First report of Sarcocystis halieti (Apicomplexa) in bearded vulture (Gypaetus barbatus).

ABSTRACT: At least three Sarcocystis species (S. falcatula, S. halieti and S. wobeseri-like) have been detected infecting raptorial birds. By histopathology and PCR-sequencing of the ITS1 marker, S. halieti was detected in a bearded vulture (Gypaetus barbatus) and a black kite (Milvus migrans) from the Catalonia region in North Spain. The 241 bp-long sequences obtained from the Sarcocystis organisms detected in both raptors showed 97.5-99.6% and 97.9-100% similarity with those of previously identified S. halieti; also, the phylogenetic trees generated placed the identified sequences together with other sequences of S. halieti available in GenBank. In sum, the description of the bearded vulture as a new intermediate host for S. halieti adds new insights on the complex epidemiology of the genus involving avian hosts.

The characterization of bearded vulture (Gypaetus barbatus) coprolites in the archaeological record.

Project description:The archaeological record of the Lagar Velho rock shelter (Lapedo Valley, Leiria, Portugal) bears testimony to several significant Upper Palaeolithic occupations, most notably the Lapedo Child burial (LV1) dating from the Gravettian. Excavations undertaken at the site since 2018 have seen the recovery of a large quantity of coprolites, above all in layer 143 (c. 29 ka cal BP). The study of these fossilized remains points to the bearded vulture (Gypaetus barbatus) as the main coprogenic agent and provides the first descriptions of these avian coprolites in archaeological assemblages. The analyses reported involved the comparison of the coprogenic samples with modern bearded vulture scats. A new morphotype is proposed for discriminating the faeces of this avian scavenger based on (1) macroscopic analyses, (2) morphometric comparisons with other fossil and modern scats and (3) their mineralogical and elemental composition. Among the criteria proposed here to identify the coprolites of the bearded vulture are their cylindrical shape, diameter, pointed extremities and homogeneous porous texture, as well as their massive internal texture, hard consistency and total absence of bone inclusions (attributable in all likelihood to a high digastric juice acidity capable of dissolving bones). Our results indicate that, as well as being used by humans for short-term stays, the Lagar Velho rock shelter was used by the bearded vulture as a nesting site. We provide new evidence from Iberia of the activity of this avian scavenger as a bone accumulator in archaeological sites.

Evidence of genetic structure in the wide-ranging bearded vulture (Gypaetus barbatus (Linnaeus, 1758)).

Project description:BackgroundThe bearded vulture is sparsely distributed across a wide geographic range that extends over three continents (Africa, Europe and Asia). Restriction to high-altitude mountainous habitats, low breeding rates, lack of food and a heightened level of persecution have left many local populations severely diminished or extinct. Understanding the genetic connectivity and population structure of this threatened vulture species is critical for accurately assessing their conservation status, and for appropriately managing local populations through captive breeding programmes or translocations. Previous genetic assessments of the species were mainly focused on the European and Asian populations and included limited representation of the geographically isolated southern African population. A single mitochondrial study, which focused on the African populations of the bearded vulture, detected limited genetic differentiation between populations in Ethiopia and southern Africa, with reduced haplotype diversity in the southern Africa population. In this study, we extend the previous genetic assessments of the species by examining the phylogeography and genetic connectivity of global G. barbatus populations using a panel of 14 microsatellite loci.ResultsAnalyses revealed spatially correlated genetic differentiation between regional populations and low levels of gene flow between these population fragments. In contrast to the mitochondrial data, the microsatellite data support the management of genetically different populations as separate entities.ConclusionsLow genetic diversity and geographic isolation are known to adversely affect the evolutionary potential of a species in the long-term. The high inbreeding found in the southern African G. barbatus and, to a lesser extent, the northern African populations highlights the need for conservation programmes to effectively manage populations of this species and maintain extant genetic diversity.

Detailed morphological structure and phylogenetic relationships of Degeeriella punctifer (Phthiraptera: Philopteridae), a parasite of the bearded vulture Gypaetus barbatus (Accipitriformes: Accipitridae).

