Histogram-based features track Alzheimer's progression in brain MRI.

ABSTRACT: Alzheimer's disease is a form of general dementia marked by amyloid plaques, neurofibrillary tangles, and neuron degeneration. The disease has no cure, and early detection is critical in improving patient outcomes. Magnetic resonance imaging (MRI) is important in measuring neurodegeneration during the disease. Computer-aided image processing tools have been used to aid medical professionals in ascertaining a diagnosis of Alzheimer's in its early stages. As characteristics of non and very-mild dementia stages overlap, tracking the progression is challenging. Our work developed an adaptive multi-thresholding algorithm based on the morphology of the smoothed histogram to define features identifying neurodegeneration and track its progression as non, very mild, mild, and moderate. Gray and white matter volume, statistical moments, multi-thresholds, shrinkage, gray-to-white matter ratio, and three distance and angle values are mathematically derived. Decision tree, discriminant analysis, Naïve Bayes, SVM, KNN, ensemble, and neural network classifiers are designed to evaluate the proposed methodology with the performance metrics accuracy, recall, specificity, precision, F1 score, Matthew's correlation coefficient, and Kappa values. Experimental results showed that the proposed features successfully label the neurodegeneration stages.

Project description:Alzheimer's disease (AD) is one of the most common neurodegenerative illnesses (dementia) among the elderly. Recently, researchers have developed a new method for the instinctive analysis of AD based on machine learning and its subfield, deep learning. Recent state-of-the-art techniques consider multimodal diagnosis, which has been shown to achieve high accuracy compared to a unimodal prognosis. Furthermore, many studies have used structural magnetic resonance imaging (MRI) to measure brain volumes and the volume of subregions, as well as to search for diffuse changes in white/gray matter in the brain. In this study, T1-weighted structural MRI was used for the early classification of AD. MRI results in high-intensity visible features, making preprocessing and segmentation easy. To use this image modality, we acquired four types of datasets from each dataset's server. In this work, we downloaded 326 subjects from the National Research Center for Dementia homepage, 123 subjects from the Alzheimer's Disease Neuroimaging Initiative (ADNI) homepage, 121 subjects from the Alzheimer's Disease Repository Without Borders homepage, and 131 subjects from the National Alzheimer's Coordinating Center homepage. In our experiment, we used the multiatlas label propagation with expectation-maximization-based refinement segmentation method. We segmented the images into 138 anatomical morphometry images (in which 40 features belonged to subcortical volumes and the remaining 98 features belonged to cortical thickness). The entire dataset was split into a 70?:?30 (training and testing) ratio before classifying the data. A principal component analysis was used for dimensionality reduction. Then, the support vector machine radial basis function classifier was used for classification between two groups-AD versus health control (HC) and early mild cognitive impairment (MCI) (EMCI) versus late MCI (LMCI). The proposed method performed very well for all four types of dataset. For instance, for the AD versus HC group, the classifier achieved an area under curve (AUC) of more than 89% for each dataset. For the EMCI versus LMCI group, the classifier achieved an AUC of more than 80% for every dataset. Moreover, we also calculated Cohen kappa and Jaccard index statistical values for all datasets to evaluate the classification reliability. Finally, we compared our results with those of recently published state-of-the-art methods.

Project description:ObjectivesTo employ different automated convolutional neural network (CNN)-based transfer learning (TL) methods for both binary and multiclass classification of Alzheimer's disease (AD) using brain MRI.MethodsHerein, we applied three popular pre-trained CNN models (ResNet101, Xception, and InceptionV3) using a fine-tuned approach of TL on 3D T1-weighted brain MRI from a subset of ADNI dataset (n = 305 subjects). To evaluate power of TL, the aforementioned networks were also trained from scratch for performance comparison. Initially, Unet network segmentedthe MRI scans into characteristic components of gray matter (GM), white matter (WM), and cerebrospinal fluid (CSF). The proposed networks were trained and tested over the pre-processed and augmented segmented and whole images for both binary (NC/AD + progressive mild cognitive impairment (pMCI)+stable MCI (sMCI)) and 4-class (AD/pMCI/sMCI/NC) classification. Also, two independent test sets from the OASIS (n = 30) and AIBL (n = 60) datasets were used to externally assess the performance of the proposed algorithms.ResultsThe proposed TL-based CNN models achieved better performance compared to the training CNN models from scratch. On the ADNI test set, InceptionV3-TL achieved the highest accuracy of 93.75% and AUC of 92.0% for binary classification, as well as the highest accuracy of 93.75% and AUC of 96.0% for multiclass classification of AD on the whole images. On the OASIS test set, InceptionV3-TL outperformed two other models by achieving 93.33% accuracy with 93.0% AUC in binary classification of AD on the whole images. On the AIBL test set, InceptionV3-TL also outperformed two other models in both binary and multiclass classification tasks on the whole MR images and achieved accuracy/AUC of 93.33%/95.0% and 90.0%/93.0%, respectively. The GM segment as input provided the highest performance in both binary and multiclass classification of AD, as compared to the WM and CSF segments.ConclusionThis study demonstrates the potential of applying deep TL approach for automated detection and classification of AD using brain MRI with high accuracy and robustness across internal and external test data, suggesting that these models can possibly be used as a supportive tool to assist clinicians in creating objective opinion and correct diagnosis.Advances in knowledgeWe used CNN-based TL approaches and the augmentation techniques to overcome the insufficient data problem. Our study provides evidence that deep TL algorithms can be used for both binary and multiclass classification of AD with high accuracy.
