Eco-friendly textile desizing with indigenously produced amylase from Bacillus cereus AS2.

ABSTRACT: Starch is added to the fabric surface to secure weaving process. During finishing these sized particles are removed from the fabric and prepared it for printing and dyeing. Chemicals de-sizing agents damage fabric surfaces and reduce the quality of the product. An alternative to these conventional desizing agents is the use of biological molecules i.e. enzymes. The current study compares traditional de-sizing to bio-based de-sizing methods, as well as the optimization of fabric desizing settings using crude amylase. Amylase-producing Bacillus cereus AS2 was isolated from indigenous soil samples. The maximal fermentative de-sizing capability was discovered at 72 h, with no fabric surface degradation. Chemical desizing showed that the fabric lost all sizing agents to TEGEWA scale 9 within 1 h in presence of 5N HCl. Optimal studies for desizing showed that 1000 IU/ml of amylase resulted in maximum de-sizing within 15 h at 60 °C and 0.5% Triton-X. Water absorbance and weight loss, both parameters were used to check the desizing efficacy and it was found that de-sizing to same scale was occurred in the case of enzyme as well as commercially desized fabric. Enzyme desized cloth was found to be free of any starch particles in SEM micrographs, identical to industrially de-sized fabric, ensuring bioprocess efficacy.

Project description:Textile-based wearable humidity sensors are of great interest for human healthcare monitoring as they can provide critical human-physiology information. The demand for wearable and sustainable sensing technology has significantly promoted the development of eco-friendly sensing solutions for potential real-world applications. Herein, a biodegradable cotton (textile)-based wearable humidity sensor has been developed using fabsil-treated cotton fabric coated with a poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS) sensing layer. The structural, chemical composition, hygroscopicity, and morphological properties are examined using X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), contact angle measurement, and scanning electron microscopy (SEM) analysis. The developed sensor exhibited a nearly linear response (Adj. R-square value observed as 0.95035) over a broad relative humidity (RH) range from 25 to 91.5%RH displaying high sensitivity (26.1%/%RH). The sensor shows excellent reproducibility (on replica sensors with a margin of error ±1.98%) and appreciable stability/aging with time (>4.5 months), high flexibility (studied at bending angles 30°, 70°, 120°, and 150°), substantial response/recovery durations (suitable for multiple applications), and highly repeatable (multicyclic analysis) sensing performance. The prospective relevance of the developed humidity sensor toward healthcare applications is demonstrated via breathing rate monitoring (via a sensor attached to a face mask), distinguishing different breathing patterns (normal, deep, and fast), skin moisture monitoring, and neonatal care (diaper wetting). The multinode wireless connectivity is demonstrated using a Raspberry Pi Pico-based system for demonstrating the potential applicability of the developed sensor as a real-time humidity monitoring system for the healthcare sector. Further, the biodegradability analysis of the used textile is evaluated using the soil burial degradation test. The work suggests the potential applicability of the developed flexible and eco-friendly humidity sensor in wearable healthcare devices and other humidity sensing applications.

Project description:BackgroundBacteria usually secrete a variety of extracellular enzymes to degrade extracellular macromolecules to meet their nutritional needs and enhance their environmental adaptability. Bacillus cereus 0-9, a biocontrol bacterial strain isolated from wheat roots, has three genes annotated as encoding amylases in the genome, but their functions are unknown, and whether they are involved in the colonization process of the bacterium remains to be further studied.MethodsMutant gene strains and fluorescently tagged strains were constructed by homologous recombination, and amylase protein was expressed in the prokaryotic Escherichia coli BL21(DE3) expression system. The iodine staining method was used to measure the activity of amylase proteins. We further observed the colonization abilities of the test strains in wheat roots through frozen section technology.ResultsThe results showed that there were three amylase-encoding genes, amyC, amyP and amyS, in the B. cereus 0-9 genome. Among the three amylase encoding genes, only amyS produced extracellular amylase whose secretion was related to signal peptide at position 1-27. The AmyS protein encoded by the amyS gene is an α-amylase. The growth of Rhizoctonia cerealis was inhibited 84.7% by B. cereus 0-9, but the biocontrol ability of the ΔamyS strain decreased to 43.8% and that of ΔamyS/amyS was restored when the amyS gene was complemented. Furthermore, the biocontrol ability of the ΔamySec strain was decreased to 46.8%, almost the same as that of the ΔamyS mutant. Due to the deletion of the amyS gene, the colonization capacities of ΔamyS (RFP) and ΔamySec (RFP) in wheat roots decreased, while that of ΔamyS/amyS (RFP) was restored after the amyS gene was complemented, indicating that the amyS gene influences the colonization of B. cereus 0-9 in wheat roots. In addition, the colonization and biocontrol abilities of the mutant were restored after the addition of sugars, such as glucose and maltose.ConclusionsB. cereus 0-9 encodes three genes annotated as amylases, amyC, amyP and amyS. Only the deletion of the amyS gene with a signal peptide did not produce extracellular amylase. The AmyS protein encoded by the amyS gene is an α-amylase. Our results indicated that the amyS gene is closely related to the colonization abilities of B. cereus 0-9 in wheat roots and the biocontrol abilities of B. cereus 0-9 to fight against R. cerealis. The extracellular amylase produced by B. cereus 0-9 can hydrolyze starch and use glucose, maltose and other nutrients to meet the needs of bacterial growth. Therefore, it is very possible that the secretion and hydrolytic activities of extracellular amylase can promote the colonization of B. cereus 0-9 in wheat roots and play important roles in the prevention and control of plant diseases. Our results contribute to exploring the mechanisms of microbial colonization in plant roots.
