Forced and spontaneous symmetry breaking in cell polarization.

ABSTRACT: How does breaking the symmetry of an equation alter the symmetry of its solutions? Here, we systematically examine how reducing underlying symmetries from spherical to axisymmetric influences the dynamics of an archetypal model of cell polarization, a key process of biological spatial self-organization. Cell polarization is characterized by nonlinear and non-local dynamics, but we overcome the theory challenges these traits pose by introducing a broadly applicable numerical scheme allowing us to efficiently study continuum models in a wide range of geometries. Guided by numerical results, we discover a dynamical hierarchy of timescales that allows us to reduce relaxation to a purely geometric problem of area-preserving geodesic curvature flow. Through application of variational results, we analytically construct steady states on a number of biologically relevant shapes. In doing so, we reveal non-trivial solutions for symmetry breaking.

Spontaneous symmetry breaking in polar fluids.

Project description:Spontaneous symmetry breaking and emergent polar order are each of fundamental importance to a range of scientific disciplines, as well as generating rich phase behaviour in liquid crystals (LCs). Here, we show the union of these phenomena to lead to two previously undiscovered polar liquid states of matter. Both phases have a lamellar structure with an inherent polar ordering of their constituent molecules. The first of these phases is characterised by polar order and a local tilted structure; the tilt direction processes about a helix orthogonal to the layer normal, the period of which is such that we observe selective reflection of light. The second new phase type is anti-ferroelectric, with the constituent molecules aligning orthogonally to the layer normal. This has led us to term the phases the SmCPH and SmAAF phases, respectively. Further to this, we obtain room temperature ferroelectric nematic (NF) and SmCPH phases via binary mixture formulation of the novel materials described here with a standard NF compound (DIO), with the resultant materials having melting points (and/or glass transitions) which are significantly below ambient temperature. The new soft matter phase types discovered herein can be considered as electrical analogues of topological structures of magnetic spins in hard matter.

Spontaneous symmetry breaking in genome evolution.

Project description:The quest for evolutionary mechanisms providing separation between the coding (exons) and noncoding (introns) parts of genomic DNA remains an important focus of genetics. This work combines an analysis of the most recent achievements of genomics and fundamental concepts of random processes to provide a novel point of view on genome evolution. Exon sizes in sequenced genomes show a lognormal distribution typical of a random Kolmogoroff fractioning process. This implies that the process of intron incretion may be independent of exon size, and therefore could be dependent on intron-exon boundaries. All genomes examined have two distinctive classes of exons, each with different evolutionary histories. In the framework proposed in this article, these two classes of exons can be derived from a hypothetical ancestral genome by (spontaneous) symmetry breaking. We note that one of these exon classes comprises mostly alternatively spliced exons.

Spontaneous symmetry breaking in plasmon lattice lasers.

Project description:Spontaneous symmetry breaking (SSB) is key for our understanding of phase transitions and the spontaneous emergence of order. In this work, we report that, for a two-dimensional (2D) periodic metasurface with gain, SSB occurs in the lasing transition. We study diffractive hexagonal plasmon nanoparticle lattices, where the K-points in momentum space provide two modes that are degenerate in frequency and identically distributed in space. Using femtosecond pulses to energize the gain medium, we simultaneously capture single-shot real-space and Fourier-space images of laser emission. By combining Fourier and real space, we resolve the two order parameters for which symmetry breaking simultaneously occurs: spatial parity and U(1) (rotational) symmetry breaking, evident respectively as random relative mode amplitude and phase. The methodology reported in this work is generally applicable to 2D plasmonic and dielectric metasurfaces and opens numerous opportunities for the study of SSB and the emergence of spatial coherence in metaphotonics.

Spontaneous symmetry breaking in interdependent networked game.

Project description:Spatial evolution game has traditionally assumed that players interact with direct neighbors on a single network, which is isolated and not influenced by other systems. However, this is not fully consistent with recent research identification that interactions between networks play a crucial rule for the outcome of evolutionary games taking place on them. In this work, we introduce the simple game model into the interdependent networks composed of two networks. By means of imitation dynamics, we display that when the interdependent factor α is smaller than a threshold value α(C), the symmetry of cooperation can be guaranteed. Interestingly, as interdependent factor exceeds α(C), spontaneous symmetry breaking of fraction of cooperators presents itself between different networks. With respect to the breakage of symmetry, it is induced by asynchronous expansion between heterogeneous strategy couples of both networks, which further enriches the content of spatial reciprocity. Moreover, our results can be well predicted by the strategy-couple pair approximation method.

Spontaneous breaking of symmetry in overlapping cell instance segmentation using diffusion models.

Project description:Instance segmentation is the task of assigning unique identifiers to individual objects in images. Solving this task requires breaking the inherent symmetry that semantically similar objects must result in distinct outputs. Deep learning algorithms bypass this break-of-symmetry by training specialized predictors or by utilizing intermediate label representations. However, many of these approaches break down when faced with overlapping labels that are ubiquitous in biomedical imaging, for instance for segmenting cell layers. Here, we discuss the reason for this failure and offer a novel approach for instance segmentation based on diffusion models that breaks this symmetry spontaneously. Our method outputs pixel-level instance segmentations matching the performance of models such as cellpose on the cellpose fluorescent cell dataset, while also permitting overlapping labels.

