Vitamin B₃ Intercalated in Layered Double Hydroxides: A Drug Delivery System for Metabolic Regulation.

ABSTRACT: The organic compound niacin or nicotinic acid, also known as vitamin B₃ (VitB₃), is essential for human nutrition and metabolic regulation. However, in high doses, it can provoke side effects, such as hyperglycemia, liver damage, and flushing. Development of a controlled release system that slowly releases VitB₃ into the organism would avoid high dosing peaks, thus contributing to decrease the occurrence of side effects in nutritional supplementation. Here, we show that the slow and controlled release of VitB₃ in an acid environment can be achieved via its intercalation in layered double hydroxides (LDHs). The synthesis of a ZnAl-VitB₃ system is shown, in which VitB₃ is intercalated in a ZnAl LDH. The presence of VitB₃ in the ZnAl-VitB₃ system was confirmed by elemental analysis, infrared (FTIR) and NMR spectroscopy, while successful intercalation in the LDHs was revealed by powder X-ray diffraction (PXRD). In vitro release tests were carried out in a concentrated HCl solution of pH 1.5, a pH similar to the human stomach environment. The results showed a steady release of VitB₃ from the LDH host, with 90% of the vitamin liberated in the first 60 min after the suspension of the LDH in the acidic solution.

Project description:BackgroundTo enhance ocular bioavailability, the traditional strategies have focused on prolonging precorneal retention and improving corneal permeability by nano-carriers with positive charge, thiolated polymer, absorption enhancer and so on. Glycylsarcosine (GS) as an active target ligand of the peptide tranpsporter-1 (PepT-1), could specific interact with the PepT-1 on the cornea and guide the nanoparticles to the treating site.PurposeThe objective of the study was to explore the active targeting intercalated nanocomposites based on chitosan-glutathione-glycylsarcosine (CG-GS) and layered double hydroxides (LDH) as novel carriers for the treatment of mid-posterior diseases.Materials and methodsCG-GS-LDH intercalated nanocomposites were prepared by the coprecipitation hydrothermal method. In vivo precorneal retention study, ex vivo fluorescence images, in vivo experiment for distribution and irritation were studied in rabbits. The cytotoxicity and cellular uptake were studied in human corneal epithelial primary cells (HCEpiC).ResultsCG-GS-LDH nanocomposites were prepared successfully and characterized by FTIR and XRD. Experiments with rabbits showed longer precorneal retention and higher distribution of fluorescence probe/model drug. In vitro cytological study, CG-GS-LDH nanocomposites exhibited enhanced cellular uptake compared to pure drug solution. Furthermore, the investigation of cellular uptake mechanisms demonstrated that both the active transport by PepT-1 and clathrin-mediated endocytosis were involved in the internalization of CG-GS-LDH intercalated nanocomposites. An ocular irritation study and a cytotoxicity test indicated that these nanocomposites produced no significant irritant effects.ConclusionsThe active targeting intercalated nanocomposites could have great potential for topical ocular drug delivery due to the capacity for prolonging the retention on the ocular surface, enhancing the drug permeability through the cornea, and efficiently delivering the drug to the targeted site.

Project description:Auxins are a class of organic substances known as plant-growth regulators, which act on plant physiology, promoting its full development. However, due to the great instability of these substances among the diversity of crops and cultivation environments, it is necessary to seek more efficient modes of application, which lead to a homogeneous distribution and promote a sustained release according to the plants demand. Seed coating, using films containing a biodegradable polymer and auxins intercalated into layered compounds, emerges as a very promising approach to a new form of growth regulator application. Thus, the presented work had three aims: (i) the synthesis and characterization of an organic-inorganic hybrid material containing a layered double hydroxide (LDH) of zinc and aluminum and the synthetic auxin 1-naphthalenoacetic acid (ZnAl-NAA-LDH), (ii) the coating of bean seeds (Phaseolus vulgaris L.) with composite films produced from mixtures of alginate polymer and ZnAl-NAA-LDH, and (iii) the evaluation of the plant response by bioassays. The hybrid ZnAl-NAA-LDH was characterized by a set of analytical techniques, including powder X-ray diffraction, thermogravimetric analysis coupled to differential scanning calorimetry and mass spectrometry, specific surface area measurement, and scanning electron microscopy. Bioassays were performed with the seeds coated with the composite film to assess the germination rate and germination speed index of the seeds, as well as biometric analyses including measurements of root area, root fresh matter, and shoot length of the plants. The bioassay performed in soil pots showed that the alginate film containing ZnAl-NAA-LDH yields an enhancement regarding root area, fresh root matter and shoot length of plants. Thus, films produced from a mixture of alginate and the hybrid material containing the growth regulator intercalated into LDH can be a viable alternative to enhance plant development, which can be included in seed management.
