Project description:Ultra-running comprises running events longer than a marathon (>42.2 km). The prolonged duration of ultra-running leads to decrements in most or all physiological parameters and considerable energy expenditure (EE) and energy deficits. SG, 47 years, 162.5 cm, 49 kg, VO2max 4 mL/kg/min-1/2.37 L/min-1, ran continuously for 7 days on a treadmill in 3 h blocks followed by 30 min breaks and slept from 1-5 a.m. Heart rate (HR) oxygen uptake (VO2), rating of perceived exertion, weight, blood lactate (mmol·L-1), haemoglobin (g·dL), haematocrit (%) and glucose (mmol·L-1), and nutrition and hydration were recorded. SG ran for 17.5 h/day, covering ~120 km/day at ~7 km/h. Energy expenditure for each 24 h period was 6878 kcal/day and energy intake (EI) was 2701 kcal/day. EE was 382 kcal/h, with 66.6% from fat and 33.4% from carbohydrate oxidation. 7 day EI was 26,989 kcal and EE was 48,147 kcal, with a total energy deficit (ED) of 21,158 kcal. Average VO2 was 1.2 L·min-1/24.7 mL·kg·min-1, Respriatory echange ratio (RER) 0.80 ± 0.03, HR 120-125 b·min-1. Weight increased from 48.6 to 49.5 kg. Haemoglobin decreased from 13.7 to 11 g·dL and haematocrit decreased from 40% to 33%. SG ran 833.05 km. SG exhibits an enhanced fat metabolism through which she had a large daily ED. Her success can be attributed to a combination of physiological and psychological factors.

Project description:Understanding the influence of running-induced acute fatigue on the homeostasis of the body is essential to mitigate the adverse effects and optimize positive adaptations to training. Fatigue is a multifactorial phenomenon, which influences biomechanical, physiological, and psychological facets. This work aimed to assess the evolution of these three facets with acute fatigue during a half-marathon. 13 recreational runners were equipped with one inertial measurement unit (IMU) on each foot, one combined global navigation satellite system-IMU-electrocardiogram sensor on the chest, and an Android smartphone equipped with an audio recording application. Spatio-temporal parameters for the running gait, along with the heart rate, its variability and complexity were computed using validated algorithms. Perceived fatigability was assessed using the rating-of-fatigue (ROF) scale at every 10 min of the race. The data was split into eight equal segments, corresponding to at least one ROF value per segment, and only level running parts were retained for analysis. During the race, contact time, duty factor, and trunk anteroposterior acceleration increased, and the foot strike angle and vertical stiffness decreased significantly. Heart rate showed a progressive increase, while the metrics for heart rate variability and complexity decreased during the race. The biomechanical parameters showed a significant alteration even with a small change in perceived fatigue, whereas the heart rate dynamics altered at higher changes. When divided into two groups, the slower runners presented a higher change in heart rate dynamics throughout the race than the faster runners; they both showed similar trends for the gait parameters. When tested for linear and non-linear correlations, heart rate had the highest association with biomechanical parameters, while the trunk anteroposterior acceleration had the lowest association with heart rate dynamics. These results indicate the ability of faster runners to better judge their physiological limits and hint toward a higher sensitivity of perceived fatigue to neuromuscular changes in the running gait. This study highlights measurable influences of acute fatigue, which can be studied only through concurrent measurement of biomechanical, physiological, and psychological facets of running in real-world conditions.

Project description:PurposeAchievement of athletes' performances is related to several factors including physiological, environmental and institutional cycles where physical characteristics are involved. The objective of this study is to analyse the performance achieved in professional sprint and middle-distance running events (100 m to 1500 m) depending on the organization of the annual calendar of track events and their environmental conditions.MethodsFrom 2002 to 2008, all performances of the Top 50 international athletes in the 100 m to 1500 m races (men and women) are collected. The historical series of world records and the 10 best annual performances in these events, amounted to a total of 26,544 performances, are also included in the study.ResultsTwo periods with a higher frequency of peak performances are observed. The first peak occurs around the 27.15(th) ±0.21 week (first week of July) and the second peak around 34.75(th) ±0.14 week (fourth week of August). The second peak tends to be the time of major international competitions (Olympic Games, World Championships, and European Championships) and could be characterized as an institutional moment. The first one, however, corresponds to an environmental optimum as measured by the narrowing of the temperature range at the highest performance around 23.25±3.26°C.ConclusionsThis is the first study to demonstrate that there are two performance peaks at a specific time of year (27th and 34th weeks) in sprint and middle distance. Both institutional and ecophysiological aspects contribute to performance in the 100 m to 1500 m best performances and define the contours of human possibilities. Sport institutions may take this into account in order to provide ideal conditions to improve the next records.

