effect of cwc21 gene deletion on pre-mRNA processing

ABSTRACT: An experiment to follow the effects of deletion of the gene coding for the cwc21 protein in the budding yeast Saccharomyces cerevisiae. This was performed after shift to 37°C from 30°C at 0, 30 and 60 minutes after shift. The pre-mRNA processing (mainly removal of introns from pre-mRNA (splicing)) was followed by RNA extraction, making cDNA labelled with either Cy3 or Cy5 and hybridizing on a custom microarray designed to assess splicing. The microarray has probes for introns, exons and exon/exon junctions found in both mature mRNA and also predicted to be used less frequently. The mutant strain was compared with an identically treated strain with the CWC21 gene intact.
