Case report: Diagnosis and autogenous vaccine treatment of herpesvirus in a green turtle (Chelonia mydas) in Santa Marta, Colombia.

ABSTRACT: This study reports the first case of fibropapillomatosis (FP) in the green turtle Chelonia mydas that has been successfully diagnosed and treated in Colombia. Worldwide, FP has reached epizootic proportions as it has been reported in marine turtles of tropical and subtropical waters, and in severe cases, it reduces the probability of survival. Treatment has been elusive as multiple surgical excisions are needed due to tumor recurrence. In this case, one green turtle with multiple tumors was diagnosed by histopathology and molecular detection of the chelonid herpesvirus 5 (ChHV5) by means of amplification and sequencing of the DNA polymerase (DNApol) gene. Two separate treatments that consisted of autogenous vaccines and surgical excisions were applied; the first one had a partial success as one out of the tumors treated reappeared after 3 months post-treatment. Treatment 2 consisted of an autogenous vaccine enriched with adjuvants and applied at increasing doses, after which, the tumor significatively decreased in size and was surgically removed. At the end of the 6 months follow-up period, no tumor recurrence was observed, and the turtle was in apparent optimal health conditions. These findings, although limited, suggest a possible treatment that might help to contain this epizootic problem.

To examine the transcriptomic events associated with embryonic arrest, RNA-sequencing analysis was performed on embryos from freshly laid eggs and eggs incubated in either normoxia (oxygen tension ~159 mmHg) or hypoxia (<8 mmHg) for 36 hours (h) after oviposition (n = 5 per group). The patterns of gene expression differed markedly among the three experimental groups. Normal embryonic development in normoxia was associated with up-regulation of genes involved in DNA replication, the cell cycle, and mitosis, but these genes were commonly down-regulated after incubation in hypoxia. Many target genes of hypoxia inducible factors, including insulin-like growth factor binding protein 1, were down-regulated by normoxic incubation but upregulated by incubation in hypoxia. Notably, some of the transcriptomic effects of hypoxia in green turtle embryos resembled those reported by others to be associated with hypoxia-induced embryonic arrest in diverse taxa, including budding yeast (Saccharomyces cerevisiae), the annelid Caenorhabditis elegans, and zebrafish (Danio rerio). Thus, while among oviparous species, hypoxia-induced pre-ovipositional embryonic arrest appears to be a unique adaptation of turtles, mechanisms underlying hypoxia-induced embryonic arrest per se may be highly conserved across the diverse taxa in which this mechanism operates. We report the transciptional changes in Chelonia mydas embryos incubated in either normoxia or hypoxia comapred to freshly laid eggs.

Project description:This is the first assessment of catch rates for the legal, artisanal green turtle, Chelonia mydas, fishery in Caribbean Nicaragua. Data were collected by community members, monitoring up to 14 landing sites from 1991 to 2011. We examined take levels, and temporal and spatial variability in catch rates for the overall fishery, by region, and community using General Additive Mixed Models (GAMMs). More than 171,556 green turtles were killed during the period, with a mean estimated minimum 8,169±2,182 annually. There was a statistically significant decline in catch rates overall. Catch rates peaked in 1997 and 2002, followed by a downward trend, particularly from mid-2008 to the end of the study period. Similar downward trends were evident in both study regions. Community specific catch rate trends also indicated declines with decreases ranging from 21% to 90%. Decrease in catch rates in Nicaragua is cause for concern even though the principal source rookery at Tortuguero, Costa Rica, shows an increase in nesting activity. Explanations for the apparent discrepancy between the increasing trend at Tortuguero and decreasing catch rate trends in Nicaragua include: i) an increase in reproductive output, ii) insufficient time has passed to observe the impact of the fishery on the rookery due to a time lag, iii) changes in other segments of the population have not been detected since only nesting activity is monitored, iv) the expansive northern Nicaragua foraging ground may provide a refuge for a sufficient portion of the Tortuguero rookery, and/or v) a larger than expected contribution of non-Tortuguero rookeries occurring in Nicaragua turtle fishing areas. Our results highlight the need for close monitoring of rookeries and in-water aggregations in the Caribbean. Where consumptive use still occurs, nations sharing this resource should implement scientifically based limits on exploitation to ensure sustainability and mitigate impacts to regional population diversity.

Project description:BackgroundSeveral anatomical studies provide evidence that green turtles (Chelonia mydas) possess the necessary anatomy for colour vision. Behavioural experiments have previously been conducted with newly emerged hatchlings, concluding that they are attracted to shorter wavelengths compared to longer wavelengths within a terrestrial environment, suggesting a possible attraction towards blue. This paper assessed the colour vision of hatchlings within an aquatic environment, and investigated whether the attraction for shorter wavelengths remains consistent within water, whether the colour saturation of the chromatic stimuli was an important factor, and whether rearing and testing individual animals in different coloured housing tanks has an impact on their visual choices.MethodsForty-one hatchling green turtles were presented with a three-choice experiment where food was attached to three different coloured plates. The plates (blue, yellow, and red) were randomly arranged in the turtle's tank and four different colour saturations were tested (100, 75, 50, and 25%). Turtles were individually placed into their housing tanks (coloured either red, white, blue or grey) with three different colour plates in front of them, from the same saturation level. The colour of the plate with food first approached and bitten by the turtle was recorded.ResultsThe colour of the tank in which an individual was reared, and where experiments were conducted, significantly influenced which food item was selected on the different coloured plates. While individual turtles preferred to select the food items associated with blue plates across the entire experiment (66.1% of the time compared to 18.2% and 15.7% for yellow and red plates respectively), the preference for blue plates was influenced by the colour of the rearing/experimental tank. Individuals raised in red, white or blue tanks appeared to consistently prefer food on blue plates, but there appeared to be no plate colour preference by turtles in grey tanks. There was no significant effect of either colour saturation or the spatial arrangement of the three colours within an experimental tank on colour choice, and no significant interaction between tank colour and colour saturation.DiscussionThesefindings confirm that the terrestrial preference towards shorter wavelength colours, such as blue, compared to longer wavelength colours remains consistent within an aquatic environment. This preference for blue continues even as the colour saturation reduces from 100% down to 25%, and the colours become darker. Thus, it is suggested that green turtle hatchlings have a strong attraction towards blue. This attraction, however, is influenced by the colour of the tank the turtles were raised in. While this supports the notion that environmental colour may influence individual turtle visual capabilities, it suggests that this relationship is more complicated, and requires further investigation.