Project description:PurposeThe aim of this study was to verify whether physiological components [vertical jumps (Squat Jump - SJ and Countermovement Jump - CMJ), eccentric utilization ratio (EUR) of vertical jumps, running economy (RE), metabolic cost (C MET ), first and second ventilatory threshold (VT1 and VT2) maximal oxygen uptake (VO2MAX)] can predict maximal endurance running performance.MethodsTwenty male runners performed maximal vertical jumps, submaximal running at constant speeds, and maximal incremental running test. Before, we measured anthropometric parameters (body mass and height) and registered the training history and volume. SJ and CMJ tests were evaluated prior to running tests. Initially, the oxygen uptake (VO2) was collected at rest in the orthostatic position for 6 min. Soon after, a 10-min warm-up was performed on the treadmill at 10 km⋅h-1, followed by two 5-min treadmill rectangular tests at 12 and 16 km⋅h-1 monitored by a gas analyzer. After that, the runners performed a maximal incremental test, where the VT1, VT2, and VO2MAX were evaluated, as well as the maximum running speed (vVO2MAX). Thus, RE and C MET were calculated with data obtained during rectangular running tests. Multivariate stepwise regression analyses were conducted to measure the relationship between independent variables (height and power of SJ and CMJ, EUR; RE and C MET 12 and 16 km⋅h-1 ; VT1, VT2, and VO2MAX), as predictors of maximal running performance (vVO2MAX), with significance level at α = 0.05.ResultsWe found that VO2MAX and RE at 16 km⋅h-1 predict 81% of performance (vVO2MAX) of endurance runners (p < 0.001).ConclusionThe main predictors of the maximal incremental running test performance were VO2MAX and RE.

Project description:Objective:We present an exploratory study for identification of sex differences in imaging biomarkers that could further refine selection of patients for acute reperfusion therapy and trials based on sex and imaging targets. Methods:The Lesion Evolution in Stroke and Ischemia On Neuroimaging (LESION) study included consecutive acute stroke patients who underwent MRI within 24 h of time from last known well and prior to therapy. Those demonstrating a potential therapeutic target on imaging were identified by presence of: (1) arterial occlusion on angiography, (2) focal ischemic region on perfusion maps, or (3) a mismatch of perfusion versus diffusion imaging lesion size. The prevalence of imaging targets within clinically relevant time intervals was calculated for each patient and examined. The relationship of time from stroke onset to probability of detection of imaging targets was evaluated. Results:Of 7007 patients screened, of which 86.7% were scanned with MRI, 1092 patients (477/615 men/women) were included in LESION. The probability of imaging target detection was significantly different between men and women, with women more likely to present with all assessed imaging targets, odds ratios between 1.36 and 1.59, P < 0.02, adjusted for NIHSS, age, and time from last known well to MRI scan. This trend held for the entire 24-h studied. Interpretation:Women present more often with treatable ischemic stroke than men. The greater probability of potentially viable and/or treatable imaging targets in women at all time points suggests that tissue injury is slower to evolve in women.

Project description:It is known that muscular force is reduced in old age. We investigate what are the effects of this phenomenon on the mechanics of running. We hypothesized that the deficit in force would result in a lower push, causing reduced amplitude of the vertical oscillation, with smaller elastic energy storage and increased step frequency. To test this hypothesis, we measured the mechanical energy of the centre of mass of the body during running in old and young subjects. The amplitude of the oscillation is indeed reduced in the old subjects, resulting in an approximately 20% smaller elastic recovery and a greater step frequency (3.7 versus 2.8 Hz, p=1.9x10(-5), at 15-17 km h(-1)). Interestingly, the greater step frequency is due to a lower aerial time, and not to a greater natural frequency of the system, which is similar in old and young subjects (3.6 versus 3.4 Hz, p=0.2). Moreover, we find that in the old subjects, the step frequency is always similar to the natural frequency, even at the highest speeds. This is at variance with young subjects who adopt a step frequency lower than the natural frequency at high speeds, to contain the aerobic energy expenditure. Finally, the external work to maintain the motion of the centre of mass is reduced in the old subjects (0.9 versus 1.2 J kg(-1) m(-1), p=5.1x10(-6)) due to the lower work done against gravity, but the higher step frequency involves a greater internal work to reset the limbs at each step. The net result is that the total work increases with speed more steeply in the old subjects than in young subjects.

Project description:Wearable exercise trackers provide data that encode information on individual running performance. These data hold great potential for enhancing our understanding of the complex interplay between training and performance. Here we demonstrate feasibility of this idea by applying a previously validated mathematical model to real-world running activities of ≈ 14,000 individuals with ≈ 1.6 million exercise sessions containing duration and distance, with a total distance of ≈ 20 million km. Our model depends on two performance parameters: an aerobic power index and an endurance index. Inclusion of endurance, which describes the decline in sustainable power over duration, offers novel insights into performance: a highly accurate race time prediction and the identification of key parameters such as the lactate threshold, commonly used in exercise physiology. Correlations between performance indices and training volume and intensity are quantified, pointing to an optimal training. Our findings hint at new ways to quantify and predict athletic performance under real-world conditions.

