Characterization of the complete mitochondrial genome and phylogenetic analysis of bean thrips Megalurothrips usitatus (Bagnall, 1913) (Thysanoptera: thripidae).

ABSTRACT: Megalurothrips usitatus (Bagnall, 1913) (Thysanoptera: Thripidae) is a widely distributed pest in Asia that primarily affects the production of snap beans and cowpea. The complete mitochondrial genome of Megalurothrips usitatus has been sequenced and annotated in this study, which is 17,209 bp long and contains 13 protein-coding genes (PCGs), two rRNAs, and 22 tRNA genes. Most of the protein-coding genes (PCGs) start with ATG except ND4 using TTG. Meanwhile, eight PCGs stop with TAA, four PCGs have an incomplete stop codon, and the gene Cytb ends with TAG. Phylogenetic analysis showed that M. usitatus is closely related to Frankliniella intonsa and F. occidentalis, providing a basis for the study of the mitochondrial evolution of Thripinae.

Project description:Megalurothrips usitatus is a serious pest on Vigna unguiculata. In this study, the complete mitochondrial genome sequence of M. usitatus was characterized and its phylogenetic relationship within the Order Thysanoptera was determined. The mitochondrial genome of M. usitatus was a circular molecule of 15426 bp in length, containing 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and the control region. It showed the typical insect mitochondrial genome arrangement. The AT content of the whole genome was 77.69% and the length of the control region was 567 bp with 78.66% AT content. The Maximum likelihood (ML) phylogenetic analysis based on mitochondrial protein-coding genes of 17 insect speciesshowed that M. usitatus is closest to Frankliniella occidentalis.

Chromosome-level genome assembly of bean flower thrips Megalurothrips usitatus (Thysanoptera: Thripidae).

Project description:Bean flower thrips Megalurothrips usitatus is a staple pest of cowpea and other legumes and causes dramatic economic losses. Its small size allows for easy concealment, and large reproductive capacity easily leads to infestations. Despite the importance of a genome in developing novel management strategies, genetic studies on M. usitatus remain limited. Thus, we generated a chromosome-level M. usitatus genome using a combination of PacBio long read and Hi-C technologies. The assembled genome was 238.14 Mb with a scaffold N50 of 13.85 Mb. The final genome was anchored into 16 pseudo-chromosomes containing 14,000 genes, of which 91.74% were functionally annotated. Comparative genomic analyses revealed that expanded gene families were enriched in fatty acid metabolism and detoxification metabolism (ABC transporters), and contracted gene families were strongly associated with chitin-based cuticle development and sensory perception of taste. In conclusion, this high-quality genome provides an invaluable resource for us to understand the thrips' ecology and genetics, contributing to pest management.

Genome-Wide Identification and Expression Profiling of Odorant-Binding Protein Genes in the Bean Flower Thrips Megalurothrips usitatus (Bagnall) (Thysanoptera: Thripidae).

Project description:Megalurothrips usitatus is an economically important vegetable pest. Because of the growing demand for reducing pesticide use on vegetables, new environmentally friendly strategies for controlling M. usitatus are urgently needed. Insect odorant-binding proteins are prospective targets for screening environmentally friendly odorant attractants for pest control. However, very little is known about OBP genes in M. usitatus. Here, we identified 14 OBPs in the M. usitatus genome using HMMER and BLAST. The chromosomal location showed that these OBPs were widely distributed across eight chromosomes. The analysis of the gene and protein structure characteristics of OBPs in M. usitatus revealed substantial diversity within the OBP gene family. The spatiotemporal expression profiles showed that ten out of 14 MusiOBPs displayed male biased expression, which were highly expressed in antennae, suggesting that they may play a crucial role in the recognition of host plant volatiles and thrips aggregation pheromones. Notably, only MusiOBP8 was significantly higher expressed in female adults, indicating a potential involvement in reproduction. Moreover, MusiOBP7 and MusiOBP13 were highly expressed in the pupae, indicating their possible role in immune responses. These results provide an important foundation for further exploration of the functions of the OBPs in M. usitatus.

Repellency of Plant Extracts against the Legume Flower Thrips Megalurothrips sjostedti (Thysanoptera: Thripidae).

