Project description:Drug delivery is a difficult task in the field of dermal therapeutics, particularly in the treatment of burns, wounds, and skin diseases. Conventional drug delivery mediums have some limitations, including poor retention on skin/wound, inconvenience in administration, and uncontrolled drug release profile. Hydrogels able to absorb large amount of water and give a spontaneous response to stimuli imposed on them are an attractive solution to overcome the limitations of conventional drug delivery media. The objective of this study is to explore a green synthesis method for the development of thermo-responsive cellulose hydrogel using cellulose extracted from oil palm empty fruit bunches (OPEFB). A cold method was employed to prepare thermo-responsive cellulose hydrogels by incorporating OPEFB-extracted cellulose and Pluronic F127 (PF127) polymer. The performance of the synthesized thermo-responsive cellulose hydrogels were evaluated in terms of their swelling ratio, percentage of degradation, and in-vitro silver sulfadiazine (SSD) drug release. H8 thermo-responsive cellulose hydrogel with 20 w/v% PF127 and 3 w/v% OPEFB extracted cellulose content was the best formulation, given its high storage modulus and complex viscosity (81 kPa and 9.6 kPa.s, respectively), high swelling ratio (4.22 ± 0.70), and low degradation rate (31.3 ± 5.9%), in addition to high t50% value of 24 h in SSD in-vitro drug release to accomplish sustained drug release. The exploration of thermo-responsive cellulose hydrogel from OPEFB would promote cost-effective and sustainable drug delivery system with using abundantly available agricultural biomass.

Project description:The palm oil industry produces large amounts of empty fruit bunches (EFB) as waste. EFB are very recalcitrant toward further processing, although their valorization could create novel incentives and bio-economic opportunities for the industries involved. Herein, EFB have been successfully subjected to the OrganoCat pretreatment-using 2,5-furandicarboxylic acid as the biogenic catalyst-to fractionate and separate this lignocellulosic material into its main components in a single step. The pretreatment of EFB leads to the deacetylation and depolymerization of noncellulosic polysaccharides and to the partial delignification of the cellulosic fiber. The OrganoCat processing of EFB yielded 45 ± 0.5 wt % cellulose-enriched pulp, 20 ± 0.7 wt % extracted lignin, 3.8 ± 0.2 wt % furfural, and 11 ± 0.6 wt % hydrolyzed sugars. The obtained EFB-pulp showed high accessibility to cellulases, resulting in a glucan conversion of 73 ± 2% after 72 h (15 ± 2% after 1 h) with commercial cellulase cocktail (Accellerase 1500). Overall, the results suggest that the treatment of the EFB material using OrganoCat may create promising paths for the full valorization of EFBs.

Project description:This study investigated the effects of various pretreatment methods on the anaerobic digestibility of oil palm empty fruit bunches (EFB) for methane production. Pretreatment methods included weak alkaline (2 % Ca(OH)2), weak acid (2 % acetic acid), acidified palm oil mill effluent (POME), biogas effluent, hydrothermal (180 °C, 190 °C, and 200 °C), and microwave pretreatments. All pretreatment methods enhanced methane yield compared to untreated EFB (189.45 mL-CH4/g-VS), with weak alkaline pretreatment being the most effective (277.11 mL-CH4/g-VS), followed by hydrothermal pretreatment at 180 °C (244.33 mL-CH4/g-VS) and biogas effluent pretreatment (238.32 mL-CH4/g-VS). The enhanced methane yield was attributed to increased cellulose content (45.5 % for weak alkaline pretreatment), reduced hemicellulose (18.0 % for hydrothermal pretreatment at 200 °C), and lignin contents (19.0 % for hydrothermal pretreatment at 200 °C), decreased crystallinity index (40.0 % for hydrothermal pretreatment at 200 °C), and increased surface area. Weak alkaline pretreatment also showed the highest net energy balance (8.73 kJ/g-VS) and a short break-even point (2 years). Microbial community analysis revealed that weak alkaline pretreatment favored the growth of syntrophic acetate-oxidizing bacteria and hydrogenotrophic methanogens, contributing to improved methane yield. This study demonstrates the potential of EFB pretreatment, particularly weak alkaline and biogas effluent pretreatment, for enhancing methane production and sustainable management of palm oil mill waste.

Project description:ObjectiveThis research was aimed to isolate cellulolytic molds in empty fruit bunches of oil palm (EFBOP) and soils from palm oil plantation area and identify their enzyme activities to digest EFBOP.ResultsA total of seven molds were successfully isolated and screened for their enzyme activities from EFBOP and the soils. The enzymes from each isolate were produced in submerged culture using Mineral Mandels and 3% of alkali pretreated pollard in triplicates. The results indicated that all of the isolates were able to hydrolyze Carboxymethyl Cellulose (CMC), Whatmann No. 1 filter paper, and also EFBOP to sugars with reducing ends that reacted to 3,5-Dinitrosalicylic acid (DNS). The CMCase activity of isolate X showed the highest while the lowest was found for isolate MT8. Filter paperase (FPase) activity of isolate X performed the highest wile the lowest were found from isolate MT3 and MT6. The saccharification activity of isolate P showed the highest while MT6 performed the lowest activity.

