Transcriptional profiling of mammary ductal epithelial progenitors

ABSTRACT: Gene expression of ductal (CFCs Sca1+CD49b+) and alveolar (CFCs Sca1-CD49b+) progenitors were compared. Ductal and alveolar progenitors were FACS sorted from mammary glands of 10 week old virgin female C57/Bl6 mice. Biological replicates (3 per each population) were generated and gene-level expression profiles were determined by hybridization to Affymetrix MoExon 1.0 ST arrays.

Project description:The purpose of this microarray experiment was to obtain reference gene expression patterns of a number of epithelial cell populations [mammary stem cells (MASC), luminal progenitors (LP), alveolar luminal stem/progenitor cells (WC virgin-these are mammary epithelial cells genetically marked by Wap-Cre in virgin females), mature luminal cells (ML, mainly represent ductal luminal cells in virgin females), and alveolar luminal cells (WC preg M-bM-^@M-^S these are alveolar cells genetically marked by Wap-Cre during mid-gestation)] present in the mammary gland of wildtype adult mice on a C57BL6 genetic background. For the isolation of RNA from mammary stem cells (MASC, Lin-CD24+CD29hi), luminal progenitors (LP, Lin-CD24hiCD29+CD61+), and mature luminal cells (ML, lin-CD24hiCD29+CD61-), the thoracic and inguinal mammary glands from 3 adult virgin female mice were harvested, minced and digested into a single cell suspension. Form each of these 3 single cell suspensions, the above populations were sorted by FACS. For alveolar luminal stem/progenitor cells and alveolar luminal cells, Lin-YFP+ mammary epithelial cells were isolated from virgin or midgestation mice genetically marked by Wap-Cre;R26Y. R26Y is a conditional YFP reporter that would be turned on upon Cre-mediated recombination.

The ratio of periportal mesenchyme to ductal epithelium acts as a proliferative rheostat in liver regeneration

Project description:Liver epithelial cells –hepatocytes and bile duct cells– intermingle with a microenvironment of endothelial cells, macrophages and fibroblasts to form the functional unit of the tissue. In homeostasis, the liver epithelium self-renews slowly, yet it is capable of remarkable regeneration following damage. We have shown that liver ductal cells can be expanded in vitro under defined medium to form 3D epithelial 'liver organoids' that recapitulate many aspects of the regeneration process in vitro, yet lack stromal cell components. Here, we describe that a subpopulation of portal mesenchymal cells expressing Sca1 and Pdgfra, exerts a dual effect on the proliferative capacity of ductal cells. On one hand, it supports liver organoid formation and expansion in the absence of exogenous growth factors and independently of cell-to-cell contact. However, when Ductal cells and SCA1+ mesenchymal cells establish physical interaction following a microfluidics encapsulation that enables the cells to self-organize into chimeric organoid structures, cell proliferation is abolished in a mesenchymal-dose dependent manner. We found that it is the ratio between mesenchyme and epithelial cells and not their absolute values that determines the net outcome of ductal proliferation both in vitro, and in vivo, during the different phases of liver regeneration. The mechanism by which Sca1+ mesenchymal cells regulate ductal cell proliferation dynamics is mediated, at least in part, by Notch cell-cell contact inhibition. Our findings underscore how the relative ratio and distribution of cell-cell contacts between the epithelium and its mesenchymal microenvironment are key regulatory processes during the dynamic regulation of tissue regeneration.

Gene expression profile of ADM, ductal and acinar cells in the setting of pancreatitis

Project description:In order to compare the gene expression profile of acinar-to-ductal metaplasia (ADM), ductal and acinar cells in the setting of pancreatitis, which were obtained by laser-capture microdissection (LMD) from frozen sections of wild type mice pancreata 2 days after caerulein administration, we performed microarray analysis. ADM has an intermediate property between ductal and acinar cells in the setting of pancreatitis, and Cxcr4 mRNA which is expressed in multipotent pancreatic progenitors, were up-regulated in ADM compared with ductal cells or acinar cells. Notably, in consistent with our immunostaining data, Dclk1 mRNA was highly expressed in ADM cells compared with ductal and acinar cells.

RNA-seq expression profiling of ductal macrophages and antigen presenting cells from virgin mouse mammary glands

Project description:Macrophages are diverse immune cells that reside in all tissues to regulate development, function and homeostasis. In the mammary gland, a highly dynamic organ that undergoes dramatic remodeling throughout life, macrophages have been implicated in development and breast tumorigenesis. Ductal macrophages, as opposed to their stromal counterparts, were found to promote ductal morphogenesis during puberty and efficiently phagocytose alveolar cells post-lactation. Within mammary tumors, only ductal macrophages expanded to form a network throughout the tumor, where they also contact intra-tumoral T cells. Furthermore, ductal macrophages accelerated the growth of tumor organoids. Thus, pro-remodeling mammary ductal macrophages represent the normal tissue counterpart of tumor macrophages in an organ that is highly susceptible to oncogenesis. To gain insight into ductal macrophage function, we sorted all antigen presenting cell populations from mouse mammary glands and undertook RNA-seq expression profiling. This showed that ductal macrophages have a distinct expression profile from those of stromal macrophages or dendritic cells from the mammary gland. Ductal keratin 15+ luminal progenitors in normal breast exhibit a basal-like breast cancer transcriptomic signature

Project description:Normal breast luminal epithelial progenitors have been implicated as cell of origin in basal-like breast cancer, but their anatomical localization remains understudied. Here, we combine micro-collection of uncultured organoids and single cell mRNA sequencing (scRNA-seq) of FACS-sorted luminal epithelial cells with multicolor imaging to profile ducts and terminal ductal lobular units (TDLUs) and compare them with breast cancer subtypes. Unsupervised clustering reveals eleven distinct clusters and a differentiation trajectory starting with keratin 15+ (K15+) progenitors enriched in ducts. Spatial mapping of luminal progenitors is confirmed at the protein level by staining with critical duct markers. Comparison of the gene expression profiles of normal luminal cells with those of breast cancer subtypes reveals a strong correlation between normal breast K15+ ductal progenitors and basal-like breast cancer. We propose that K15+ basal-like breast cancers originate in ductal progenitors, which emphasizes the importance of not only lineages but also cellular position within the ductal-lobular tree.

HSP27 (Heat Shock Protein 27, HSPB1) overexpression effect on bleomycin-induced PF (Pulmonary fibrosis) mice model

Project description:To analyze the role of HSP27 in bleomycin (BLM)-induced pulmonary fibrosis in vivo, we used TG mice that conventionally overexpressed HSP27. Our previously reported that HSP27 plays an important role in pulmonary fibrosis. In mice overexpressing HSPB1, BLM-induced fibrotic regions and collagen deposits were increased compared with those in control BL6 mice. Following intratracheal treatment of BLM, the HSP25 TG mice showed an abundance of mononuclear and neutrophils cells in the alveoli, greater destruction of alveolar septa, intra-alveolar hyaline membrane formation, and a marked increase in collagen deposition compared with control BL6 mice.