Project description:Megalurothrips sjostedti Trybom is an important pest of cowpea (Vigna unguiculata L.) in Africa. To propose an alternative to chemical control, the repellency of 24 plant extracts was evaluated against adult female thrips of M. sjostedti in the laboratory. Plant extracts in ethanol were separately applied on a filter paper disk in a still air visual cue olfactometer. The results showed highly significant differences in repellency among extract type, concentration and their interactions. We classified the level of repellency into four categories as strong, good, moderate and weak or non- repellent based on hierarchical ascendant classification. We identified Piper nigrum, Cinnamomum zeylanicum, Cinnamomum cassia as strong repellents. Five extracts were classified as good, eight as moderate and the remaining eight extracts were weak or non-repellent. Repellency of the extracts increased with the concentration suggesting that the behavioral response of M. sjostedti was dose-dependent. Mono- and sesquiterpene hydrocarbon compounds from seven highly repellent extracts were identified by gas chromatography-mass spectrometry (GC/MS). The use of repellent extracts could be useful in developing integrated pest management strategies for thrips on legume crops. In this regard, the specific modes of action of the identified compounds need to be investigated to incorporate them into the existing crop protection strategies.

Identification and Virulence Characterization of Two Akanthomyces attenuatus Isolates Against Megalurothrips usitatus (Thysanoptera: Thripidae).

Project description:Megalurothrips usitatus (Bagnall) is one of the most harmful pests of leguminous plants. In order to expand our knowledge on the infection of M. usitatus by entomopathogenic fungi, two newly identified isolates of Akanthomyces attenuatus (Zare & Gams) were tested for their pathogenicity against M. usitatus. Both isolates of A. attenuatus (SCAUDCL-38 and SCAUDCL-56) were isolated from soil and were identified by morphological and molecular analyses. The adult females of M. usitatus were treated with five different concentrations (1 × 104, 1 × 105, 1 × 106, 1 × 107, and 1 × 108 conidia/mL) of the isolates. Our results revealed 76.25% and 57.5% mortality of M. usitatus after five days of treatment with 1 × 108 conidia/mL of SCAUDCL-38 and SCAUDCL-56, respectively. The median lethal concentrations (LC50) of SCAUDCL-38 and SCAUDCL-56 calculated through linear regression analysis after five days of fungal treatment of M. usitatus were 1.9 × 106 and 1.5 × 107 conidia/mL, respectively, whereas the median lethal time (LT50) observed for 1 × 108 conidia/mL of SCAUDCL-38 and SCAUDCL-56 were 3.52 days and 4.9 days, respectively. A. attenuatus isolates SCAUDCL-38 and SCAUDCL-56 are highly pathogenic strains of M. usitatus. These findings offer valuable information on the development and commercialization of alternative control measures against M. usitatus.

Parthenogenesis affects interspecific competition between Megalurothrips usitatus and Frankliniella intonsa (Thysanoptera: Thripidae) in changing environment: evidence from life table study.

Project description:The thrips Megalurothrips usitatus Bagnall and Frankliniella intonsa Trybom (Thysanoptera: Thripidae) are important pests in cowpea-growing areas of south China. Parthenogenesis is an important reproductive form of thysanopterans, and plays an important role in maintaining population growth. To understand the developmental and reproductive characteristics of these 2 thrips species during parthenogenesis, we compared the age-stage life tables of M. usitatus and F. intonsa on cowpea pods under natural regimes during the summer and winter. The results showed that the total preadult period and total preoviposition period of M. usitatus were significantly longer than those of F. intonsa in both seasons. Moreover, longevity of adult M. usitatus (29.53 days) was shorter compared with adult F. intonsa (34.00 days) in summer, whereas higher fecundity (220.8 eggs/female) and more oviposition days (37.83 days) were observed in M. usitatus compared with F. intonsa in winter (fecundity = 179.83 eggs/female, oviposition days = 33.03 days). The net and gross reproductive rates of M. usitatus were significantly greater than those of F. intonsa during winter. In addition, the intrinsic and finite rates of increase of M. usitatus were significantly lower than those of F. intonsa, and the mean generation time of M. usitatus was significantly longer than that of F. intonsa both in summer and winter. These results indicated that parthenogenesis has species specificity among thrips, which in turn affects population development, especially under changing environments.

Complete mitochondrial genome sequence for the Thrips hawaiiensis (Thysanoptera: Thripidae).