Project description:In light of environmental issues, lignocellulosic empty fruit bunch (EFB) biomass is promoted as a carbon-neutral, environmentally friendly, and renewable alternative feedstock. A comprehensive environmental assessment of EFB biorefineries is critical for determining their sustainability in parallel with the bioeconomy policy. Nonetheless, no life cycle assessment (LCA) has been performed on co-producing food and biochemicals (furfural and glucose) derived from EFB biomass. This research is the first to evaluate the environmental performance of the furfural and glucose co-production processes from EFB biomass. Environmental analysis is conducted using a prospective gate-to-gate LCA for four impact categories, including global warming potential (GWP), acidification (ADP), eutrophication (EP), and human toxicity (HT). Aspen Plus is used to simulate the co-production process of furfural and glucose as well as generate mass and energy balances for LCA inventory data usage. The findings suggest that the environmental footprint in respect of GWP, ADP, EP, and HT is 4846.85 kg CO2 equivalent per ton EFB, 7.24 kg SO2 equivalent per ton EFB, 1.52 kg PO4 equivalent per ton EFB, and 2.62E-05 kg 1,4-DB equivalent per ton EFB, respectively. The normalized overall impact scores for GWP, ADP, EP, and HT are 1.16E-10, 2.28E-11, 6.12E-10, and 2.18E-17 years/ton of EFB, respectively. In summary, the proposed integrated plant is not only economically profitable but also environmentally sustainable. In the attempt to enhance the Malaysian economic sector based on the EFB, this study has the potential to serve as an indicator of the environmental sustainability of the palm oil industry.Graphical abstractSupplementary informationThe online version contains supplementary material available at 10.1007/s10668-022-02633-8.

Project description:Oil palm empty fruit bunches (OPEFB) are the most abundant, inexpensive, and environmentally friendly lignocellulosic biomass in Malaysia. Investigations on the microbial diversity of decaying OPEFB may reveal microbes with complex enzymes that have the potential to enhance the conversion of lignocellulose into second-generation biofuels as well as the production of other value-added products. In the present study, fungal and bacterial diversities in decaying OPEFB were identified using Illumina MiSeq sequencing of the V3 region of the 16S rRNA gene and V4 region of the 18S rRNA gene. Fungal diversity in decaying OPEFB was dominated by the phylum Ascomycota (14.43%), while most of the bacterial sequences retrieved belonged to Proteobacteria (76.71%). Three bacterial strains isolated from decaying OPEFB, designated as S18, S20, and S36, appeared to grow with extracted OPEFB-lignin and Kraft lignin (KL) as the sole carbon source. 16S rRNA gene sequencing identified the 3 isolates as Paenibacillus sp.. The molecular weight distribution of KL before and after degradation showed significant depolymerization when treated with bacterial strains S18, S20, and S36. The presence of low-molecular-weight lignin-related compounds, such as vanillin and 2-methoxyphenol derivatives, which were detected by a GC-MS analysis, confirmed the KL-degrading activities of isolated Paenibacillus strains.

Project description:This paper aims to examine the effect of cellulose nanocrystals (CNCs) derived from oil palm empty fruit bunch fiber (EFB) incorporating cement mortar on its structural performances. Cellulose nanocrystals (CNCs) were extracted from ?-cellulose extracted from EFB using an acid hydrolysis process with a concentration of acid used was 64% w/v under the temperature of 45?°C for 60?minutes. The Cellulose nanocrystals (CNCs) were mixed into the cement mortar ranging from 0 to 0.8% w/w and its mechanical properties were determined. The developed CNCs mortar was characterized for their compressive and flexural properties as well as microstructure. The influence of CNCs concentration, curing method, dispersion of CNCs on mortar's mechanical performance was thoroughly examined to find out the optimum condition. Overall results revealed that an addition of 0.4% cellulose nanocrystals has shown to increase the compressive and flexural strength to 46% and 20%, respectively cured under the wrapping method. The hydration of cementitious composites also improved significantly with the addition of CNCs by the formation of highly crystalline of portlandite observed under the XRD test. This present work demonstrates the importance of palm oil empty fruit bunch waste as a sustainable resource of cellulose nanocrystals admixture to achieve structural strength of cement mortar and promotes green technologies in construction.

