Electro-Responsive Conductive Blended Hydrogel Patch.

ABSTRACT: The proposed electro-responsive hydrogel has great benefit for transdermal drug delivery system (TDDS) applications. To improve the physical or chemical properties of hydrogels, a number of researchers have previously studied the mixing efficiencies of the blended hydrogels. However, few studies have focused on improving the electrical conductivity and drug delivery of the hydrogels. We developed a conductive blended hydrogel by mixing alginate with gelatin methacrylate (GelMA) and silver nanowire (AgNW). We demonstrated that and the tensile strength of blended hydrogels were increased by a factor of 1.8 by blending GelMA and the electrical conductivity was enhanced by a factor of 18 by the addition of AgNW. Furthermore, the GelMA-alginate-AgNW (Gel-Alg-AgNW) blended hydrogel patch enabled on-off controllable drug release, indicating 57% doxorubicin release in response to electrical stimulation (ES) application. Therefore, this electro-responsive blended hydrogel patch could be useful for smart drug delivery applications.

Conductive GelMA-Collagen-AgNW Blended Hydrogel for Smart Actuator.

Project description:Blended hydrogels play an important role in enhancing the properties (e.g., mechanical properties and conductivity) of hydrogels. In this study, we generated a conductive blended hydrogel, which was achieved by mixing gelatin methacrylate (GelMA) with collagen, and silver nanowire (AgNW). The ratio of GelMA, collagen and AgNW was optimized and was subsequently gelated by ultraviolet light (UV) and heat. The scanning electron microscope (SEM) image of the conductive blended hydrogels showed that collagen and AgNW were present in the GelMA hydrogel. Additionally, rheological analysis indicated that the mechanical properties of the conductive GelMA-collagen-AgNW blended hydrogels improved. Biocompatibility analysis confirmed that the human umbilical vein endothelial cells (HUVECs) encapsulated within the three-dimensional (3D), conductive blended hydrogels were highly viable. Furthermore, we confirmed that the molecule in the conductive blended hydrogel was released by electrical stimuli-mediated structural deformation. Therefore, this conductive GelMA-collagen-AgNW blended hydrogel could be potentially used as a smart actuator for drug delivery applications.

An Angle-Independent Multi-Color Display Electro-Responsive Hydrogel Film.

Project description:In nature, some organisms have the ability to camouflage to adapt to environmental changes; they blend with the environment by changing their skin colors. Such a phenomenon is of great significance for the research of adaptive camouflage materials. In this study, we propose a novel design scheme for the study of angle-independent photonic materials and successfully prepare an electrically tunable multi-color display angle-independent inverse opal photonic gel (IOPG). After photopolymerization of hydroxyethyl methacrylate with ionizable monomer acrylic acid (AA) in a long-range disordered opal template and etching, the angle-independent inverse opal photonic gel is obtained, presenting a single structural color. The electrically responsive color changes can be achieved at different angles. The color of the disordered AA-IOPG changes from green to blue-green when applying +4 V bias voltage and from green to orange when applying -4 V bias voltage. The electrochromism of the disordered AA-IOPG is mainly due to the local pH change caused by water electrolysis under bias voltage, which leads to a change of the swelling ratio. The disordered AA-IOPG shows high color tunability and durability through repeated opposite bias voltage tests, indicating that it is a promising conductive photonic material.

Multi-responsive and conductive bilayer hydrogel and its application in flexible devices.

Project description:Multi-stimuli-responsive hydrogels are intelligent materials that present advantages for application in soft devices compared with conventional machines. In this paper, we prepared a bilayer hydrogel consisting of a poly(2-(dimethylamino)ethyl methacrylate) layer and a poly(N-isopropylacrylamide) layer. The hydrogel responded to temperature, pH, NaCl, and ethanol by undergoing bending deformation. At 40 °C, it only took 23 s for the hydrogel to bend nearly 300°. Carbon black was also introduced into the hydrogel network to render it conductive. Based on its multi-stimuli-responsive properties and conductivity, the hydrogel was used to construct a 4-arm gripper, thermistor, and finger movement monitor. The time required to grip and release an object was 141 s. The resistance changed with temperature, which affected the brightness of an LED. Finger motions were monitored, and the bending angle could be distinguished.

Cardiac-Adaptive Conductive Hydrogel Patch Enabling Construction of Mechanical-Electrical Anisotropic Microenvironment for Heart Repair.

Project description:The biomimetic construction of a microstructural-mechanical-electrical anisotropic microenvironment adaptive to the native cardiac tissue is essential to repair myocardial infarction (MI). Inspired by the 3D anisotropic characteristic of the natural fish swim bladder (FSB), a novel flexible, anisotropic, and conductive hydrogel was developed for tissue-specific adaptation to the anisotropic structural, conductive, and mechanical features of the native cardiac extracellular matrix. The results revealed that the originally stiff, homogeneous FSB film was tailored to a highly flexible anisotropic hydrogel, enabling its potential as a functional engineered cardiac patch (ECP). In vitro and in vivo experiments demonstrated the enhanced electrophysiological activity, maturation, elongation, and orientation of cardiomyocytes (CMs), and marked MI repair performance with reduced CM apoptosis and myocardial fibrosis, thereby promoting cell retention, myogenesis, and vascularization, as well as improving electrical integration. Our findings offer a potential strategy for functional ECP and provides a novel strategy to bionically simulate the complex cardiac repair environment.

Responsive and self-healing structural color supramolecular hydrogel patch for diabetic wound treatment.