Project description:Thrips hawaiiensis (Morgan) (Thysanoptera: Thripidae) is a common Thysanoptera insect widely distributed in Asia and the Pacific, it damages various plants. In this study the complete mitochondrial genome of T. hawaiiensis was sequenced and characterized by using next-generation sequencing technique. The total length of the complete genome is 15,357 bp and A + T content of 77.8% (GeneBank accession No. MW582621). The T. hawaiiensis mitochondrial genome consists of 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes (tRNAs) and 2 non-coding control regions (Dloop region). According to previous studies, only a few complete mitochondrial genomes from Order Thysanoptera have been reported. Thus, T. hawaiiensis complete mitochondrial genome sequence reported will provide molecular information for mitochondrial genome research on Thysanoptera.

Identification and validation of reference genes for normalization of gene expression analysis using qRT-PCR in Megalurothrips usitatus (thysanoptera: thripidae).

Project description:Introduction: Gene expression analysis by reverse transcription quantitative polymerase chain reaction (qRT-PCR) has been widely used in research including insects. The selection of appropriate reference genes is the key to obtaining accurate and reliable results from qRT-PCR. However, studies on the expression stability of reference genes in Megalurothrips usitatus remain lacking. Methods: In this study, qRT-PCR was used to analyze the expression stability of candidate reference genes in M. usitatus. The expression levels of six candidate reference gene transcription of M. usitatus were analyzed. GeNorm, NormFinder, BestKeeper, and ΔCt were used to analyze the expression stability of M. usitatus treated with biological factors (developmental period treatment) and abiotic factors (light, temperature, insecticide treatment, respectively). Comprehensive stability ranking of candidate reference genes was recommended by RefFinder. Results and Discussion: Results showed that ribosomal protein S (RPS) was the most suitable expression in insecticide treatment. Ribosomal protein L (RPL) was the most suitable expression at developmental stage and light treatment, whereas elongation factor was the most suitable expression in temperature treatment. RefFinder was used to comprehensively analyze the above four treatments, and the results showed that RPL and actin (ACT) showed high stability in each treatment. Therefore, this study identified these two genes as reference genes in the qRT-PCR analysis of different treatment conditions of M. usitatus. Ourfindings will be beneficial for improving the accuracy of qRT-PCR analysis for future functional analysis of the target gene expression in M. usitatus.

Microbiome profiling of the onion thrips, Thrips tabaci Lindeman (Thysanoptera: Thripidae).

Project description:The gut microbial community structure of adult Thrips tabaci collected from 10 different agro-climatically diverse locations of India was characterized by using the Illumina MiSeq platform to amplify the V3 region of the 16S rRNA gene of bacteria present in the sampled insects. Analyses were performed to study the bacterial communities associated with Thrips tabaci in India. The complete bacterial metagenome of T. tabaci was comprised of 1662 OTUs of which 62.25% belong to known and 37.7% of unidentified/unknown bacteria. These OTUs constituted 21 bacterial phyla of 276 identified genera. Phylum Proteobacteria was predominant, followed by Actinobacteria, Firmicutes, Bacteroidetes and Cyanobacteria. Additionally, the occurrence of the reproductive endosymbiont, Wolbachia was detected at two locations (0.56%) of the total known OTUs. There is high variation in diversity and species richness among the different locations. Alpha-diversity metrics indicated the higher gut bacterial diversity at Bangalore and lowest at Rahuri whereas higher bacterial species richness at T. tabaci samples from Imphal and lowest at Jhalawar. Beta diversity analyses comparing bacterial communities between the samples showed distinct differences in bacterial community composition of T. tabaci samples from different locations. This paper also constitutes the first record of detailed bacterial communities associated with T. tabaci. The location-wise variation in microbial metagenome profile of T. tabaci suggests that bacterial diversity might be governed by its population genetic structure, environment and habitat.

Cypermethrin resistance and reproductive types in onion thrips, Thrips tabaci (Thysanoptera: Thripidae).

Project description:Cypermethrin resistance and reproductive types were examined for T. tabaci strains. Some arrhenotokous and thelytokous strains encoded the sodium channel mutation (T929I) involved in cypermethrin resistance. However, the resistance levels varied to some degree among the strains. A cytochrome P450 inhibitor, piperonyl butoxide, showed different synergistic effects on the strains examined. These results suggest that fundamental and additional levels of cypermethrin resistance in T. tabaci are conferred respectively by reduced sensitivity of the sodium channel and by cytochrome P450-mediated detoxification.