Project description:The present work reports the biobleaching effect on OPEFB pulp upon utilisation of extracellular xylano-pectinolytic enzymes simultaneously yielded from Bacillus amyloliquefaciens ADI2. The impacts of different doses, retention times, pH, and temperatures required for the pulp biobleaching process were delineated accordingly. Here, the OPEFB pulp was subjected to pre-treatment with xylano-pectinolytic enzymes generated from the same alkalo-thermotolerant isolate that yielded those of higher quality. Remarkable enhanced outcomes were observed across varying pulp attributes: for example, enzyme-treated pulp treated to chemical bleaching sequence generated improved brightness of 11.25%. This resulted in 11.25% of less chlorine or chemical consumption required for obtaining pulp with optical attributes identical to those generated via typical chemical bleaching processes. Ultimately, the reduced consumption of chlorine would minimise the organochlorine compounds found in an effluent, resulting in a lowered environmental effect of paper-making processes overall as a consequence. This will undoubtedly facilitate such environmentally-friendly technology incorporation in the paper pulp industry of today.

Project description:BackgroundAspergillus niger, along with many other lignocellulolytic fungi, has been widely used as a commercial workhorse for cellulase production. A fungal cellulase system generally includes three major classes of enzymes i.e., β-glucosidases, endoglucanases and cellobiohydrolases. Cellobiohydrolases (CBH) are vital to the degradation of crystalline cellulose present in lignocellulosic biomass. However, A. niger naturally secretes low levels of CBH. Hence, recombinant production of A. niger CBH is desirable to increase CBH production yield and also to allow biochemical characterisation of the recombinant CBH from A. niger.MethodsIn this study, the gene encoding a cellobiohydrolase B (cbhB) from A. niger ATCC 10574 was cloned and expressed in the methylotrophic yeast Pichia pastoris X-33. The recombinant CBHB was purified and characterised to study its biochemical and kinetic characteristics. To evaluate the potential of CBHB in assisting biomass conversion, CBHB was supplemented into a commercial cellulase preparation (Cellic® CTec2) and was used to hydrolyse oil palm empty fruit bunch (OPEFB), one of the most abundant lignocellulosic waste from the palm oil industry. To attain maximum saccharification, enzyme loadings were optimised by response surface methodology and the optimum point was validated experimentally. Hydrolysed OPEFB samples were analysed using attenuated total reflectance FTIR spectroscopy (ATR-FTIR) to screen for any compositional changes upon enzymatic treatment.ResultsRecombinant CBHB was over-expressed as a hyperglycosylated protein attached to N-glycans. CBHB was enzymatically active towards soluble substrates such as 4-methylumbelliferyl-β-D-cellobioside (MUC), p-nitrophenyl-cellobioside (pNPC) and p-nitrophenyl-cellobiotrioside (pNPG3) but was not active towards crystalline substrates like Avicel® and Sigmacell cellulose. Characterisation of purified CBHB using MUC as the model substrate revealed that optimum catalysis occurred at 50 °C and pH 4 but the enzyme was stable between pH 3 to 10 and 30 to 80 °C. Although CBHB on its own was unable to digest crystalline substrates, supplementation of CBHB (0.37%) with Cellic® CTec2 (30%) increased saccharification of OPEFB by 27%. Compositional analyses of the treated OPEFB samples revealed that CBHB supplementation reduced peak intensities of both crystalline cellulose Iα and Iβ in the treated OPEFB samples.DiscussionSince CBHB alone was inactive against crystalline cellulose, these data suggested that it might work synergistically with other components of Cellic® CTec2. CBHB supplements were desirable as they further increased hydrolysis of OPEFB when the performance of Cellic® CTec2 was theoretically capped at an enzyme loading of 34% in this study. Hence, A. niger CBHB was identified as a potential supplementary enzyme for the enzymatic hydrolysis of OPEFB.

Project description:Lignin is one of the most abundant natural resources that can be exploited for the bioproduction of value-added commodity chemicals. Oil palm empty fruit bunches (OPEFBs), byproducts of palm oil production, are abundant lignocellulosic biomass but largely used for energy and regarded as waste. Pretreatment of OPEFB lignin can yield a mixture of aromatic compounds that can potentially serve as substrates to produce commercially important chemicals. However, separation of the mixture into desired individual substrates is required, which involves expensive steps that undermine the utility of OPEFB lignin. Here, we report successful engineering of microbial hosts that can directly utilize heterogeneous mixtures derived from OPEFB lignin to produce commodity chemicals, adipic acid and levulinic acid. Furthermore, the corresponding bioconversion pathway was placed under a genetic controller to autonomously activate the conversion process as the cells are fed with a depolymerized OPEFB lignin mixture. This study demonstrates a simple, one-pot biosynthesis approach that directly utilizes derivatives of agricultural waste to produce commodity chemicals.
