RNA-seq of mesenchymal stromal cells cultured in the presence of breast cancer cell secretomes

ABSTRACT: Mesenchymal stromal cells were cultured in 3D PEG hydrogels for 7 days in the presence of serum-free media or conditioned media from a panel of breast cancer cells (MCF-7, MDA-MB-231, MDA-MB-231 lung-tropic, MDA-MB-231 brain-tropic, MDA-MB-231 bone-tropic). In all cases, the secretomes were collected after cancer cells were in serum-free media for 24h.

Project description:We report the gene expression patterns in MDA-MB-231 (a line selected for low metastatic ability), MDA-MB-231-1833 (its bone-tropic metastatic derivative line), MDA-MB-231p27CK-DD (a phosphomimetic cell line), MDA-MB-231-1833shp27 (p27 knockdown cell line), MDA-MB-231-1833PF1502 (PI3K inhibitor treatment). It shows that the gene expression pattern are regulated in a p27 phosphorylation-dependent manner.

Modulation of the cancer cell transcriptome by culture media formulations and cell density

Project description:We investigated how varying the composition of cell culture formulations and growing cancer cells at different densities might affect tumor cells genotype. Specifically, we compared gene expression profiles generated by human MDA-MB-231 human breast cancer cells cultured in different media (MEM, DMEM, or RPMI 1640) containing different concentrations of fetal bovine serum (FBS) or different sera (equine or bovine) that were grown at different cell densities. We have 6 different group (1. MDA-MB-231; MEM-10% FBS-90% confluence (MEM-1 and MEM-2). 2. MDA-MB-231; DMEM-10% FBS-90% confluence (DMEM-1 and DMEM-2). 3. MDA-MB-231; RPMI1640-10% FBS-90% confluence (RPMI-1 and RPMI-2) 4. MDA-MB-231; MEM-10% Horse serum-90% confluence (Horse-1 and Horse-2). 5. MDA-MB-231; MEM-0.1% FBS-90% confluence (FBS0.1-1 and FBS0.1-2). 6. MDA-MB-231; MEM-10% FBS-50% confluence (Con50-1 and Con50-2). Each group has duplicated. We analyzed; 1. Cell density (90% vs 50% confluence). 2. FBS concentration (10% FBS vs 0.1% FBS). 3. Sera (10% FBS vs 10% Horse serum). 4. Media (MEM vs DMEM vs RPMI1640)

Influence of LRP-1 on MDA-MB-231 secretome

Project description:LRP-1 (low-density lipoprotein receptor-related protein-1) receptor is a multifunctional endocytosis receptor that is part of the LDL receptor family. Due to its capacity to control the pericellular level of various growth factors and proteases, LRP-1 plays a crucial role in controlling the dynamics of the membrane proteome. LRP-1 overexpression in breast cancer, prompted us to take an interest in its involvement in tumor progression. An RNA interference strategy in MDA-MB-231 line was used, based on shRNA stable expression. In addition to integrated experimental strategies (in vitro and in vivo) through combined approaches of biochemistry, molecular biology, cell biology, multimodal preclinical imaging, proteomics allowed us to compare shLRP-1 MDA-MB-231 tumor conditioned media to shCtrl MDA-MB-231 tumor conditioned media in order to identify secreted molecular targets modulate by LRP-1 repression and thus provide a better understanding of its regulatory action within the TNBC microenvironment.

Gene expression in human breast cancer cell lines regulated by CXCR6 gene

Project description:This study aimed to further our understanding of the role that CXCR6 plays in breast cancer progression. Microarrays were searched for some genes that had correlated expression with CXCR6 mRNA. According to fold-change screening between restoring expression of CXCR6 and its respective control cells in MDA-MB-231 and MCF-7 cells, both up-regulated and down-regulated genes were shown. The restoring of CXCR6 expression in MDA-MB-231 and MCF-7 cells were respectively noted as MDA-MB-231-CXCR6 and MCF-7-CXCR6 cells and the respective controls were noted as MDA-MB-231-GFP and MCF-7-GFP cells.
