The invisible witness: air and dust as DNA evidence of human occupancy in indoor premises.

ABSTRACT: Humans constantly shed deoxyribonucleic acid (DNA) into the surrounding environment. This DNA may either remain suspended in the air or it settles onto surfaces as indoor dust. In this study, we explored the potential use of human DNA recovered from air and dust to investigate crimes where there are no visible traces available-for example, from a recently vacated drugs factory where multiple workers had been present. Samples were collected from three indoor locations (offices, meeting rooms and laboratories) characterized by different occupancy types and cleaning regimes. The resultant DNA profiles were compared with the reference profiles of 55 occupants of the premises. Our findings showed that indoor dust samples are rich sources of DNA and provide an historical record of occupants within the specific locality of collection. Detectable levels of DNA were also observed in air and dust samples from ultra-clean forensic laboratories which can potentially contaminate casework samples. We provide a Bayesian statistical model to estimate the minimum number of dust samples needed to detect all inhabitants of a location. The results of this study suggest that air and dust could become novel sources of DNA evidence to identify current and past occupants of a crime scene.

Project description:The aim of this study was to analyse the quality of indoor air in sport facilities in one of the sport centres in Poland with respect to microclimatic parameters (temperature, humidity, and air flow velocity), particulate matter concentrations (PM10, PM4, PM2.5, and PM1), gas concentrations (oxygen, ozone, hydrogen sulphide, sulphur dioxide, volatile organic compounds, and benzopyrene), and microbial contamination (the total number of bacteria, specifically staphylococci, including Staphylococcus aureus, haemolytic bacteria, Enterobacteriaceae, Pseudomonas fluorescens, actinomycetes, and the total number of fungi and xerophilic fungi). Measurements were made three times in May 2022 at 28 sampling points in 5 different sporting areas (the climbing wall, swimming pool, swimming pool changing room, and basketball and badminton courts) depending on the time of day (morning or afternoon) and on the outside building. The obtained results were compared with the standards for air quality in sports facilities. The air temperature (21-31 °C) was at the upper limit of thermal comfort, while the air humidity (RH < 40%) in the sports halls in most of the locations was below demanded values. The values for dust pollution in all rooms, except the swimming pool, exceeded the permissible limits, especially in the afternoons. Climatic conditions correlated with a high concentration of dust in the indoor air. Particulate matter concentrations of all fractions exceeded the WHO guidelines in all researched premises; the largest exceedances of standards occurred for PM2.5 (five-fold) and for PM10 (two-fold). There were no exceedances of gaseous pollutant concentrations in the air, except for benzopyrene, which resulted from the influence of the outside air. The total number of bacteria (5.1 × 101-2.0 × 104 CFU m-3) and fungi (3.0 × 101-3.75 × 102 CFU m-3) was exceeded in the changing room and the climbing wall hall. An increased number of staphylococci in the afternoon was associated with a large number of people training. The increased concentration of xerophilic fungi in the air correlated with the high dust content and low air humidity. Along with the increase in the number of users in the afternoon and their activities, the concentration of dust (several times) and microorganisms (1-2 log) in the air increased by several times and 1-2 log, respectively. The present study indicates which air quality parameters should be monitored and provides guidelines on how to increase the comfort of those who practice sports and work in sports facilities.

Project description:Monitoring indoor air quality (IAQ) in schools is critical because children spend most of their daytime inside. One of the main air pollutant sources in urban areas is road traffic, which greatly influences air quality. Thus, this study addresses, in depth, the linkages of meteorology, ambient air pollution, and indoor activities with IAQ in a traffic-influenced school situated south of Madrid. The measurement period was from 22 November to 21 December 2017. Simultaneous measurements of indoor and outdoor PM1, PM2.5, and PM10 mass concentrations, ultrafine particle number concentration (PNC) and equivalent black carbon (eBC) were analyzed under different meteorological conditions. PNC and eBC outdoor concentrations and their temporal trend were similar among the sampling points, with all sites being influenced in the same way by traffic emissions. Strong correlations were found between indoor and outdoor concentrations, indicating that indoor pollution levels were significantly affected by outdoor sources. Especially, PNC and eBC had the same indoor/outdoor (I/O) trend, but indoor concentrations were lower. The time delay in indoor vs. outdoor concentrations varied between 0.5 and 2 h, depending on wind speed. Significant differences were found between different meteorological conditions (ANOVA p-values < 2.14 × 10-6). Atmospheric stability periods led to an increase in indoor and outdoor pollutant levels. However, the highest I/O ratios were found during atmospheric instability, especially for eBC (an average of 1.2). This might be related to rapid changes in the outdoor air concentrations induced by meteorology. Significant variations were observed in indoor PM10 concentrations during classroom occupancy (up to 230 µg m-3) vs. non-occupancy (up to 19 µg m-3) days, finding levels higher than outdoor ones. This was attributed to the scholarly activities in the classroom. Conversely, PNC and eBC concentrations only increased when the windows of the classroom were open. These findings have helped to establish practical recommendations and measures for improving the IAQ in this school and those of similar characteristics.

Project description:Environmental microbes have been associated with both protective and adverse health effects in children and adults. Epidemiological studies often rely on broad biomarkers of microbial exposure (i.e. endotoxin, 1 → 3-beta-d-glucan), but fail to identify the taxonomic composition of the microbial community. Our aim was to characterize the bacterial and fungal microbiome in different types of environmental samples collected in studies of human health effects. We determined the composition of microbial communities present in home, school and outdoor air samples by amplifying and sequencing regions of rRNA genes from bacteria (16S) and fungi (18S and ITS). Samples for this pilot study included indoor settled dust (from both a Boston area birth cohort study on Home Allergens and Asthma (HAA) (n = 12) and a study of school exposures and asthma symptoms (SICAS) (n = 1)), as well as fine and coarse concentrated outdoor ambient particulate (CAP) samples (n = 9). Sequencing of amplified 16S, 18S, and ITS regions was performed on the Roche-454 Life Sciences Titanium pyrosequencing platform. Indoor dust samples were dominated by Gram-positive bacteria (Firmicutes and Actinobacteria); the most abundant bacterial genera were those related to human flora (Streptococcus, Staphylococcus, Corynebacterium and Lactobacillus). Outdoor CAPs were dominated by Gram-negative Proteobacteria from water and soil sources, in particular the genera Acidovorax, and Brevundimonas (which were present at very low levels or entirely absent in indoor dust). Phylum-level fungal distributions identified by 18S or ITS regions showed very similar findings: a predominance of Ascomycota in indoor dust and Basidiomycota in outdoor CAPs. ITS sequencing of fungal genera in indoor dust showed significant proportions of Aureobasidium and Leptosphaerulina along with some contribution from Cryptococcus, Epicoccum, Aspergillus and the human commensal Malassezia. ITS sequencing detected more than 70 fungal genera in indoor dust not observed by culture. Microbiome sequencing is feasible for different types of archived environmental samples (indoor dust, and low biomass air particulate samples), and offers the potential to study how whole communities of microbes (including unculturable taxa) influence human health.
