Project description:Due to the technical, environmental and economic problems caused by asphaltene precipitation, such as oil production reduction, well shut-ins and the necessity of EOR usage, the prediction of asphaltene precipitation seems to be vital. Considering the larger size of asphaltene molecules compared to the other hydrocarbon, it is reasonable to predict the precipitation using the Flory-Huggins theory. In this study, Flory-Huggins solution theory has been modified regarding the solvent molar volume. The modified model was used to predict the asphaltene precipitation of four oil samples in the absence and presence of the inhibitors. Then, the modeling data given by the Flory-Huggins theory was validated with the experimental data obtained by ASTM D-6560 standard method. The mean error at this modeling was 2-13%, which seems acceptable. The proposed model for the cases where an inhibitor is not involved has higher accuracy. The modified Flory-Huggins theory confirmed that the addition of inhibitors at all concentrations postpones the onset point. The average error of the modified model was found to be 4.5-9.8%, which is in a good range. Also, the model accuracy is less for situations where the asphaltene content of the crude oil is higher. Based on this study, the modification of Flory-Huggins theory, regarding the solvent molar volume leads to a lower error.

Project description:A self-consistent analytical solution for binodal concentrations of the two-component Flory-Huggins phase separation model is derived. We show that this form extends the validity of the Ginzburg-Landau expansion away from the critical point to cover the whole phase space. Furthermore, this analytical solution reveals an exponential scaling law of the dilute phase binodal concentration as a function of the interaction strength and chain length. We demonstrate explicitly the power of this approach by fitting experimental protein liquid-liquid phase separation boundaries to determine the effective chain length and solute-solvent interaction energies. Moreover, we demonstrate that this strategy allows us to resolve differences in interaction energy contributions of individual amino acids. This analytical framework can serve as a new way to decode the protein sequence grammar for liquid-liquid phase separation.

Project description:Thermoresponsive polymers are a promising material for drug nanocarrier preparation, which makes the study of their aggregation in physiological conditions very important. In this paper, the thermal behaviour of the thermoresponsive polymers poly(N-isopropylacrylamide), poly(2-isopropyl-2-oxazoline-co-2-n-propyl-2-oxazoline) and poly[(2-hydroxyethyl methacrylate)-co-oligo(ethylene glycol) methyl ether methacrylate] were studied in phosphate buffer (PBS) and solutions of its salts in concentration as in PBS. The thermal response of the polymers was measured using UV-Vis and dynamic light scattering (DLS). The salts shifted the cloud point temperature (TCP) of the (co)polymers to higher values compared to the TCP of aqueous polymer solutions. In PBS and NaCl solutions, all polymers exhibited an unexpected and previously unreported transmittance profile. During heating, an additional aggregation of polymers appeared above the TCP accompanied by the formation of a precipitate. In monosodium phosphate solutions and pure water, the studied polymers showed lower critical solution temperature (LCST-type) behaviour. DLS measurements showed that a salt influenced the size of the resulting polymer particles. The sizes and stability of particles depended on the heating rate. In PBS and NaCl solutions, the size of particles in the dispersion decreased above 60 °C, and the precipitate appeared on the bottom of the cuvette. The additional aggregation of polymer and its falling out of solution may hinder the removal of carriers from the body and has to be taken into account when preparing nanocarriers.

Project description:Amorphous solid dispersion (ASD) is one of the most promising enabling formulations featuring significant water solubility and bioavailability enhancements for biopharmaceutical classification system (BCS) class II and IV drugs. An accurate thermodynamic understanding of the ASD should be established for the ease of development of stable formulation with desired product performances. In this study, we report a first experimental approach combined with classic Flory-Huggins (F-H) modelling to understand the performances of ASD across the entire temperature and drug composition range. At low temperature and drug loading, water (moisture) was induced into the system to increase the mobility and accelerate the amorphous drug-amorphous polymer phase separation (AAPS). The binodal line indicating the boundary between one phase and AAPS of felodipine, PVPK15 and water ternary system was successfully measured, and the corresponding F-H interaction parameters (χ) for FD-PVPK15 binary system were derived. By combining dissolution/melting depression with AAPS approach, the relationship between temperature and drug loading with χ (Φ, T) for FD-PVPK15 system was modelled across the entire range as χ = 1.72 - 852/T + 5.17·Φ - 7.85·Φ2. This empirical equation can provide better understanding and prediction for the miscibility and stability of drug-polymer ASD at all conditions.