Project description:The treatment of diabetic wounds remains a great challenge for medical community. Here, we present a novel structural color supramolecular hydrogel patch for diabetic wound treatment. This hydrogel patch was created by using N-acryloyl glycinamide (NAGA) and 1-vinyl-1,2,4-triazole (VTZ) mixed supramolecular hydrogel as the inverse opal scaffold, and temperature responsive poly(N-isopropylacrylamide) (PNIPAM) hydrogel loaded with vascular endothelial cell growth factor (VEGF) as a filler. Supramolecular hydrogel renders hydrogel patch with superior mechanical properties, in which NAGA and VTZ also provide self-healing and antibacterial properties, respectively. Besides, as the existence of PNIPAM, the hydrogel patch was endowed with thermal-responsiveness property, which could release actives in response to temperature stimulus. Given these excellent performances, we have demonstrated that the supramolecular hydrogel patch could significantly enhance the wound healing process in diabetes rats by downregulating the expression of inflammatory factors, promoting collagen deposition and angiogenesis. Attractively, due to responsive optical property of inverse opal scaffold, the hydrogel patch could display color-sensing behavior that was suitable for the wound monitoring and management as well as guidance of clinical treatment. These distinctive features indicate that the presented hydrogel patches have huge potential values in biomedical fields.

Operando Decoding Ion-Conductive Switch in Stimuli-Responsive Hydrogel by Nanodiamond-Based Quantum Sensing.

Project description:Thermal-responsive hydrogels are developed as ion-conductive switchs for energy storage devices, however, the molecule mechanism of switch on/off remains unclear. Here, poly(N-isopropylacrylamide-co-acrylamide) hydrogel is synthesized as a model material and nanodiamond (ND) based quantum sensing for phase change study is developed. First, micro-scale phase separation with cross-linked mesh structure after sol-gel transition is visualized in situ and water molecules are trapped by polymer chains and on a chemically "frozen" state. Then, the nano-scale inhomogeneous distributions of viscosity, thermal conductivity and ionic mobility in hydrogel at high temperature are observed by measuring the rotation, translation and zero-field splitting of NDs. Besides, the ionic mobility of hydrogel is found to be dependent not only on temperature but also on polymer concentration. These observations suggested that the physical "wall" induced by inhomogeneous phase separation at microscopic scale blocked the ion conduction pathways, providing a potential intrinsic explanation for ion migration shut-down of ionic hydrogels at high temperature.

Responsive-Hydrogel Aquabots.

Project description:It remains a challenge to produce soft robots that can mimic the responsive adaptability of living organisms. Rather than fabricating soft robots from bulk hydrogels,hydrogels are integrated into the interfacial assembly of aqueous two-phase systems to generate ultra-soft and elastic all-aqueous aquabots that exhibit responsive adaptability, that can shrink on demand and have electrically conductive functions. The adaptive functions of the aquabots provide a new platform to develop minimally invasive surgical devices, targeted drug delivery systems, and flexible electronic sensors and actuators.

Theoretical modeling of dendrite growth from conductive wire electro-polymerization.

Project description:Electropolymerization is a bottom-up materials engineering process of micro/nano-scale that utilizes electrical signals to deposit conducting dendrites morphologies by a redox reaction in the liquid phase. It resembles synaptogenesis in the brain, in which the electrical stimulation in the brain causes the formation of synapses from the cellular neural composites. The strategy has been recently explored for neuromorphic engineering by establishing link between the electrical signals and the dendrites' shapes. Since the geometry of these structures determines their electrochemical properties, understanding the mechanisms that regulate polymer assembly under electrically programmed conditions is an important aspect. In this manuscript, we simulate this phenomenon using mesoscale simulations, taking into account the important features of spatial-temporal potential mapping based on the time-varying signal, the motion of charged particles in the liquid due to the electric field, and the attachment of particles on the electrode. The study helps in visualizing the motion of the charged particles in different electrical conditions, which is not possible to probe experimentally. Consistent with the experiments, the higher AC frequency of electrical activities favors linear wire-like growth, while lower frequency leads to more dense and fractal dendrites' growth, and voltage offset leads to asymmetrical growth. We find that dendrites' shape and growth process systematically depend on particle concentration and random scattering. We discover that the different dendrites' architectures are associated with different Laplace and diffusion fields, which govern the monomers' trajectory and subsequent dendrites' growth. Such unconventional engineering routes could have a variety of applications from neuromorphic engineering to bottom-up computing strategies.

An injectable conductive hydrogel with dual responsive release of rosmarinic acid improves cardiac function and promotes repair after myocardial infarction.

Project description:Myocardial infarction (MI) causes irreversible damage to the heart muscle, seriously threatening the lives of patients. Injectable hydrogels have attracted extensive attention in the treatment of MI. By promoting the coupling of mechanical and electrical signals between cardiomyocytes, combined with synergistic therapeutic strategies targeting the pathological processes of inflammation, proliferation, and fibrotic remodeling after MI, it is expected to improve the therapeutic effect. In this study, a pH/ROS dual-responsive injectable hydrogel was developed by modifying xanthan gum and gelatin with reversible imine bond and boronic ester bond double crosslinking. By encapsulating polydopamine-rosmarinic acid nanoparticles to achieve on-demand drug release in response to the microenvironment of MI, thereby exerting anti-inflammatory, anti-apoptotic, and anti-fibrosis effects. By adding conductive composites to improve the conductivity and mechanical strength of the hydrogel, restore electrical signal transmission in the infarct area, promote synchronous contraction of cardiomyocytes, avoid induced arrhythmias, and induce angiogenesis. Furthermore, the multifunctional hydrogel promoted the expression of cardiac-specific markers to restore cardiac function after MI. The in vivo and in vitro results demonstrate the effectiveness of this synergistic comprehensive treatment strategy in MI treatment, showing great application potential to promote the repair of infarcted hearts.