Project description:Background: Properly performed training is a matter of importance for endurance athletes (EA). It allows for achieving better results and safer participation. Recently, the development of machine learning methods has been observed in sports diagnostics. Velocity at anaerobic threshold (VAT), respiratory compensation point (VRCP), and maximal velocity (Vmax) are the variables closely corresponding to endurance performance. The primary aims of this study were to find the strongest predictors of VAT, VRCP, Vmax, to derive and internally validate prediction models for males (1) and females (2) under TRIPOD guidelines, and to assess their machine learning accuracy. Materials and Methods: A total of 4001 EA (nmales = 3300, nfemales = 671; age = 35.56 ± 8.12 years; BMI = 23.66 ± 2.58 kg·m-2; VO2max = 53.20 ± 7.17 mL·min-1·kg-1) underwent treadmill cardiopulmonary exercise testing (CPET) and bioimpedance body composition analysis. XGBoost was used to select running performance predictors. Multivariable linear regression was applied to build prediction models. Ten-fold cross-validation was incorporated for accuracy evaluation during internal validation. Results: Oxygen uptake, blood lactate, pulmonary ventilation, and somatic parameters (BMI, age, and body fat percentage) showed the highest impact on velocity. For VAT R2 = 0.57 (1) and 0.62 (2), derivation RMSE = 0.909 (1); 0.828 (2), validation RMSE = 0.913 (1); 0.838 (2), derivation MAE = 0.708 (1); 0.657 (2), and validation MAE = 0.710 (1); 0.665 (2). For VRCP R2 = 0.62 (1) and 0.67 (2), derivation RMSE = 1.066 (1) and 0.964 (2), validation RMSE = 1.070 (1) and 0.978 (2), derivation MAE = 0.832 (1) and 0.752 (2), validation MAE = 0.060 (1) and 0.763 (2). For Vmax R2 = 0.57 (1) and 0.65 (2), derivation RMSE = 1.202 (1) and 1.095 (2), validation RMSE = 1.205 (1) and 1.111 (2), derivation MAE = 0.943 (1) and 0.861 (2), and validation MAE = 0.944 (1) and 0.881 (2). Conclusions: The use of machine-learning methods allows for the precise determination of predictors of both submaximal and maximal running performance. Prediction models based on selected variables are characterized by high precision and high repeatability. The results can be used to personalize training and adjust the optimal therapeutic protocol in clinical settings, with a target population of EA.

Project description:Exercise can prevent and improve the pathophysiology of diseases and promote healthy aging. Thus, understanding the mechanisms that regulate the beneficial effects of exercise may lead to the development of new strategies to enhance quality of life and to counteract chronic diseases. Voluntary wheel running is an interesting model to study the effects of exercise in mice. Compared to forced treadmill exercise, voluntary wheel running presents several advantages such as: 1) running pattern is similar to natural running behavior of mice; 2) it is performed under non-stressed conditions, according to the rhythmicity of the animal; 3) it does not require direct interference from the researcher, and can be easily applied in long-term studies. Mice run spontaneously when given access to running wheels, for a total distance of ?4 to 20 km per day and a total activity time of ?3 to 7 hours a day. Hence, voluntary wheel running can result in robust endurance-like adaptation in skeletal and cardiac muscles and protect from sarcopenia. However, due to the lack of control over exercise parameters in voluntary exercise models, it is important for the researcher to understand the patterns and variability of wheel running in mice, as well as the factors that can affect voluntary running activity. Overall, voluntary wheel running in mice is a very interesting approach to study the chronic adaptation to exercise, analyze the effects of exercise, and test exercise capacity in different experimental models.

Project description:Human gait is as unique to an individual as is their fingerprint. It remains unknown, however, what gait characteristics differentiate well between individuals that could define the uniqueness of human gait. The purpose of this work was to determine the gait characteristics that were most relevant for a neural network to identify individuals based on their running patterns. An artificial neural network was trained to recognize kinetic and kinematic movement trajectories of overground running from 50 healthy novice runners (males and females). Using layer-wise relevance propagation, the contribution of each variable to the classification result of the neural network was determined. It was found that gait characteristics of the coronal and transverse plane as well as medio-lateral ground reaction forces provided more information for subject identification than gait characteristics of the sagittal plane and ground reaction forces in vertical or anterior-posterior direction. Additionally, gait characteristics during the early stance were more relevant for gait recognition than those of the mid and late stance phase. It was concluded that the uniqueness of human gait is predominantly encoded in movements of the coronal and transverse plane during early stance.
