NuRD-mediated gene repression in mouse embryonic stem cells


ABSTRACT: To obtain a global view of NuRD-mediated transcriptional regulation, gene expression profiles of wild-type and Mbd3-null ES cells were compared by microarray analysis. In the absence of Mbd3, which encodes a core structural component of NuRD, the complex does not form and embryonic development stalls at the implantation stage.

Pluripotent cells possess the ability to differentiate into any cell type. Commitment to differentiate into specific lineages requires strict control of gene expression to coordinate the downregulation of lineage inappropriate genes while enabling the expression of lineage-specific genes.
