Streamlining the interface between electronics and neural systems for bidirectional electrochemical communication.

ABSTRACT: Seamless neural interfaces conjoining neurons and electrochemical devices hold great potential for highly efficient signal transmission across neural systems and the external world. Signal transmission through chemical sensing and stimulation via electrochemistry is remarkable because communication occurs through the same chemical language of neurons. Emerging strategies based on synaptic interfaces, iontronics-based neuromodulation, and improvements in selective neurosensing techniques have been explored to achieve seamless integration and efficient neuro-electronics communication. Synaptic interfaces can directly exchange signals to and from neurons, in a similar manner to that of chemical synapses. Hydrogel-based iontronic chemical delivery devices are operationally compatible with neural systems for improved neuromodulation. In this perspective, we explore developments to improve the interface between neurons and electrodes by targeting neurons or sub-neuronal regions including synapses. Furthermore, recent progress in electrochemical neurosensing and iontronics-based chemical delivery is examined.

Project description:Progress in decoding neural signals has enabled the development of interfaces that translate cortical brain activities into commands for operating robotic arms and other devices. The electrical stimulation of sensory areas provides a means to create artificial sensory information about the state of a device. Taken together, neural activity recording and microstimulation techniques allow us to embed a portion of the central nervous system within a closed-loop system, whose behavior emerges from the combined dynamical properties of its neural and artificial components. In this study we asked if it is possible to concurrently regulate this bidirectional brain-machine interaction so as to shape a desired dynamical behavior of the combined system. To this end, we followed a well-known biological pathway. In vertebrates, the communications between brain and limb mechanics are mediated by the spinal cord, which combines brain instructions with sensory information and organizes coordinated patterns of muscle forces driving the limbs along dynamically stable trajectories. We report the creation and testing of the first neural interface that emulates this sensory-motor interaction. The interface organizes a bidirectional communication between sensory and motor areas of the brain of anaesthetized rats and an external dynamical object with programmable properties. The system includes (a) a motor interface decoding signals from a motor cortical area, and (b) a sensory interface encoding the state of the external object into electrical stimuli to a somatosensory area. The interactions between brain activities and the state of the external object generate a family of trajectories converging upon a selected equilibrium point from arbitrary starting locations. Thus, the bidirectional interface establishes the possibility to specify not only a particular movement trajectory but an entire family of motions, which includes the prescribed reactions to unexpected perturbations.

Project description:Free communication is one of the cornerstones of modern civilisation. While manual keyboards currently allow us to interface with computers and manifest our thoughts, a next frontier is communication without manual input. Brain-computer interface (BCI) spellers often achieve this by decoding patterns of neural activity as users attend to flickering keyboard displays. To date, the highest performing spellers report typing rates of ~10.00 words/minute. While impressive, these rates are typically calculated for experienced users repetitively typing single phrases. It is therefore not clear whether naïve users are able to achieve such high rates with the added cognitive load of genuine free communication, which involves continuously generating and spelling novel words and phrases. In two experiments, we developed an open-source, high-performance, non-invasive BCI speller and examined its feasibility for free communication. The BCI speller required users to focus their visual attention on a flickering keyboard display, thereby producing unique cortical activity patterns for each key, which were decoded using filter-bank canonical correlation analysis. In Experiment 1, we tested whether seventeen naïve users could maintain rapid typing during prompted free word association. We found that information transfer rates were indeed slower during this free communication task than during typing of a cued character sequence. In Experiment 2, we further evaluated the speller's efficacy for free communication by developing a messaging interface, allowing users to engage in free conversation. The results showed that free communication was possible, but that information transfer was reduced by voluntary textual corrections and turn-taking during conversation. We evaluated a number of factors affecting the suitability of BCI spellers for free communication, and make specific recommendations for improving classification accuracy and usability. Overall, we found that developing a BCI speller for free communication requires a focus on usability over reduced character selection time, and as such, future performance appraisals should be based on genuine free communication scenarios.

Project description:Aim: This investigation aims to advance the understanding of neural dynamics that underlies live and natural interactions during spoken dialogue between two individuals. Introduction: The underlying hypothesis is that functional connectivity between canonical speech areas in the human brain will be modulated by social interaction. Methods: Granger causality was applied to compare directional connectivity across Broca's and Wernicke's areas during verbal conditions consisting of interactive and noninteractive communication. Thirty-three pairs of healthy adult participants alternately talked and listened to each other while performing an object naming and description task that was either interactive or not during hyperscanning with functional near-infrared spectroscopy (fNIRS). In the noninteractive condition, the speaker named and described a picture-object without reference to the partner's description. In the interactive condition, the speaker performed the same task but included an interactive response about the preceding comments of the partner. Causality measures of hemodynamic responses from Broca's and Wernicke's areas were compared between real, surrogate, and shuffled trials within dyads. Results: The interactive communication was characterized by bidirectional connectivity between Wernicke's and Broca's areas of the listener's brain. Whereas this connectivity was unidirectional in the speaker's brain. In the case of the noninteractive condition, both speaker's and listener's brains showed unidirectional top-down (Broca's area to Wernicke's area) connectivity. Conclusion: Together, directional connectivity as determined by Granger analysis reveals bidirectional flow of neuronal information during dynamic two-person verbal interaction for processes that are active during listening (reception) and not during talking (production). Findings are consistent with prior contrast findings (general linear model) showing neural modulation of the receptive language system associated with Wernicke's area during a two-person live interaction. Impact statement The neural dynamics that underlies real-life social interactions is an emergent topic of interest. Dynamically coupled cross-brain neural mechanisms between interacting partners during verbal dialogue have been shown within Wernicke's area. However, it is not known how within-brain long-range neural mechanisms operate during these live social functions. Using Granger causality analysis, we show bidirectional neural activity between Broca's and Wernicke's areas during interactive dialogue compared with a noninteractive control task showing only unidirectional activity. Findings are consistent with an Interactive Brain Model where long-range neural mechanisms process interactive processes associated with rapid and spontaneous spoken social cues.