Project description:Habitat loss is one of the main threats to species survival and, in the case of parasites, it is their hosts that provide their habitat. Therefore, extinction even at local scale of host taxa also implies the extinction of their parasites in a process known as co-extinction. This is the case of the bearded vulture (Gypaetus barbatus), which almost became extinct at the beginning of the twentieth century. After several attempts, this species was successfully reintroduced into the Alps at the end of the twentieth century. We collected 25 lice specimens from an electrocuted bearded vulture from Susa (Italian Alps) that were morphologically identified as Degeeriella punctifer. Six individuals were studied by scanning electron microscopy, with particular emphasis on their cephalic sensorial structures, while four further specimens were characterized at molecular level by amplifying partial regions of the 12SrRNA, COX1 and elongation factor 1 alpha (EF-1) genes. From a morphological perspective, the number, type and arrangement of the sensillae on the two distal antennal segments is quite similar to that of other species of the family Philopteridae (Phthiraptera: Ischnocera). The mandibles and tarsal claws allow lice to cling firmly to their host's feathers. Phylogenetic analyses help unravel the paraphyletic nature of the genus Degeeriella and demonstrate the clear differentiation between lice parasitizing Accipitriformes and Falconiformes, as well as the close relationship between D. punctifer, D. fulva, D. nisus and Capraiella sp. that, along with other genera, parasitize rollers (Aves: Coraciiformes).

Differential range use between age classes of southern African Bearded Vultures Gypaetus barbatus.

Project description:Bearded Vulture Gypaetus barbatus movements were investigated in southern Africa to determine whether an individual's age, sex or breeding status influenced its ranging behaviour and to provide the information required to guide conservation activities. Data from satellite transmitters fitted to 18 individuals of four age classes were used to determine range size and use. Because of the nature of the movements of marked individuals, these data could be used to determine the overall foraging range of the entire population, which was estimated to be 51 767 km(2). Although juvenile, immature and sub-adult birds used different parts of the overall range, their combined foraging range was 65% (33 636 km(2)) of the overall range. Average adult home ranges (286 km(2)) were only around 1% the size of the average foraging ranges of non-adults (10 540 -25 985 km(2)), with those of breeding adults being even smaller (95 km(2)). Home ranges of breeding adults did not vary in size between seasons but adults utilized their home range more intensively whilst breeding, moving greater distances during the incubation and chick hatching period. Range size and use increased as non-adults aged. Immatures and sub-adults had larger range sizes during winter, but range use of non-adults did not vary seasonally. Range size and use did not differ between the sexes in any of the age classes. Information on home range size and use enables specific areas within the species' range to be targeted for management planning, education and conservation action.

Cosmetic colouring by Bearded Vultures Gypaetus barbatus: still no evidence for an antibacterial function.

Project description:Bearded Vultures regularly visit ferruginous springs for cosmetic purposes to obtain their reddish plumage colouration. Different hypotheses have been proposed to explain this deliberate application of adventitious colouration: (1) to signal individual dominance status; (2) to exploit an anti-bacterial effect of iron oxides or ochre to reduce feather degradation by bacteria and, in parallel (3) to enable incubating birds to transfer this protection to their developing embryos to increase hatching success. Here, we re-evaluate the antibacterial hypothesis using three experimental approaches: (a) by applying feather-degrading bacteria to stained and unstained bearded vulture feathers; (b) by assessing the antibacterial activity of ochre; and (c) by comparing the breeding success of orange individuals with pale ones. Our findings suggest that the in vitro addition of feather degrading Bacillus licheniformis to naturally stained Bearded Vulture feathers did not retard feather degradation compared to controls. Iron particles from red soil (ochre) or iron salts had no antibacterial effect on the growth of three species of bacteria (Escherichia coli, Kocuria rhizophila and Bacillus licheniformis), incubated either in the dark or under visible light. Finally, breeding success did not differ between territories occupied by pale individuals versus orange ones. These results run counter to the hypothesis that iron oxides have an antibacterial role in Bearded Vultures. The use of red soils by Bearded Vultures may function as a territorial status signal, but may also be involved in other processes, such as pair formation and the long-term maintenance of the pair bond, as suggested for the closely related Egyptian vulture Neophron percnopterus.