Project description:Soluble fibres are gaining increasing interest for functional food applications like bread, but their interaction with gluten and effects on dough rheology are not fully elucidated. This study hypothesized that soluble fibres influence gluten structure and dough rheology by acting as plasticizers and humectants. Plasticizing properties depend on the effective number of hydrogen bonding sites available in the fibre molecule (N OH,s ). Humectant properties are related to the water interaction parameter derived from analysis of the sorption behaviour. Oligo-fructoses, inulins, polydextrose and a glucose syrup were added individually and in mixtures to wheat dough to test the hypothesis. PCA and multi-linear regressions showed that the G' from temperature sweeps increased with an increase in the effective volume fraction of hydrogen bonding sites ( Φ w , e f f ) in the solvent and in the water interaction parameter (χ eff ). The enhanced G' corresponded to a reduction in tan(δ), indicating an increased elastic behaviour. The parameters Φ w , e f f and χ eff also explained the changes in phase transitions during heating, i.e. Tonset and Tpeak of starch gelatinization (R2 > 0.9). Image analysis of the gluten network revealed that fibre structure and physico-chemical properties influenced the gluten network by altering branching rate, lacunarity, and protein strand width. Comparing inulins and polydextrose of similar molecular weights (Mw) indicated that interactions with gluten were influenced more by N OH,s than Mw. High Mw inulins, with a linear structure, promoted junctions in the gluten network through hydrogen bonds, and possibly phase separation in gluten-rich and inulin-rich phases. In contrast, the more hydrophilic, branched polydextrose reduced junction formation in the gluten network due to fewer N OH,s . This study provides new insights into the physico-chemical properties of soluble fibres and their role in wheat dough functionality.

Project description:This study presents the theory for liquid-liquid phase separation for systems of molecules modeling monoclonal antibodies. Individual molecule is depicted as an assembly of seven hard spheres, organized to mimic the Y-shaped antibody. We consider the antibody-antibody interactions either through Fab, Fab' (two Fab fragments may be different), or Fc domain. Interaction between these three domains of the molecule (hereafter denoted as A, B, and C, respectively) is modeled by a short-range square-well attraction. To obtain numerical results for the model under study, we adapt Wertheim's thermodynamic perturbation theory. We use this model to calculate the liquid-liquid phase separation curve and the second virial coefficient B2. Various interaction scenarios are examined to see how the strength of the site-site interactions and their range shape the coexistence curve. In the asymmetric case, where an attraction between two sites is favored and the interaction energies for the other sites kept constant, critical temperature first increases and than strongly decreases. Some more microscopic information, for example, the probability for the particular two sites to be connected, has been calculated. Analysis of the experimental liquid-liquid phase diagrams, obtained from literature, is presented. In addition, we calculate the second virial coefficient under conditions leading to the liquid-liquid phase separation and present this quantity on the graph B2 versus protein concentration.

Project description:In this study, deep learning (DL)-based estimation of the Flory-Huggins χ parameter of A-B diblock copolymers from two-dimensional cross-sectional images of three-dimensional (3D) phase-separated structures were investigated. 3D structures with random networks of phase-separated domains were generated from real-space self-consistent field simulations in the 25-40 χN range for chain lengths (N) of 20 and 40. To confirm that the prepared data can be discriminated using DL, image classification was performed using the VGG-16 network. We comprehensively investigated the performances of the learned networks in the regression problem. The generalization ability was evaluated from independent images with the unlearned χN. We found that, except for large χN values, the standard deviation values were approximately 0.1 and 0.5 for A-component fractions of 0.2 and 0.35, respectively. The images for larger χN values were more difficult to distinguish. In addition, the learning performances for the 4-class problem were comparable to those for the 8-class problem, except when the χN values were large. This information is useful for the analysis of real experimental image data, where the variation of samples is limited.