Spontaneous symmetry breaking propulsion of chemically coated magnetic microparticles.

Project description:Chemically coated micro/nanoparticles are often used in medicine to enhance drug delivery and increase drug up-take into specific areas of the body. Using a recently discovered spontaneous symmetry breaking propulsion mechanism, we demonstrate that chemically coated microparticles can swim through mucus solution under precise navigation and that certain functionalizations can dynamically change propulsion behavior. For this investigation biotin, Bitotin-PEG3-amine, and biotin chitosan were chemically functionalized onto the surfaces of magnetic microparticles using an avidin-biotin complex. These chemicals were chosen because they are used prolifically in drug delivery applications, with PEG and chitosan having well known mucoadhesive effects. Coated microparticles were then suspended in mucus synthesized from porcine stomach mucins and propelled using rotating magnetic fields. The relationship between different chemical coatings, microparticle velocity, and controllability were thoroughly explored and discussed. Results indicate that the biotinylated surface coatings altered the propulsion behavior of microparticles, with performance differences interlinked to both magnetic field properties and localized mucus properties. Precisely controlled drug carrying microparticles are envisioned to help supplant traditional drug delivery methods and enhance existing medical techniques utilizing micro/nanoparticles.

Spontaneous mirror symmetry breaking in heterocatalytically coupled enantioselective replicators.

Project description:Chiral hypercycle replicators (first-order autocatalysis together with mutual cross-catalysis) formed from achiral or racemizing resources may lead to spontaneous mirror symmetry breaking (SMSB) without the need for additional heterochiral inhibition reactions, such as those of the Frank-like models, which are an obstacle for the emergence of evolutionary selection properties. The results indicate that the chemical models for the emergence of primordial autocatalytic self-reproducing systems, of and by themselves, can also explain naturally the emergence of biological homochirality.

Spontaneous breaking of time-reversal symmetry in topological superconductors.

Project description:We study the behavior of spinless fermions in superconducting state, in which the phases of the superconducting order parameter depend on the direction of the link. We find that the energy of the superconductor depends on the phase differences of the superconducting order parameter. The solutions for the phases corresponding to the energy minimuma, lead to a topological superconducting state with the nontrivial Chern numbers. We focus our quantitative analysis on the properties of topological states of superconductors with different crystalline symmetry and show that the phase transition in the topological superconducting state is result of spontaneous breaking of time-reversal symmetry in the superconducting state. The peculiarities in the chiral gapless edge modes behavior are studied, the Chern numbers are calculated.

Symmetry-breaking polarization driven by a Cdc42p GEF-PAK complex.

Project description:BackgroundIn 1952, Alan Turing suggested that spatial patterns could arise from homogeneous starting conditions by feedback amplification of stochastic fluctuations. One example of such self-organization, called symmetry breaking, involves spontaneous cell polarization in the absence of spatial cues. The conserved GTPase Cdc42p is essential for both guided and spontaneous polarization, and in budding yeast cells Cdc42p concentrates at a single site (the presumptive bud site) at the cortex. Cdc42p concentrates at a random cortical site during symmetry breaking in a manner that requires the scaffold protein Bem1p. The mechanism whereby Bem1p promotes this polarization was unknown.ResultsHere we show that Bem1p promotes symmetry breaking by assembling a complex in which both a Cdc42p-directed guanine nucleotide exchange factor (GEF) and a Cdc42p effector p21-activated kinase (PAK) associate with Bem1p. Analysis of Bem1p mutants indicates that both GEF and PAK must bind to the same molecule of Bem1p, and a protein fusion linking the yeast GEF and PAK bypasses the need for Bem1p. Although mammalian cells lack a Bem1p ortholog, they contain more complex multidomain GEFs that in some cases can directly interact with PAKs, and we show that yeast containing an artificial GEF with similar architecture can break symmetry even without Bem1p.ConclusionsYeast symmetry-breaking polarization involves a GEF-PAK complex that binds GTP-Cdc42p via the PAK and promotes local Cdc42p GTP-loading via the GEF. By generating fresh GTP-Cdc42p near pre-existing GTP-Cdc42p, the complex amplifies clusters of GTP-Cdc42p at the cortex. Our findings provide mechanistic insight into an evolutionarily conserved pattern-forming positive-feedback pathway.

Spontaneous chiral symmetry breaking in a random driven chemical system.

Project description:Living systems have evolved to efficiently consume available energy sources using an elaborate circuitry of chemical reactions which, puzzlingly, bear a strict restriction to asymmetric chiral configurations. While autocatalysis is known to promote such chiral symmetry breaking, whether a similar phenomenon may also be induced in a more general class of configurable chemical systems-via energy exploitation-is a sensible yet underappreciated possibility. This work examines this question within a model of randomly generated complex chemical networks. We show that chiral symmetry breaking may occur spontaneously and generically by harnessing energy sources from external environmental drives. Key to this transition are intrinsic fluctuations of achiral-to-chiral reactions and tight matching of system configurations to the environmental drives, which together amplify and sustain diverged enantiomer distributions. These asymmetric states emerge through steep energetic transitions from the corresponding symmetric states and sharply cluster as highly-dissipating states. The results thus demonstrate a generic mechanism in which energetic drives may give rise to homochirality in an otherwise totally symmetrical environment, and from an early-life perspective, might emerge as a competitive, energy-harvesting advantage.
