Effects of biochar application and nutrient fluctuation on the growth, and cadmium and nutrient uptake of Trifolium repens with different planting densities in Cd-contaminated soils

Project description:This study was undertaken to investigate combined toxic and genotoxic effects of cadmium (Cd) and arsenic (As) on white clover, a pollutant sensitive plant frequently used as environmental bioindicator. Plants were exposed to soil spiked with increasing concentrations of cadmium sulfate (20, 40 and 60 mg Kg-1) or sodium arsenite (5, 10 and 20 mg Kg-1) as well as with their combinations. Metal(loid) bioavailability was assessed after soil contamination, whereas plant growth, metal(loid) concentration in plant organs and DNA damage were measured at the end of plant exposition. Results showed that individual and joint toxicity and genotoxicity were related to the concentration of Cd and As measured in plant organs, and that As concentration was the most relevant variable. Joint effects on plant growth were additive or synergistic, whereas joint genotoxic effects were additive or antagonistic. The interaction between Cd and As occurred at both soil and plant level. In soil the presence of As limited the bioavailability of Cd, whereas the presence of Cd increased the bioavailability of As. Nevertheless only As biovailability determined the amount of As absorbed by plants. The amount of Cd absorbed by plant was not linearly correlated with the fraction of bioavailable Cd in soil suggesting the involvement of additional factors, such as plant uptake mechanisms. These results reveal that the simultaneous presence in soil of Cd and As, although producing an additive or synergistic toxic effect on Trifolium repens L. growth, generates a lower DNA damage.

Project description:Soil contamination by heavy metals such as Cd can pose a risk to the environment and human health. However, Cd is difficult to immobilize at low concentration levels in soil. Individually, Bacillus subtilis and biochar have been shown to be inefficient at immobilizing Cd in soil. In this study, corncob biochar was generated at different pyrolysis temperatures (300 °C-550 °C), and the Cd immobilization efficiency and performance of corncob biochar loaded with B. subtilis (CB@B) and corncob biochar alone (CB) were evaluated in solutions and in soil. The characterization (SEM and FTIR) of CB generated at different pyrolysis temperatures and CB generated at different pyrolysis temperatures in CB@B (300 °C-550 °C) indicated that a superior pore structure and abundant O-functional groups were obtained at a pyrolysis temperature of 400 °C for both CB@B and CB. The X-ray diffraction and X-ray photoelectron spectroscopy results indicate that the formation of Cd compounds was associated with the positive combined biosorption effect of the bacteria and biochar, electronic adsorption, activity of the O-functional groups (C[double bond, length as m-dash]O, COOH, OH, and Si-O-Si), and complexation between extracellular substances and Cd2+. Adsorption experiments were conducted in a solution to assess the effects of various operating parameters such as the time, pH, and adsorbent dose. The 400 °C-CB@B and 400 °C-CB samples achieved the largest reductions in the Cd concentration at 81.21% and 5.70%, respectively. Then, CaCl2 extraction experiments were conducted in soil, and using 0.25%-CB@B, a 55.21% decrease was realized in the Cd concentration after 56 days and a 16.71% increase was realized in soil pH to 8.38. No significant difference was observed in the CB-treated groups, among which 1.0%-CB achieved the largest reduction of 26.08% after 56 days and a 3.20% increase in the soil pH to 7.41. The Tessier sequential extraction method obtained similar trends. Overall, 400 °C-CB@B demonstrated outstanding immobilization efficiency and durability, indicating that it provided a safe and nutrient-rich habitat for B. subtilis to realize a synergistic effect for Cd immobilization.

Project description:The recently described complex nature of some dehydrin-coding sequences in Trifolium repens could explain the considerable variability among transcripts originating from a single gene.1 For some of the sequences the existence of natural antisense transcripts (NAT s), which could form sense-antisense (SAS) pairs, was predicted. The present study demonstrates that cis-natural antisense transcripts of 2 dehydrin types (YnKn and YnSKn) accumulate in white clover plants subjected to treatments with polyethylene glycol (PEG), abscisic acid (ABA), and high salt concentration. The isolated YnKn cis-NAT s mapped to sequence site enriched in alternative start codons. Some of the sense-antisense pairs exhibited inverse expression with differing profiles which depended on the applied stress. A natural antisense transcript coding for an ABC F family protein (a trans-NAT ) which shares short sequence homology with YnSKn dehydrin was identified in plants subjected to salt stress. Forthcoming experiments will evaluate the impact of NAT s on transcript abundances, elucidating the role of transcriptional and post-transcriptional interferences in the regulation of dehydrin levels under various abiotic stresses.