Spatial and temporal movements in Pyrenean bearded vultures (Gypaetus barbatus): Integrating movement ecology into conservation practice.

Project description:Understanding the movement of threatened species is important if we are to optimize management and conservation actions. Here, we describe the age and sex specific spatial and temporal ranging patterns of 19 bearded vultures Gypaetus barbatus tracked with GPS technology. Our findings suggest that spatial asymmetries are a consequence of breeding status and age-classes. Territorial individuals exploited home ranges of about 50 km2, while non-territorial birds used areas of around 10 000 km2 (with no seasonal differences). Mean daily movements differed between territorial (23.8 km) and non-territorial birds (46.1 km), and differences were also found between sexes in non-territorial birds. Daily maximum distances travelled per day also differed between territorial (8.2 km) and non-territorial individuals (26.5 km). Territorial females moved greater distances (12 km) than males (6.6 km). Taking into account high-use core areas (K20), Supplementary Feeding Sites (SFS) do not seem to play an important role in the use of space by bearded vultures. For non-territorial and territorial individuals, 54% and 46% of their home ranges (K90), respectively, were outside protected areas. Our findings will help develop guidelines for establishing priority areas based on spatial use, and also optimize management and conservation actions for this threatened species.

The importance of the nutritive value of old bones in the diet of Bearded vultures Gypaetus barbatus.

Project description:Vultures are central-place foragers and need to optimize their foraging behaviour to offset travel costs by increasing their energy gain. This process is more obvious in certain vulture species that do not feed their young by regurgitation and so must carry food items back to the nest. The Bearded Vulture Gypaetus barbatus is the only species with a bone-diet based. We analysed the chemical composition of bones and the age-related changes in their nutritive value to assess the differences in energy content between bones of differing age, body part and species. We found differences between specific anatomical parts, species and the age of the bones. Fresh bones contain 108% as much energy as fresh meat and, interestingly, dry bones retain 90% of the protein found in fresh bones. Dry femurs weighing 140 g retain enough protein to be comparable to 111 g of fresh meat, in energy terms. Compared to meat-eating species, the specialized osteophagous diet of the Bearded Vulture seems to have certain advantages. A better understanding of nutrient levels in food remains could help to improve theoretical foraging models, assist in conservation management, and even improve our understanding of the use of bones by early hominids.

Project description:Sarcocystis is a genus of intracellular parasitic protozoa that infects various species of mammals, birds, and reptiles worldwide. At least 46 Sarcocystis species naturally infect rodents as intermediate hosts producing tissue cysts. This study aimed to provide the first report and molecular characterisation of Sarcocystis spp. in muscles from plains viscacha (Lagostomus maximus) in Argentina. Muscle samples of 53 plains viscachas from three provinces of Argentina were processed by homogenisation and optical microscopy to detect tissue cysts. Positive samples were analysed by PCR-sequencing, using the following markers: 18S rRNA, ITS1, and coxI. The 18S rRNA and coxI consensus sequences were aligned with other sequences from Sarcocystis spp., and phylogenetic trees were constructed. Of all animals processed, 13.2% (7/53) harboured Sarcocystis sp. cysts. 18S rRNA consensus sequences were obtained from four muscle samples and one individual cyst, and they showed 99.88-100% similarity, except for the cyst sequence, which showed 97.11% homology. Similarities of only 96-97% were recorded in the 18S rRNA fragment with other Sarcocystis spp. whose sequences are available in the GenBank. The five coxI fragment sequences obtained were 100% identical and showed an identity of 99.41-99.48% with S. canis. For ITS1 only short and low-quality sequences were obtained. In the phylogenetic trees, all the sequences from plains viscachas were positioned together in a branch separated from other Sarcocystis spp. These results could be related to new Sarcocystis spp. producing sarcocysts in plains viscachas. Besides, comprehensive cyst morphological analysis using TEM from the new Sarcocystis species will allow a description of the cyst wall ultrastructure. In this sense, further studies are needed to deepen these findings and elucidate other potential intermediate and possible definitive hosts.