Project description:Chlorophylls and their derivatives are currently used in a wide range of applications. To replace the volatile organic solvents commonly applied for their extraction from biomass, aqueous solutions of non-ionic surfactants are studied herein in the extraction of chlorophylls from spinach leaves. Aqueous solutions of several surfactants were screened, demonstrating that their hydrophilic-lipophilic balance (HLB) plays the pivotal role on the extraction performance, with the best results obtained for surfactants with a HLB ranging between 10 and 13. A response surface methodology (RSM) was then used to optimize operational conditions (surfactant concentration, solid-liquid ratio and temperature), leading to a maximum extraction yield of chlorophylls of 0.94 mg/g. After the extraction step, the chlorophylls-rich extract was concentrated by heating above the surfactant-water cloud point, leading to the separation into two-phases, and to a concentration factor of 9 and a recovery of 97% of chlorophylls in the surfactant-rich phase. The antioxidant activity of the extracts was finally appraised, showing that the antioxidant activity of the aqueous chlorophylls-rich extracts is higher than that obtained with volatile organic solvents. The obtained results show the potential of aqueous solutions of non-ionic surfactants to extract highly hydrophobic compounds from biomass and their possible direct use in cosmetic and nutraceutical applications, without requiring an additional recovery or purification step.

Project description:Predicting the electronic properties of aqueous liquids has been a long-standing challenge for quantum mechanical methods. However, it is a crucial step in understanding and predicting the key role played by aqueous solutions and electrolytes in a wide variety of emerging energy and environmental technologies, including battery and photoelectrochemical cell design. We propose an efficient and accurate approach to predict the electronic properties of aqueous solutions, on the basis of the combination of first-principles methods and experimental validation using state-of-the-art spectroscopic measurements. We present results of the photoelectron spectra of a broad range of solvated ions, showing that first-principles molecular dynamics simulations and electronic structure calculations using dielectric hybrid functionals provide a quantitative description of the electronic properties of the solvent and solutes, including excitation energies. The proposed computational framework is general and applicable to other liquids, thereby offering great promise in understanding and engineering solutions and liquid electrolytes for a variety of important energy technologies.

Project description:Aqueous solutions of some polymers exhibit a lower critical solution temperature (LCST); that is, they form phase-separated aggregates when heated above a threshold temperature. Such polymers found many promising (bio)medical applications, including in situ thermogelling with controlled drug release, polymer-supported radiotherapy (brachytherapy), immunotherapy, and wound dressing, among others. Yet, despite the extensive research on medicinal applications of thermoresponsive polymers, their biodistribution and fate after administration remained unknown. Thus, herein, they studied the pharmacokinetics of four different thermoresponsive polyacrylamides after intramuscular administration in mice. In vivo, these thermoresponsive polymers formed depots that subsequently dissolved with a two-phase kinetics (depot maturation, slow redissolution) with half-lives 2 weeks to 5 months, as depot vitrification prolonged their half-lives. Additionally, the decrease of TCP of a polymer solution increased the density of the intramuscular depot. Moreover, they detected secondary polymer depots in the kidneys and liver; these secondary depots also followed two-phase kinetics (depot maturation and slow dissolution), with half-lives 8 to 38 days (kidneys) and 15 to 22 days (liver). Overall, these findings may be used to tailor the properties of thermoresponsive polymers to meet the demands of their medicinal applications. Their methods may become a benchmark for future studies of polymer biodistribution.
